Interleukin-21(IL-21)is produced mainly by activated CD4^(+)T cells and promotes the proliferation,survival and differentiation of immune cells.This study characterised a high affinity binding receptor of IL-21(IL-21 Rα)from grass carp(Ctenopharyngodon idella,Ci)and analyzed the expression of CiIL-21 and CiIL-21 Rα.It has been shown that they were modulated by PAMPs and during infection with bacterial and viral pathogens.Recombinant CiIL-21 protein produced in bacteria was active in inducing the expression of genes involved in Th responses.A monoclonal CiIL-21 antibody was generated against the rCiIL-21 and showed good reactive specificity with the rCiIL-21 proteins expressed in E.coli cells,and HEK293 T cells and the native protein from PHA-stimulated kidney leukocytes.In addition,the IL-21-producing cells detected by the CiIL-21 monoclonal antibody were found to be increased to in the kidney,spleen and hindgut after infection with Flavobacterium columnare.Our data indicate that IL-21 is involved in the regulation of immune response to bacterial and viral infection.

CD4 T helper cells are an important group of cells in the immune system of vertebrates and express CD4 receptor on the cell surface.In mammals,the CD4 receptor is encoded by a single copy gene,whilst in fish,two copies of cd4 genes,namely cd4-1 and cd4-2,are found.In this study,the ectodomains of grass carp(Ctenopharyngodon idella,Ci)CD4-1 and CD4-2 were expressed in the E.coli cells and used to generate monoclonal antibodies in mice.Western blotting,confocal microscopy and flow cytometry were performed to characterize the monoclonal antibodies.It has been shown that the CiCD4-1 and CiCD4-2 monoclonal antibodies had good specificity to react with the recombinant ectodomains of CiCD4-1 and CiCD4-2 expressed in the CHO-S cells and the native CD4 molecules of grass carp.The CiCD4-1 monoclonal antibody did not recognize CiCD4-2 and verse versa.In addition,the CD4-1 and CD4-2 monoclonal antibodies specifically recognized the CD4-1 and CD4-2 receptors expressed in the HEK293 cells and native molecules of fish cells.Further,the percentages of lymphocytes in immune tissues of healthy fish were analyzed by flow cytometry.It was found that 17.6%of lymphocytes were CD4-1^(+)cells and 22.5%were CD4-2^(+)in the head kidney.In the spleen,13.1%of lymphocytes were CD4-1^(+)and 18.6%were CD4-2^(+)while 7.3%of blood lymphocytes were CD4-1^(+)cells and 8.8%were CD4-2^(+)cells.The availability of the CD4-1 and CD4-2 monoclonal antibodies provides antibody-based tools for further elucidation of the functions of CD4 T cells in grass carp.

A cell line,termed ZFIN,was established from the caudal fin of zebrafish and was shown to be susceptible to spring viremia of carp virus(SVCV).The ZFIN cells are epithelial like cells and have a moderate plasmid transfection efficiency of 13.9%.Using an RNA-seq approach,differentially expressed genes(DEGs)regulated by SVCV were identified.Infection of SVCV gave rise to 3931 DEGs and up-regulated DEGs were mostly enriched into the biological regulation and cellular processes,among which pathways for the type I interferon signaling and the response to exogenous dsRNA were the top two GO terms.Several KEGG signaling pathways including TLR signaling pathway,RLR receptor signaling pathway,cytosolic DNA-sensing pathway,NLR signaling pathway,cytokine-cytokine receptor interaction and ferroptosis were significantly enriched.Antiviral genes including ifnφ1,isg15 and mx were significantly up-regulated.In addition,key DEGs involved in autophagy were identified.The results indicate that the ZFIN cell line provides a useful in vitro tool for study on the gene functions and cellular responses to viral infection in fish.

A previously developed hybrid coupled model(HCM)is composed of an intermediate tropical Pacific Ocean model and a global atmospheric general circulation model(AGCM),denoted as HCMAGCM.In this study,different El Niño flavors,namely the Eastern-Pacific(EP)and Central-Pacific(CP)types,and the associated global atmospheric teleconnections are examined in a 1000-yr control simulation of the HCMAGCM.The HCMAGCM indicates profoundly different characteristics among EP and CP El Niño events in terms of related oceanic and atmospheric variables in the tropical Pacific,including the amplitude and spatial patterns of sea surface temperature(SST),zonal wind stress,and precipitation anomalies.An SST budget analysis indicates that the thermocline feedback and zonal advective feedback dominantly contribute to the growth of EP and CP El Niño events,respectively.Corresponding to the shifts in the tropical rainfall and deep convection during EP and CP El Niño events,the model also reproduces the differences in the extratropical atmospheric responses during the boreal winter.In particular,the EP El Niño tends to be dominant in exciting a poleward wave train pattern to the Northern Hemisphere,while the CP El Niño tends to preferably produce a wave train similar to the Pacific North American(PNA)pattern.As a result,different climatic impacts exist in North American regions,with a warm-north and cold-south pattern during an EP El Niño and a warm-northeast and cold-southwest pattern during a CP El Niño,respectively.This modeling result highlights the importance of internal natural processes within the tropical Pacific as they relate to the genesis of ENSO diversity because the active ocean–atmosphere coupling is allowed only in the tropical Pacific within the framework of the HCMAGCM.

