Eosinophils promote CD8+ T cell memory generation to potentiate anti-bacterial immunity

Memory CD8+T cell generation is crucial for pathogen elimination and effective vaccination against infection.The cellular and molecular circuitry that underlies the generation of memory CD8+T cells remains elusive.Eosinophils can modulate inflammatory allergic responses and interact with lymphocytes to regulate their functions in immune defense.Here we report that eosinophils are required for the generation of memory CD8+T cells by inhibiting CD8+T cell apoptosis.Eosinophil-deficient mice display significantly impaired memory CD8+T cell response and weakened resistance against listeria monocytogenes(L.m.)infection.Mechanistically,eosinophils secrete interleukin-4(IL-4)to inhibit JNK/Caspase-3 dependent apoptosis of CD8+T cells upon L.m.infection in vitro.Furthermore,active eosinophils are recruited into the spleen and secrete more IL-4 to suppress CD8+T cell apoptosis during early stage of L.m.infection in vivo.Adoptive transfer of wild-type(WT)eosinophils but not IL-4-deficient eosinophils into eosinophil-deficient mice could rescue the impaired CD8+T cell memory responses.Together,our findings suggest that eosinophil-derived IL-4 promotes the generation of CD8+T cell memory and enhances immune defense against L.m.infection.Our study reveals a new adjuvant role of eosinophils in memory T cell generation and provides clues for enhancing the vaccine potency via targeting eosinophils and related cytokines.

A pH-Regulated Colorimetric Sensor Array for Discrimination of Biothiols Based on Two Different-Sized β-Cyclodextrin-Functionalized AuNPs

The abnormal changes of biothiols are directly related to health condition of human body,and the effective identification and quantification of biothiols is of great significance for screening and diagnosis of disease.This study described the development of a p H-regulated colorimetric sensor array for discrimination of five kinds of biothiols including cysteine(Cys),glutathione(GSH),homocysteine(Hcy),cysteamine(CA)and N-acetylcysteine(NAC).The proposed sensor array was established using 5-nm-and 20-nm-sizedβ-cyclodextrin-functionalized gold nanoparticles(β-CD@Au NPs)as nonspecific receptor and signal transduction elements.Due to the different binding affinity between biothiols andβ-CD@Au NPs in various p H environments,the different aggregation behaviors of nanoparticles produced unique colorimetric response patterns,which were able to be distinguished by bare eyes and UV-vis spectrophotometer.Accordingly,principal component analysis(PCA)and hierarchical cluster analysis(HCA)were employed for pattern recognition and generated a clustering map for a clear differentiation of biothiols at the level ofμmol/L.Furthermore,it can be proved that the method was successfully applied to the analysis of biothiols in human urine samples.

Clinical Analysis and Mental Health Survey of Hemophilia Carriers:a Cross-sectional Study

Objective:Hemophilia carriers(HCs),who are heterozygous for mutations in the clotting factor VIII/clotting factor IX gene(F8 or F9),may have a wide range of clotting factor levels,from very low,similar to afflicted males,to the upper limit of normal,and may experience mental health issues.The purpose of this study was to provide genetic information on mothers of hemophilia patients and to understand the clotting factor activity and phenotype of HCs.Additionally,we aimed to investigate the mental health status of HCs in China.Methods:A total of 127 hemophilia mothers,including 93 hemophilia A(HA)mothers and 34 hemophilia B(HB)mothers,were enrolled in this study.Long distance PCR,multiplex PCR,and Sanger sequencing were used to analyze mutations in F8 or F9.Coagulation factor activity was detected by a one-stage clotting assay.The Symptom Checklist 90(SCL-90,China/Mandarin version)was given to HCs at the same time to assess their mental health.Results:A total of 90.6%of hemophilia mothers were diagnosed genetically as carriers,with inversion in intron 22 and missense mutations being the most common mutation types in HA and HB carriers,respectively.The median clotting factor level in carriers was 0.74 IU/mL(ranging from 0.09 to 1.74 IU/mL)compared with 1.49 IU/mL(ranging from 0.93 to 1.89 IU/mL)in noncarriers,of which 14.3%of HCs had clotting factor levels of 0.40 IU/mL or below.A total of 53.8%(7/13)of HA carriers with low clotting factor levels(less than 0.50 IU/mL)had a history of bleeding,while none of the HB carriers displayed a bleeding phenotype.The total mean score and the global severity index of the SCL-90 for surveyed HCs were 171.00(±60.37)and 1.78(±0.59),respectively.A total of 67.7%of the respondents had psychological symptoms,with obsessive-compulsive disorder being the most prevalent and severe.The pooled estimates of all nine factors were significantly higher than those in the general population(Plia mothers was 90.6%,with a median cl

Langasite-based SAW high-temperature vibration sensor with temperature decoupling

Monitoring high-temperature vibrations is critical in aerospace engineering, industrial manufacturing, and energy production,where harsh conditions necessitate robust vibration sensors for detecting anomalous signals. However, the accuracy of such sensors is often compromised by crosstalk between temperature and vibration signals. This study introduces a high-temperature vibration sensor based on langasite(LGS) surface acoustic wave(SAW) technology, designed to withstand temperatures up to500°C. The sensor demonstrates high sensitivity, ranging from 12.54 kHz/g at 25°C to 15.63 k Hz/g at 500°C. A comprehensive mechanical and electrical coupling model for the SAW vibration sensor was developed by integrating theoretical equations with numerical simulations to optimize the sensor's performance. Additionally, a novel decoupling algorithm for temperature and vibration was established, achieving thermomechanical decoupling with precise vibration parameters. Experimental results indicated a maximum relative deviation of 4.67% for the algorithm. In conclusion, the proposed LGS SAW vibration sensor emerges as a promising solution for the accurate detection of multiple parameters in high-temperature vibration monitoring.

