Demineralized bone matrix(DBM)has been widely used clinically for dental,craniofacial and skeletal bone repair,as an osteoinductive and osteoconductive material.3D printing(3DP)enables the creation of bone tissue engineering scaffolds with complex geometries and *** methacryloylated gelatin nanoparticles(GNP-MAs)3DP inks have been developed,which display gel-like behavior for high print fidelity and are capable of post-printing photocrosslinking for control of scaffold swelling and ***,novel DBM nanoparticles(DBM-NPs,∼400 nm)were fabricated and characterized prior to incorporation in 3DP *** objectives of this study were to determine how these DBM-NPs would influence the printability of composite colloidal 3DP inks,assess the impact of ultraviolet(UV)crosslinking on 3DP scaffold swelling and degradation and evaluate the osteogenic potential of DBM-NP-containing composite colloidal *** addition of methacryloylated DBM-NPs(DBM-NP-MAs)to composite colloidal inks(100:0,95:5 and 75:25 GNP-MA:DBM-NP-MA)did not significantly impact the rheological properties associated with printability,such as viscosity and shear recovery or *** crosslinking with a UV dosage of 3 J/cm2 directly impacted the rate of 3DP scaffold swelling for all GNP-MA:DBM-NP-MA ratios with an∼40%greater increase in scaffold area and pore area in uncrosslinked versus photocrosslinked scaffolds over 21 days in phosphate-buffered saline(PBS).Likewise,degradation(hydrolytic and enzymatic)over 21 days for all DBM-NP-MA content groups was significantly decreased,∼45%less in PBS and collagenase-containing PBS,in UV-crosslinked versus uncrosslinked *** incorporation of DBM-NP-MAs into scaffolds decreased mass loss compared to GNP-MA-only scaffolds during collagenase *** in vitro osteogenic study with bone marrow-derived mesenchymal stem cells demonstrated osteoconductive properties of 3DP scaffolds for the DBM-NP-MA contents examine

Parental Awareness and Perception of Their Children’s Body Size

Objective: To investigate parents’ misperception of their active children’s size. Subjects: One hundred and forty male or female parents and their children from a youth soccer league participated. Actual Child Body Mass Index (BMI) percentiles and BMI categories were compared to the results of a Body Size Estimation Task and lifestyles questionnaire results. Results: Parents underestimated the body size of their children and placed them in a lower BMI category than the children actually belonged in. As was the case in other studies children in the unhealthy or at risk to become overweight category where seen as a healthy weight. Unlike other studies, many healthy weight children were seen as underweight. Conclusions: An explanation of body size misperception and underestimation of body size may be “change blindness”. The growing prevalence of obesity in children may be better addressed by focusing on the parents’ apparent lack of concern about excess childhood weight and the parents’ identification of excess childhood weight as “just normal” rather than seeing excess weight as a potential problem.

This study assessed apparently normal mitral valves from patients with congestive heart failure(CHF)using biochemical and echocardiographic measures of extracellular matrix(ECM)and anatomy. Mitral regurgitation(MR)is frequently found in patients with CHF. This MR is considered purely functional, yet animal studies suggest that altered left ventricular(LV)function leads to increased cellularity and fibrosis of the mitral valve. Therefore, we hypothesized that patients with CHF might have partly organic MR, via dysfunctional valvular remodeling. Mitral valves from transplant recipient hearts of patients with CHF(23 dilated, 14 ischemic)were analyzed for deoxyribonucleic acid(DNA), collagen, glycosaminoglycan(GAG), and water concentrations and compared with autopsy controls. Cardiac dimensions and functional parameters(measured from recent echocardiograms)were compared with biochemical parameters using a repeated measures generalized linear model. The mitral valves in CHF had up to 78%more DNA(p kness and DNA concentration(+, p=0.003), annular diameter and chordal collagen(+, p=0.03), and water concentration and both left atrial diameter(-, p=0.008)and LV collagen concentration(-, p=0.04). Mitral valves in CHF are biochemically different from normal, with ECM changes that are influenced by the altered cardiac dimensions. This remodeling suggests that MR in patients with CHF may not be purely functional, and that these valves are not “normal.”

Aim:We aimed to further investigate the role of hepcortespenlisimut-L(Hepko-V5 or V5),a new oral immunotherapy developed by us,for hepatocellular carcinoma(HCC)***:The interim data from ongoing PhaseⅢplacebo-controlled,randomized trial were evaluated on the initial group of patients in advanced stage of HCC with emphasis on liver function and tumor marker alpha-fetoprotein ***,an in vitro study was undertaken to elucidate the mechanism of action of V5 by measuring with flow cytometry the expression of cytokines such as IL-2,INF-γ,and TNF-αand cell activation markers CD69 and ki67 on CD4-and CD8-positive lymphocytes isolated from peripheral blood of healthy ***:As early as one month after treatment initiation,there was a clear improvement in alanine transaminase,aspartate transaminase,alkaline phosphatase,and bilirubin levels among HCC patients who received daily dose of V5,but not in the placebo ***,alpha-fetoprotein(AFP)levels among V5 recipients decreased,while in the ;placebo group they *** results are in line with in vitro observations indicating immune activation,as evidenced by many-fold enhancement of CD69,ki67,and INF-γexpression and at the same time marked anti-inflammatory effect resulting in 10-fold decrease in TNF-αoutput and lack of influence on IL-2 ***:Hepcortespenlisimut-L,a tableted oral formulation derived from heat-inactivated pooled blood of patients with HCC and viral hepatitis shows beneficial clinical effect,as demonstrated by improvement in liver function and reduction of tumor marker AFP *** correlate with in vitro observations showing potent activation of the immune response and pronounced oral tolerance effect.
