Low prevalence of germline hmSH6 mutations in colorectal cancer families from Spain

AIm: To investigate the prevalence and penetrance of hmSH6 mutations in Spanish HNPCC families that was negative for mutation in hmLH1 or ***: We used PCR-based DGGE assay and direct Sequencing to screen for hmSH6 gene in 91 HNPCC ***: we have identified 10 families with germ-line mutations in the DNA sequence. These mutations included two intronic variation, three missense mutation, one nonsense mutation, and four silent mutations. Among the 10 germ-line mutations identified in the Spanish cohort,8 were novel, perhaps, suggesting different mutational spectra in the Spanish population. detailed pedigrees were constructed for the three families with a possible pathogenic hmSH6 mutation. The two silent mutations H388H and L758L, detected in a person affected of colorectal cancer at age 29, produce loss of the wild-type allele in the tumor sample. Immunohistochemical analysis showed that expression of mSH6 protein was lost only in the tumors from the carriers of V878A and Q263X ***: Altogether, our results indicate that disease-causing germ-line mutations of hmSH6 are very less frequent in Spanish HNPCC families.

Objective To determine whether topical applications of thiosulfinate-enriched Allium sativum extract(TASE)can accelerate acute cutaneous wound healing(WH)in a murine *** Keratinocyte viability and in vitro wound closure were assessed in keratinocyte *** of topical TASE(0.5μg/mL of allicin in 97%ethanol)on acute cutaneous WH were determined in a murine model of acute cutaneous *** mice were alternately assigned to the vehicle-and TASE-treated groups(n=6 per group).Expression levels of mRNA for keratinocyte differentiation marker-related proteins(filaggrin,loricrin and involucrin)and lipid synthetic enzymes(elongation of very long chain fatty acids protein 4(ELOVL4),fatty acid synthase(FA2H),3-hydroxy-3-methyl-glutaryl-coenzyme A reductase(HmGCoA),and serine palmitoyltransferase(SPT))were assessed using real-time quantitative polymerase chain reaction on day 3 and 8 after wounding,while transepidermal water loss(TEWL)rates were measured in wounded *** TASE accelerated WH both in vivo(40%vs.22%reduction in wound area,Pmoreover,topical applications of TASE upregulated the expression levels of epidermal mRNA for ELOVL4,HmGCoA,SPT,filaggrin,loricrin and involucrin(Pmparison with vehicle alone on day 8(33.06±2.09 g/(m^2·h)vs.24.60±2.04 g/(m^2·h),Pmulated keratinocyte proliferation and formation of epidermal permeability barrier function,leading to acceleration of acute cutaneous *** products containing TASE could be used to manage acute cutaneous WH.

The conversion of subalpine forests into grasslands for pastoral use is a well-knownphenomenon, although for most mountain areas the timing of deforestation has not been determined. The presence of charcoal fragments in soil profiles affected by shallow landsliding enabled us to date the occurrence of fires and the periods of conversion ofsubalpine forest into grasslands in the Urbión mountains, Iberian Range, Spain. We found that the treeline in the highest parts of the northwestern massifs of the Iberian Range(the Urbión, demanda, Neila, and Cebollera massifs) is currently between 1500 and 1600 m a.s.l., probably because of pastoral use of the subalpine belt, whereas in the past it would have reached almost the highest divides(at approximately 2100–2200 m a.s.l.). The radiocarbon dates obtained indicate that the transformation of the subalpine belt occurred during the late Neolithic, Chalcolithic, Bronze Age, Iron Age, and middle Ages. Forest clearing was probably moderate during fires prior to the middle Ages, as the small size of the sheep herds and the local character of the markets only required small clearings, and therefore more limited fires. Thus, it is likely that the forest recovered burnt areas in a few decades; this suggests the management of the forest and grasslands following a slash-andburn system. During the middle and modern Ages deforestation and grassland expansion affected most of the subalpine belt and coincided with the increasing prevalence of transhumance, as occurred in other mountains in the Iberian Peninsula(particularly the Pyrenees). Although the occurrence of shallow landslides following deforestation between the Neolithic and the Roman Period cannot be ruled out, the most extensive shallow landsliding processes would have occurred from the middle Ages until recent times.

