Losartan reduced connexin43 expression in left ventricular myocardium of spontaneously hypertensive rats

Objective:To assess the effect of angiotensin II type 1(AT1)receptor antagonist losartan on myocardium con- nexin43(Cx43)gap junction(GJ)expression in spontaneously hypertensive rats(SHRs)and investigate possible mechanisms. Methods:Sixteen 9-week-old male SHRs and 8 age-matched male Wistar-Kyoto(WKY)rats were included in this *** were randomly divided into two groups to receive losartan at 30 mg/(kg·d)by oral gavage once daily for 8 weeks(SHR-L)or vehicle(0.9%saline)to act as controls(SHR-V);WKY rats receiving vehicle for 8 weeks served as normotensive *** the end of the experiment,rats were sacrificed and the hearts were *** of Cx43 and nuclear factor-kappaB p65 (NF-κB p65)proteins in all three groups were observed and further investigations on the effect of angiotensin II type 1 receptor antagonist losartan(30 mg/(kg·d),8 weeks)on Cx43 expression were conducted with Western blot and immunohistochemistry. NF-κB p65 protein in nuclear extracts was determined by Western ***:Left ventricular(LV)hypertrophy was prominent in SHRs,Cx43 and NF-κB p65 protein expressions were obviously upregulated and Cx43 distribution was dispersed over the cell *** with losarton reduced the over-expressions of Cx43 and NF-κB p65 in LV *** distribution of Cx43 gap junction also became much regular and confined to intercalated disk after losartan ***- sion:Cx43 level was upregulated in LV myocardium of SHR during early stage of *** II type 1 receptor antagonist losartan prevented Cx43 gap junction remodeling in hypertrophied left ventricles,possibly through the NF-κB pathway.

AIM: To investigate whether puerarin can augment endothelial progenitor cells (EPCs) numbers, promote EPC proliferative, migratory, adhesive, and in vitro vasculogenesis capacity. METHODS: EPCs were characterized as adherent cells by double staining of DiLDL-uptake and lectin binding under a laser scanning confocal microscope. Expression of KDR, VEGFR-2, and AC 133 was detected by flow cytometry. EPCs proliferation, migration and in vitro vasculogenesis were determined with MTT assay, modified Boyden chamber assay, and in vitro vasculogenesis kit, respectively. EPCs adhesive assay was performed by replating those on fibronectin-coated dishes, then adherent cells were counted. RESULTS: Incubation of isolated human MNCs with puerarin 0.1-3 mmol/L increased the number of EPCs, EPC proliferative, migratory, adhesive, and in vitro vasculogenesis capacity in a concentration and time-dependent manner, which reached maximum at 3 mmol/L, 24 h (approximately 1-fold increase, P<0.01). The interactions of the complexes of Cu(phen) 2+ 2 and Cu(bpy) 2+ 2 with DNA were investigated with electrochemistry and spectroelectrochemistry. Binding constants( K n + ) and the reaction model were obtained from their electrochemical behavior and electrochemical data. The binding constants( K 2+ ) for Cu(phen) 2+ 2 DNA and Cu(bpy) 2+ 2 DNA were 8 51×10 3 L·mol -1 and 3 27×10 3 L·mol -1 , and K +s were 3 09×10 4 L·mol -1 and 5 66×10 3 L·mol -1 respectively. The spectroscopic characteristics of the complexes of Cu(phen) 2+ 2 DNA and Cu(bpy) 2+ 2 DNA were studied in detail. Some significant spectroelectrochemical data were obtained. The interaction of DNA with complexes was described.

The association between the estrogen receptor a gene （ESR1） Pvull polymorphism （c.454-397T〉C） and coronary artery disease （CAD） is controversial. Thus, we conducted a meta-analysis to evaluate the relationship. Data were collected from 21 studies encompassing 9926 CAD patients and 16710 controls. Odds ratios （ORs） with 95% confidence intervals （CIs） were used to assess the relationship between Pvull polymorphism and CAD. The poly- morphism in control populations in all studies followed Hardy-Weinberg equilibrium. We found a significant association between ESR1 Pvull polymorphism and CAD risk in all subjects. When the data were stratified by region, a significant association between ESR1 Pvull polymorphism and CAD risk was observed in Asian populations but not in Western populations. The current study suggests that ESR1 Pvull polymorphism has an important role in CAD susceptibility.
