Effects of Salvia miltiorrhiza Bunge extract and its single components on monosodium urate-induced pain in vivo and lipopolysaccharide-induced inflammation in vitro

OBJECTIVE:To investigate the possible antinociceptive effects of Salvia(S.)miltiorrhiza Bunge and its single components in monosodium urate(MSU)-induced pain model in mice and lipopolysaccharide(LPS)-induced inflammation model in RAW264.7 ***:Pretreatment of *** Bunge extract(from 1 to 50μg/mL)concentration-dependently attenuated LPS-induced nitric oxide(NO)*** extract of *** Bunge(50 or 100 mg/kg)also caused reversals of decreased threshold for pain in the MSU-treated group as measured by Von-Frey ***,we assessed the antinociceptive and anti-inflammatory properties of the active single components from *** Bunge such as 15,16-dihydrotanshinoneⅠ,tanshinoneⅠ,cryptotanshinone,miltirone,tanshinoneⅡA,and salvianolic acid *** of them showed an anti-inflammatory effect in LPS-induced NO release model and an antinociceptive effect in MSU-treated pain ***:Our results suggest that *** Bunge extract may exert anti-inflammatory effect by reducing LPS-induced NO release and an antinociceptive property in MSU-treated pain ***,tanshinoneⅡA,miltirone,cryptotanshinone,and 15,16-dihydrotanshinoneⅠ,not only appear to be responsible for LPS-induced NO release induced by *** Bunge,but also in the production of *** Bunge extract-induced antinociception in MSU-treated pain ***:Therefore,the analgesic and anti-inflammatory property of *** Bunge indicate it as a therapeutic potential candidate for the treatment of pain and inflammation.

A high-carbon steel joint,SK5（0.84 wt% C）,was successfully welded by friction stir welding（FSW）,both without and with a gas torch,in order to control the cooling rate during *** welding,the weld zone comprised gray and black regions,corresponding to microstructural variation：a martensite structure and a duplex structure of ferrite and cementite,*** volume fraction of the martensite structure and the Vickers hardness in the welds were decreased with the using of the gas torch,which was related with the lower cooling rate.

Bioresponsive hydrogels are smart materials that respond to various external stimuli and exhibit great potential as biosensors owing to their capability of real-time and label-free ***,we propose a sensing platform based on bioresponsive hydrogels,employing the concept of moiré *** sets of line patterns with different pitch sizes are prepared;a hydrogel grating whose pitch size changes according to external stimuli and a reference grating with constant pitch *** volume changes of the hydrogel caused by external stimuli changes the pitch size of the hydrogel grating,and subsequently,the pitch sizes of the moiré patterns(moiré signal),whose values can be obtained in a real-time and label-free manner through customized moiré microscopy and signal *** confirming that the pH-induced swelling of hydrogel could be monitored using moiré patterns,we performed moiré pattern-based detection of specific proteins using protein-responsive hydrogel that underwent shrinking via interaction with target ***-derived neurotrophic factor and platelet-derived growth factor were selected as the model proteins,and our proposed system successfully detected both proteins at nanomolar *** both cases,the pitch size change of hydrogel grating was monitored much more sensitively using moiré patterns than through direct *** changes in the moiré signals caused by target proteins were detected in ex-vivo environments using a custom-made intraocular lens incorporating the hydrogel grating,demonstrating the capability of the proposed system to detect various markers in intraocular aqueous humor,when implanted in the eye.

Human umbilical cord blood-derived mesenchymal stem cells promote regeneration of crush-injured rat sciatic nerves