In the face of the elevated incidence and mortality rate of septic shock in the ICU,this retrospective study seeks to investigate the indicative and predictive value of high-mobility group box 1(HMGB1)and miR-146b in patients with septic shock.Quantitative RTPCR was employed in this study to quantify the HMGB1 and miR-146b levels in plasma samples obtained from the patient group and healthy controls.The investigation involved the comparison between the two groups and tracking changes in the patient group over time.The finding revealed that upon admission,the patient group exhibited markedly elevated relative expression levels of HMGB1,which subsequently decreased over time.Conversely,the patient group displayed significantly reduced relative expression levels of miR-146b upon admission,which subsequently increased over time compared to the control group.Receiver operating characteristic(ROC)curves showed good predictive value for HMGB1 and miR-146b.The experimental results suggest that HMGB1 and miR-146b serve as valuable and convenient biomarkers for evaluating the severity of septic shock and predicting mortality.Additionally,it is proposed that serum miR-146b may be inducible and potentially exerts a negative regulatory effect on the expression of HMGB1.

In this paper,an angular position sensor(APS)designed for a resonant miniaturized scanning mirror(M-SM)is presented.The APS operates based on the principle of differential variable capacitance,significantly expanding the detectable bandwidth from a few hertz to several kilohertz.By modeling the motion characteristics,the sampling rates of the biaxial scanning angles are 1473.6 times and 539.4 times higher than those of conventional sensors.Initially,the motion characteristics model is presented as a simple harmonic motion,converting sampled capacitance into continuous capacitance.Subsequently,the nonparallel state of the M-SM and sensor is transformed into a parallel state through the space coordinate system transformation.Furthermore,a 2D nonlinear angle transfer function is developed to convert the differential capacitance into an angle,thereby mitigating the nonlinear errors resulting from large angles.Achieving an accuracy better than 0.014°,the measuring range expands from±0.5729°(±10 mrad)to±5.026°(±87 mrad).Additionally,the capturing mode and tracking mode are proposed to monitor real-time angular changes of the M-SM with an accuracy of 0.017°.High-precision APSs have enhanced beam pointing accuracy and resolution and can thereby be used to advance the development of laser components,including light detection and ranging(LiDAR).

By using the multi-taper method(MTM)of singular value decomposition(SVD),this study investigates the interdecadal evolution(10-to 30-year cycle)of precipitation over eastern China from 1951 to 2015 and its relationship with the North Pacific sea surface temperature(SST).Two significant interdecadal signals,one with an 11-year cycle and the other with a 23-year cycle,are identified in both the precipitation and SST fields.Results show that the North Pacific SST forcing modulates the precipitation distribution over China through the effects of the Pacific Decadal Oscillation(PDO)-related anomalous Aleutian low on the western Pacific subtropical high(WPSH)and Mongolia high(MH).During the development stage of the PDO cold phase associated with the 11-year cycle,a weakened WPSH and MH increased the precipitation over the Yangtze River Basin,whereas an intensified WPSH and MH caused the enhanced rain band to move northward to North China during the decay stage.During the development stage of the PDO cold phase associated with the 23-year cycle,a weakened WPSH and MH increased the precipitation over North China,whereas an intensified WPSH and the weakened MH increased the precipitation over South China during the decay stage.The 11-year and 23-year variabilities contribute differently to the precipitation variations in the different regions of China,as seen in the 1998flooding case.The 11-year cycle mainly accounts for precipitation increases over the Yangtze River Basin,while the 23-year cycle is responsible for the precipitation increase over Northeast China.These results have important implications for understanding how the PDO modulates the precipitation distribution over China,helping to improve interdecadal climate prediction.

To the Editor:Probiotics are a promising treatment modality for functional constipation(FC);however,the factors affecting individual responses to probiotics remain unclear.Growing evidence has identified that there is a strong relationship among the gut microbiota and constipation and related gut-brain axis.[1]The gut microbiota may modulate the gut functions via gut metabolites or trigger the release of gut hormones,such as peptide YY,gastric inhibitory polypeptide,and 5-hydroxytryptamine.[2]In turn,gut hormones affect gut secretion,motility,and sensation through their receptors located on epithelial,enteric,and smooth muscle cells.[2]Based on these findings,exogenous probiotics have been used in patients with constipation;however,few have produced consistent results.[3]In this study,we investigated the efficacy and safety of several specific probiotics strains for constipation,and also examined the potential reasons for the individualized effects of probiotics,which may facilitate interventional decisionmaking for FC patients.

MEMS light detection and ranging(LiDAR)is becoming an indispensable sensor in vehicle environment sensing systems due to its low cost and high performance.The beam scanning trajectory,sampling scheme and gridding are the key technologies of MEMS LiDAR imaging.In Lissajous scanning mode,this paper improves the sampling scheme,through which a denser Cartesian grid of point cloud data at the same scanning frequency can be obtained.By summarizing the rules of the Cartesian grid,a general sampling scheme independent of the beam scanning trajectory patterns is proposed.Simulation and experiment results show that compared with the existing sampling scheme,the resolution and the number of points per frame are both increased by 2 times with the same hardware configuration and scanning frequencies for a MEMS scanning miror(MEMS-SM).This is beneficial for improving the point cloud imaging performance of MEMS LiDAR.

Low efficiency is the main obstacle to using prime editing in maize(Zea mays).Recently,prime-editing efficiency was greatly improved in mammalian cells and rice(Oryza sativa)plants by engineering primeediting guide RNAs(pegRNAs),optimizing the prime editor(PE)protein,and manipulating cellular determinants of prime editing.In this study,we tested PEs optimized via these three strategies in maize.We demonstrated that the ePE5max system,composed of PEmax,epegRNAs(pegRNA-evopreQ.1),nicking single guide RNAs(sgRNAs),and MLH1dn,efficiently generated heritable mutations that conferred resistance to herbicides that inhibit 5-enolpyruvylshikimate-3-phosphate synthase(EPSPS),acetolactate synthase(ALS),or acetyl CoA carboxylase(ACCase)activity.Collectively,we demonstrate that the ePE5max system has sufficient efficiency to generate heritable(homozygous or heterozygous)mutations in maize target genes and that the main obstacle to using PEs in maize has thus been removed.