Nanostructured materials with localized surface plasmon resonance for photocatalysis

Localized surface plasmon resonance (LSPR) enhanced photocatalysis has fascinated much interest and considerable efforts have been devoted toward the development of plasmonic photocatalysts. In the past decades, noble metal nanoparticles (Au and Ag) with LSPR feature have found wide applications in solar energy conversion. Numerous metal-based photocatalysts have been proposed including metal/semiconductor heterostructures and plasmonic bimetallic or multimetallic nanostructures. However, high cost and scarce reserve of noble metals largely limit their further practical use, which drives the focus gradually shift to low-cost and abundant nonmetallic nanostructures. Recently, various heavily doped semiconductors (such as WO_(3-x), MoO_(3-x), Cu_(2-x)S, TiN) have emerged as potential alternatives to costly noble metals for efficient photocatalysis due to their strong LSPR property in visible-near infrared region. This review starts with a brief introduction to LSPR property and LSPR-enhanced photocatalysis, the following highlights recent advances of plasmonic photocatalysts from noble metal to semiconductor-based plasmonic nanostructures. Their synthesis methods and promising applicability in plasmon-driven photocatalytic reactions such as water splitting, CO_(2) reduction and pollution decomposition are also summarized in details. This review is expected to give guidelines for exploring more efficient plasmonic systems and provide a perspective on development of plasmonic photocatalysis.

Evaluation of Well Spacing for Primary Development of Fractured Horizontal Wells in Tight Sandstone Gas Reservoirs

Methods for horizontal well spacing calculation in tight gas reservoirs are still adversely affected by the complexity of related control factors,such as strong reservoir heterogeneity and seepage mechanisms.In this study,the stress sensitivity and threshold pressure gradient of various types of reservoirs are quantitatively evaluated through reservoir seepage experiments.On the basis of these experiments,a numerical simulation model(based on the special seepage mechanism)and an inverse dynamic reserve algorithm(with different equivalent drainage areas)were developed.The well spacing ranges of Classes I,II,and III wells in the Q gas field are determined to be 802–1,000,600–662,and 285–400 m,respectively,with their average ranges as 901,631,and 342.5 m,respectively.By considering both the pairs of parallel well groups and series well groups as examples,the reliability of the calculation results is verified.It is shown that the combination of the two models can reduce errors and provide accurate results.

Dosing low-level ferrous iron in coagulation enhances the removal of micropollutants, chlorite and chlorate during advanced water treatment

Drinking water utilities are interested in upgrading their treatment facilities to enhance micropollutant removal and byproduct control.Pre-oxidation by chlorine dioxide(ClO_(2))followed by coagulation-flocculation-sedimentation and advanced oxidation processes(AOPs)is one of the promising solutions.However,the chlorite(ClO_(2)^(-))formed from the ClO_(2) preoxidation stage cannot be removed by the conventional coagulation process using aluminum sulfate.ClO_(2)^(–)negatively affects the post-UV/chlorine process due to its strong radical scavenging effect,and it also enhances the formation of chlorate(ClO_(3)^(–)).In this study,dosing micromolar-level ferrous iron(Fe(II))into aluminum-based coagulants was proposed to eliminate the ClO_(2)^(–)generated from ClO_(2) pre-oxidation and benefit the post-UV/chlorine process in radical production and ClO_(3)^(–)reduction.Results showed that the addition of 52.1-μmol/L FeSO_(4) effectively eliminated the ClO_(2)^(-)generated from the pre-oxidation using 1.0 mg/L(14.8μmol/L)of ClO 2.Reduction of ClO_(2)^(-)increased the degradation rate constant of a model micropollutant(carbamazepine)by 55.0%in the post-UV/chlorine process.The enhanced degradation was verified to be attributed to the increased steady-state concentrations of HO^(-)·and ClO_(2)·by Fe(II)addition.Moreover,Fe(II)addition also decreased the ClO_(3)^(–)formation by 53.8%in the UV/chlorine process and its impact on the formation of chloroorganic byproducts was rather minor.The findings demonstrated a promising strategy to improve the drinking water quality and safety by adding low-level Fe(II)in coagulation in an advanced drinking water treatment train.