Several studies have demonstrated that human umbilical cord blood-derived mesenchymal stem cells can promote neural regeneration following brain injury. However, the therapeutic effects of human umbilical cord blood-derived mesenchymal stem cells in guiding peripheral nerve regeneration remain poorly understood. This study was designed to investigate the effects of human umbilical cord blood-derived mesenchymal stem cells on neural regeneration using a rat sciatic nerve crush injury model. Human umbilical cord blood-derived mesenchymal stem cells （1 ~ 106） or a PBS control were injected into the crush-injured segment of the sciatic nerve. Four weeks after cell injection, brain-derived neurotrophic factor and tyrosine kinase receptor B mRNA expression at the lesion site was increased in comparison to control. Furthermore, sciatic function index, Fluoro Gold-labeled neuron counts and axon density were also significantly increased when compared with control. BACKGROUND Ligamentum flavum cysts,which are most common in mobile junctional levels of the spine,can be a rare cause of spinal *** have been several case reports of ligamentum flavum ***,there is yet to be a documented case report of a calcified ligamentum flavum ***,we report the first case of a calcified ligamentum flavum cyst causing ankle and toe *** SUMMARY A 66-year-old male visited our hospital complaining of claudication as well as thigh and calf pain in his left leg,all beginning two weeks *** examination revealed motor weakness of the left ankle dorsiflexion and great toe *** spinal computed tomography scans showed spinal stenosis combined with a calcified mass at the left side of the L4-5 *** reso-nance imaging showed dural sac compression caused by the calcified mass at the left ligamentum flavum of the L4-5 *** performed decompressive laminectomy and excision of the calcified mass combined with posterior lumbar interbody fusion at the L4-5 ***-operatively,we found a firm and nodule like mass originating from the ventral surface of ligamentum *** examination suggested a calcified pseudocyst without a capsular *** the operation,the patient’s motor weakness in the ankle and great toe improved *** The patient’s ankle and great toe weakness were improved successfully after surgical removal of the calcified cyst.

Protective effects of Ecklonia cava extract on the toxicity and oxidative stress induced by hair dye in in-vitro and in-vivo models

Oxidative hair dyes containingρ-phenylenediamine(PPD)are reported to induce an allergic reaction by promoting oxidative stress when absorbed through the *** the associated risk,these hair dyes remain popular owing to their convenience and sharpness of *** makes it important to minimize the cytotoxicity and oxidative stress induced by PPD-containing hair *** cava extract has been evaluated in different studies for its protective effects against external stress in fibroblasts and *** study was aimed at using in-vitro and in-vivo models to investigate the extract’s effects on cytotoxicity of and oxidative stress induced by PPD-containing hair *** of CIEL*a*b*Color space was first used to determine the range of *** extract that would not interfere with the coloring ability of the dye upon ***,the set ranges of *** extract(5% and 7%)were added to the hair dye and their toxicity assessed by evaluating the viability of fibroblasts and *** effects on developmental phenotypes and induction of oxidative stress by hair dye were evaluated and compared with those of hair dyes containing different contents of *** extract using an in-vivo zebrafish *** results showed that *** extract in hair dye could significantly decrease the cytotoxicity and levels of oxidative stress caused by hair dyes containing PPD in both in-vitro and in-vivo *** results suggest that the addition of 7% *** extract to 250μg/mL hair dye does not interfere with the coloring ability of the dye while showing significant protective eff ects against the hair *** study proposes that the use of *** extract as an adduct to hair dyes containing PPD reduces the cytotoxicity and oxidative stress induced by these hair dyes.

Human periodontal ligament stem cells repair mental nerve injury

Human periodontal ligament stem cells are easily accessible and can differentiate into Schwann cells. We hypothesized that human periodontal ligament stem cells can be used as an alternative source for the autologous Schwann cells in promoting the regeneration of injured peripheral nerve. To validate this hypothesis, human periodontal ligament stem cells （1 × 106） were injected into the crush-injured left mental nerve in rats. Simultaneously, autologous Schwann cells （1 × 106） and PBS were also injected as controls. Real-time reverse transcriptase polymerase chain reaction showed that at 5 days after injection, mRNA expression of low affinity nerve growth factor receptor was sig-nificantaly increased in the left trigeminal ganglion of rats with mental nerve injury. Sensory tests, histomorphometric evaluation and retrograde labeling demonstrated that at 2 and 4 weeks after in-jection, sensory function was significantly improved, the numbers of retrograde labeled sensory neurons and myelinated axons were significantly increased, and human periodontal ligament stem cells and autologous Schwann cells exhibited similar therapeutic effects. These findings suggest that transplantation of human periodontal ligament stem cells show a potential value in repair of mental nerve injury.

The extract of Celtis choseniana Nakai alleviates testosterone-in-duced benign prostatic hyperplasia through inhibiting 5αreductase type 2 and the Akt/NF-κB/AR pathway
