AIM:To study the association between inflammatory bowel disease(IBD) and genetic variations in eosinophil protein X(EPX) and eosinophil cationic protein(ECP).METHODS:DNA was extracted from ethylene diamine tetraacetic acid blood of 587 patients with Crohn's disease(CD),592 with ulcerative colitis(UC) and 300 healthy *** EPX405(G > C,rs2013109),ECP434(G > C,rs2073342) and ECP562(G > C,rs2233860) gene polymorphisms were analysed,by the 5'-nuclease allelic discrimination *** determination of intracellular content of EPX and ECP in granulocytes,39 blood samples was collected and extracted with a buffer containing cetyltrimethylammonium *** intracellular content of EPX was analysed using an enzyme-linked immunosorbent *** intracellular content of ECP was analysed with the UniCAP system as described by the *** tests for calculations of results were χ 2 test,Fisher's exact test,ANOVA,Student-Newman-Keuls test,and Kaplan-Meier survival curve with log-rank test for trend,the probability values of P lly ***:The genotype frequency for males with UC and with an age of disease onset of ≥ 45 years(n = 57) was for ECP434 and ECP562,GG = 37%,GC = 60%,CC = 4% and GG = 51%,GC = 49%,CC = 0% *** was significantly different from the healthy subject's genotype frequencies of ECP434(GG = 57%,GC = 38%,CC = 5%;P = 0.010) and ECP562(GG = 68%,GC = 29%,CC = 3%;P = 0.009).The genotype frequencies for females,with an age of disease onset of ≥ 45 years with CD(n = 62),was for the ECP434 and ECP562 genotypes GG = 37%,GC =52%,CC = 11% and GG = 48%,GC = 47% and CC = 5% *** was also statistically different from healthy controls for both ECP434(P = 0.010) and ECP562(P = 0.013).The intracellular protein concentration of EPX and ECP was calculated in μg/10 6 eosinophils and then correlated to the EPX 405 *** protein content of EPX was highest in the patients with the C

Immune oppression array elucidating immune escape and survival mechanisms in uveal melanoma

AIM： To examine the genetic profile of primary uveal melanoma （UM） as compared to UM in immune ***： Dendritic cells （DC） loaded with lysates of UM cells of high metastatic potential were used to stimulate CTls（CTls）. When CTls co-cultured with the UM cells, most UM cells could be eliminated. Survival UM cells grew slowly and were considered to be survival variants and examined by a microarray analysis. These differential genes were analyzed further with Venn Diagrams and functions related to immune escape. We additionally examined transcriptional changes of manually selected survival variants of UM cells and of clinical UM samples by quantitative real-time polymerase chain reaction （qRT-PCR）, and analyzed the correlation of these expressions and patients’ ***： Gene expression analyses revealed a marked up-regulation of SlAMF7 and CCl22 and a significant down-regulation of KRT10, FXYD3 and ABCC2. The expression of these genes in the relapsed UM was significantly greater than those in primary UM. UM patients with overexpression of these genes had a shorter survival period as compared with those of their ***： Gene expression, in particular of SlAMF7, CCl22, KRT10, FXYD3 and ABCC2, differed between primary UM cells and survival variants of UM cells.

Robot impedance control and passivity analysis with inner torque and velocity feedback loops

Impedance control is a well-established technique to control interaction forces in robotics. However, real implementations of impedance control with an inner loop may suffer from several limitations. In particular, the viable range of stable stiffness and damping values can be strongly affected by the bandwidth of the inner control loops （e.g., a torque loop） as well as by the filtering and sampling frequency. This paper provides an extensive analysis on how these aspects influence the stability region of impedance parameters as well as the passivity of the system. This will be supported by both simulations and experimental data. Moreover, a methodology for designing joint impedance controllers based on an inner torque loop and a positive velocity feedback loop will be presented. The goal of the velocity feedback is to increase （given the constraints to preserve stability） the bandwidth of the torque loop without the need of a complex controller.

3D Printing Technology for Tapered Optical Fiber Protection With Gas Sensing Possibilities

We present a new procedure for protecting micro-optical fibers(tapered fibers)by using the 3-dimensional(3D)printing technology.A standard single-mode optical fiber was tapered down to the diameter of 1mm and embedded in a polymeric matrix obtained by an additive manufacturing *** show that the proposed structure protects the fiber taper against environmental humidity while keeping permeability to gas flow and the possibility of the realization of gas detection *** our knowledge,this is the first time 3D printed casings were applied to protect fiber tapers from humidity *** envisage this new approach will allow the development of new fiber taper devices to better resist in humid environments.