In Arabidopsis thaliana, multiple genes involved in shoot apical meristem （SAm） transitions have been char- acterized, but the mechanisms required for the dynamic attainment of vegetative, inflorescence, and floral meristem （Vm, Im, Fm） cell fates during SAm transitions are not well understood. Here we show that a mADS-box gene, XAANTAL2 （XAL2/AGL14）, is necessary and sufficient to induce flowering, and its regula- tion is important in Fm maintenance and determinacy, xal2 mutants are late flowering, particularly under short-day （SD） condition, while XAL2 overexpressing plants are early flowering, but their flowers have vege- tative traits. Interestingly, inflorescences of the latter plants have higher expression levels of LFY, AP1, and TFL1 than wild-type plants. In addition we found that XAL2 is able to bind the TFL1 regulatory regions. On the other hand, the basipetal carpels of the 35S：：XAL2 lines lose determinacy and maintain high levels of WUS expression under SD condition. To provide a mechanistic explanation for the complex roles of XAL2 in SAm transitions and the apparently paradoxical phenotypes of XAL2 and other mADS-box （SOCl, AGL24） over- expressors, we conducted dynamic gene regulatory network （GRN） and epigenetic landscape modeling. We uncovered a GRN module that underlies Vm, Im, and Fm gene configurations and transition patterns in wild- type plants as well as loss and gain of function lines characterized here and previously. Our approach thus provides a novel mechanistic framework for understanding the complex basis of SAm development.

Impact of potato psyllid （Hemiptera： Triozidae） feeding on free amino acid composition in potato

The impacts of potato psyllid （Bactericera cockerelli） feeding on potato foliage on the free amino acids （FAAs） composition in potato leaf and tubers were determined under greenhouse conditions. The free amino acids in plant extracts were separated by high-performance liquid chromatography, and in both leaf and tuber samples, at least 17 FAAs were detected. Psyllid feeding significantly changed the levels of several FAAs in both leaf and tuber samples. The concentration of leucine increased 1.5-fold, whereas that of serine and proline increased 2- and 3-fold, respectively. In contrast, the concentrations ofglutamic acid, aspartic acid and lyscine were significantly reduced by 42.0%, 52.1% and 27.5%, respectively. There were also significant changes in the levels of FAAs in the Zebra chip （ZC） infected tubers compared with the healthy tubers, and the levels of six of the FAAs increased, and the levels of nine of the FAAs decreased. The results from this study indicate that potato psyllid causes major changes in free amino acid composition of plant tissues, and this change in plant metabolism may contribute to the plant stress as indicated by increased levels of proline in the leaves and hence promoting the development of plant diseases such as ZC disease.

OBJECTIVES Elderly patients show a higher incidence of ischemic and bleeding events after percutaneous transluminal coron-ary intervention(PCI).We sought to investigate outcomes in elderly patients treated with antithrombotic strategy guided by bleeding and ischemic risks after revascularization with last generation everolimus-eluting stent(EES).mETHODS Prospective multicenter registry including patients over 75 years revascularized with EES and antithrombotic ther-apy guided by clinical presentation,PCI complexity and PRECISE DAPT ***-primary safety endpoints were:(1)composite of cardiac death,myocardial infarction and stent thrombosis and;(2)bleeding(BARC 2-5).Primary efficacy endpoint was target lesion revascularization.A matched group of patients revascularized with current drug-eluting stents and no such tailored antith-rombotic therapy was used as *** Finally,1064 patients were included in SIERRA-75 cohort,80.8±4.2 years,36.6%women,71%acute coronary syn-dromes(ACS)and 53.6%complex ***-primary safety endpoint of major adverse cardiovascular events was met in 6.2%,co-primary safety endpoint of bleeding in 7.8%and primary efficacy endpoint of TKLR in 1.5%.The multivariable adjusted model showed no significant association of the prescribed short/long dual antiplatelet therapy(DAPT)durations with any endpoint suggesting a well tailored *** stent thrombosis reported in the subgroup with 1-3 months DAPT *** compared to control group,bleeding BARC 2-5 was significantly lower in SIERRA-75 group(7.4%vs.10.2%,P=0.04)as well as the net safety-efficacy endpoint(14.3%vs.18.5%,P=0.02).CONCLUSIONS In elderly population,the application of this risks-adjusted antithrombotic protocol after revascularization with last generation EES seems to be associated with an improved prognosis in terms of ischemic and bleeding outcomes.