BACKGROUND Prohibitin 1(PHB1)has been identified as an antiproliferative protein that is highly conserved and ubiquitously expressed,and it participates in a variety of essential cellular functions,including apoptosis,cell cycle regulation,prolifera-tion,and survival.Emerging evidence indicates that PHB1 may play an important role in the progression of hepatocellular carcinoma(HCC).However,the role of PHB1 in HCC is controversial.AIM To investigate the effects of PHB1 on the proliferation and apoptosis of human HCC cells and the relevant mechanisms in vitro.METHODS HCC patients and healthy individuals were enrolled in this study according to the inclusion and exclusion criteria;then,PHB1 levels in the sera and liver tissues of these participates were determined using EliSA,RT-PCR,and immunohistoche-mistry.Human HepG2 and SMMC-7721 cells were transfected with the pEGFP-PHB1 plasmid and PHB1-specific shRNA(shRNA-PHB1)for 24-72 h.Cell prolif-eration was analysed with an MTT assay.Cell cycle progression and apoptosis were analysed using flow cytometry(FACS).The mRNA and protein expression levels of the cell cycle-related molecules p21,Cyclin A2,Cyclin E1,and CDK2 and the cell apoptosis-related molecules cytochrome C(Cyt C),p53,Bcl-2,Bax,caspase 3,and caspase 9 were measured by real-time PCR and Western blot,respectively.RESULTS Decreased levels of PHB1 were found in the sera and liver tissues of HCC patients compared to those of healthy individuals,and decreased PHB1 was positively correlated with low differentiation,TNM stage III-IV,and alpha-fetoprotein≥400μg/L.Overexpression of PHB1 significantly inhibited human HCC cell proliferation in a time-dependent manner.FACS revealed that the overexpression of PHB1 arrested HCC cells in the G0/G1 phase of the cell cycle and induced apoptosis.The proportion of cells in the G0/G1 phase was significantly increased and the proportion of cells in the S phase was decreased in HepG2 cells that were transfected with pEGFP-PHB1 compared with untreated control and empty vector-trans

Objective This study aimed to understand the epidemic status and phylogenetic relationships of rotavirus group A(RVA)in the Pearl River Delta region of Guangdong Province,China.Methods This study included individuals aged 28 days–85 years.A total of 706 stool samples from patients with acute gastroenteritis collected between January 2019 and January 2020 were analyzed for 17 causative pathogens,including RVA,using a Gastrointestinal Pathogen Panel,followed by genotyping,virus isolation,and complete sequencing to assess the genetic diversity of RVA.Results The overall RVA infection rate was 14.59%(103/706),with an irregular epidemiological pattern.The proportion of co-infection with RVA and other pathogens was 39.81%(41/103).Acute gastroenteritis is highly prevalent in young children aged 0–1 year,and RVA is the key pathogen circulating in patients 6–10 months of age with diarrhea.G9P[8](58.25%,60/103)was found to be the predominant genotype in the RVA strains,and the 41 RVA-positive strains that were successfully sequenced belonged to three different RVA genotypes in the phylogenetic analysis.Recombination analysis showed that gene reassortment events,selection pressure,codon usage bias,gene polymorphism,and post-translational modifications(PTMs)occurred in the G9P[8]and G3P[8]strains.Conclusion This study provides molecular evidence of RVA prevalence in the Pearl River Delta region of China,further enriching the existing information on its genetics and evolutionary characteristics and suggesting the emergence of genetic diversity.Strengthening the surveillance of genotypic changes and gene reassortment in RVA strains is essential for further research and a better understanding of strain variations for further vaccine development.

Qualitative exploration of home life experiences and care needs among elderly patients with temporary intestinal stomas

BACKGROUND This study employed a phenomenological research approach within qualitative research to explore the challenges encountered by elderly individuals with temporary colostomies in managing their daily lives and care needs.Protecting the anus surgery combined with temporary colostomy has emerged as a prevalent treatment modality for low rectal cancer.However,the ileostomy is susceptible to peri-stoma skin complications,as well as fluid,electrolyte,and nutritional imbalances,posing challenges to effective management.The successful selfmanagement of patients is intricately linked to their adjustment to temporary colostomy;nonetheless,there remains a dearth of research examining the factors influencing self-care among temporary colostomy patients and the obstacles they confront.AIM To investigate the lived experiences,perceptions,and care requirements of temporary colostomy patients within their home environment,with the ultimate goal of formulating a standardized management protocol.METHODS Over the period of June to August 2023,a purposive sampling technique was utilized to select 12 patients with temporary intestinal stomas from a tertiary hospital in Shanghai,China.Employing a phenomenological research approach,a semi-structured interview guide was developed,and qualitative interviews were conducted using in-depth interview techniques.The acquired data underwent coding,analysis,organization,and summarization following Colaizzi’s seven-step method.RESULTS The findings of this study revealed that the experiences and needs of patients with temporary intestinal stomas can be delineated into four principal themes:Firstly,Temporary colostomy patients bear various burdens and concerns about the uncertainty of disease progression;secondly,patients exhibit limited self-care capabilities and face information deficits,resulting in heightened reliance on healthcare professionals;thirdly,patients demonstrate the potential for internal motivation through proactive self-adjustment;and finally,patients express a significa