Histiocytes have a pivotal role in wound repair and intestinal epithelial recovery-the most important goal to sustain gut ***,an in vivo description of colonic histiocytes by confocal laser endomicroscopy(ClE) is ***,we report the case of a 45-yearsold male patient who was referred to our clinic with weight loss and a history of two consecutive Clostridium difficile colitis episodes,the latter cured 3 wk before present *** microbiology was *** colonoscopy showed atrophy and a light mucosal oedema in the distal *** on-going endoscopy,we performed a fluorescein-aided ClE which revealed large polygonal(histiocytes-like) cells with copious cytoplasm and large nuclei in the lamina propria of the sigmoid colon as well as regenerative epithelial *** assessment of biopsies from the same areas confirmed the endomicroscopical findings:Periodic acid-Schiff-and CD68-positive foamy histiocytes in the colonic lamina propria and an advanced epithelial *** stool microbiology was repeatedly negative and polymerase chain reaction-analysis from colonic biopsies could not detect any mRNA for Thropheryma whippleii and common pathogens,we interpreted this particular setting as a mucosal healing process after consecutive Clostridium difficile *** conclusion,by describing these colonic histiocytes,we highlight the clinical usefulness of ClE in describing the entity of histiocytes in vivo and in real-time during the process of post-infectious mucosal healing in the colon.

Autoantibody profiles in autoimmune hepatitis and chronic hepatitis C identifies similarities in patients with severe disease

AIM To determine how the auto-antibodies(Abs) profiles overlap in chronic hepatitis C infection(CHC) and autoimmune hepatitis(AIH) and correlate to liver *** levels of antinuclear Ab, smooth muscle antibody(SMA) and liver/kidney microsomal-1(lKM-1) Ab and markers of liver damage were determined in the sera of 50 patients with CHC infection, 20 AIH patients and 20 healthy controls using enzyme linked immunosorbent assay and other immune assays. RESUlTS We found that AIH patients had more severe liver disease as determined by elevation of total Ig G, alkaline phosphatase, total serum bilirubin and serum transaminases and significantly higher prevalence of the three non-organ-specific autoantibodies(auto-Abs) than CHC patients. Antinuclear Ab, SMA and lKM-1 Ab were also present in 36% of CHC patients and related to disease severity. CHC cases positive for auto-Abs were directly comparable to AIH in respect of most markers of liver damage and total Ig G. These cases had longer disease duration compared with auto-Ab negative cases, but there was no difference in gender, age or viral load. KlM-1+ Ab CHC cases showed best overlap with AIH. CONClUSION Auto-Ab levels in CHC may be important markers of disease severity and positive cases have a disease similar to AIH. Auto-Abs might have a pathogenic role as indicated by elevated markers of liver damage. Future studies will unravel any novel associations between these two diseases, whether genetic or other.

Assisted reproductive technologies invoIving the use of spermatozoa and eggs for in vitro fertilization(IVF)have come as the solution for many infertile couples to become ***,in some cases,the use of ejaculated spermatozoa delivers poor IVF *** studies have suggested the use of testicular spermatozoa in severe male in fertility cases,but no guideli nes regarding their utilization are currently *** the present study,we found the mRNA protamine 1/protamine 2(P1/P2)ratio to be a valuable biomarker of poor sperm function that could be used as a diagnostic key for the identification of cases that would benefit from the use of testicular spermatozoa.A total of 23 couples undergoing egg donation cycles with at least one previous cycle failure were *** couples underwent two consecutive intracytoplasmic sperm injection(ICSI)cycles with either ejaculated or testicular spermatozoa(TESA).The sperm mRNA P1/P2 ratio,fertilization rate,blastocyst rate,and pregnancy and live birth rate were *** showed improved ICSI and clinical outcomes in cycles with testicular spermatozoa in men with altered mRNA P1/P2 *** cycles presented significantly higher rates of fertilization(mean±standard deviation:76.1%±15.1%vs 65.5%±18.8%),blastocyst formation(55.0%±20.3%vs 30.8%±23.8%),and good morphological quality blastocyst(28.9%±22.9%vs 13.5%±17.9%)and also improvements on pregnancy(60.9%vs 0%)and healthy birth rates(56.5%vs 0%)than EJACUlATE *** results described here suggest that in patients with previous IVF/ICSI failures and aberrant mRNA protamine ratios,the use of testicular spermatozoa may be a good alternative to improve clinical outcomes.