Insights into the origin of precipitation moisture for tropical cyclones during rapid intensification process

In this study,we identiﬁed the moisture sources for the precipitation associated with tropical cyclones(TCs)during the rapid intensiﬁcation(RI)process from 1980 to 2018 by applying a lagrangian moisture source diagnostic *** detected sixteen regions on a global scale for RI events distributed as follows:four in the North Atlantic(NATL),two in the Central and East Paciﬁc Ocean(NEPAC),the North Indian Ocean(NIO)and South Indian Ocean(SIO),and three in the South Paciﬁc Ocean(SPO)and the Western North Paciﬁc Ocean(WNP).The moisture uptake(mU)mostly was from the regions where TCs underwent *** Western NATL,tropical NATL,Caribbean Sea,the Gulf of mexico and the Central America and mexico landmass supported~85.4%of the precipitating moisture in the NATL,while the latter source and the eastern North Paciﬁc Ocean provided the higher amount of moisture in NEPAC(~84.3%).The Arabian Sea,the Bay of Bengal and the Indian Peninsula were the major moisture sources in NIO,contributing approximately 81.3%.The eastern and western parts of the Indian Ocean supplied most of the atmospheric humidity in SIO(~83.8%).The combined contributions(~87.9%)from the western and central SPO and the Coral Sea were notably higher in ***,TCs in the WNP basin mostly received moisture from the western North Paciﬁc Ocean,the Philippine Sea and the China Sea,accounting for 80.1%.The remaining moisture support in each basin came from the summed contributions of the remote ***,RI TCs gained more moisture up to 2500 km from the cyclone centre than those slow intensiﬁcation(SI)and the total mU was approximately three times higher during RI than ***,the patterns of the mU differences respond to the typical pathways of moisture transport in each basin.

AIm:To evaluates the effectiveness and safety of the first generation,NS3/4A protease inhibitors(PIs) in clinical practice against chronic C virus,especially in patients with advanced fibrosis. mETHODS:Prospective study and non-experimental analysis of a multicentre cohort of 38 Spanish hospitals that includes patients with chronic hepatitis C genotype 1,treatment-na?ve(TN) or treatment-experienced(TE),who underwent triple therapy with the first generation NS3/4A protease inhibitors,boceprevir(BOC) and telaprevir(TVR),in combination with pegylated interferon and ribavirin. The patients were treatment in routine practice settings. Data on the study population and on adverse clinical and virologic effects were compiled during the treatment period and during follow ***:One thousand and fifty seven patients were included,405(38%) were treated with BOC and 652(62%) with TVR. Of this total,30%(n = 319) were TN and the remaining were TE:28%(n = 298) relapsers,12%(n = 123) partial responders(PR),25%(n = 260) null-responders(NR) and for 5%(n = 57) with prior response unknown. The rate of sustained virologic response(SVR) by intention-to-treatment(ITT) was greater in those treated with TVR(65%) than in those treated with BOC(52%)(P modified intention-to-treatment(m ITT) no were found significant differences. By degree of fibrosis,56% of patients were F4 and the highest SVR rates were recorded in the non-F4 patients,both TN and TE. In the analysis by groups,the TN patients treated with TVR by ITT showed a higher SVR(P = 0.005). However,by m ITT there were no significant differences between BOC and TVR. In the multivariate analysis by m ITT,the significant SVR factors were relapsers,IL28 B CC and non-F4; the type of treatment(BOC or TVR) was not significant. The lowest SVR values were presented by the F4-NR patients,treated with BOC(46%) or with TVR(45%). 28% of the patients interrupted the treatment,mainly by non-viral response(51%):this outcome was
