Endonuclease-rolling circle amplification-based method for sensitive analysis of DNA-binding protein

A sensitive approach for the qualitative detection of DNA-binding protein on the microarray was developed. DNA complexes in which a partial duplex region is formed from a biotin-primer and a circle single strand DNA （ssDNA） were spotted on a microarray. The endonuclease recognition site （ERS） and the DNA-binding sites （DBS） were arranged side by side within the duplex region. The working principle of the detection system is described as follows： when the DNA-binding protein capture the DBS, the endonuclease could not attach to the ERS, and the immobilized primer in the DNA complex could be extended along the circle ssDNA by rolling circle amplification （RCA）. When no protein protects the DBS, the ERS could be attacked by the endonuclease and subsequently no rolling circle amplification occurs. Thereby we can detect the sequence specific DNA-binding activity with high-sensitivity due to the signal amplification of RCA.

There are lots of researches on fixture layout optimization for large thin-walled *** researches focus on the positioning problem,i.e.,optimizing the positions of a constant number of ***,how to determine the number of fixtures is *** most cases,the number of fixtures located on large thin-walled parts is determined based on engineering experience,which leads to huge fixture number and extra ***,this paper constructs an optimization model to minimize the number of *** constraints are set in the optimization model to ensure that the part deformation is within the surface profile *** addition,the assembly gap between two parts is also *** conduct the optimization,this paper develops an improved particle swarm optimization(IPSO)algorithm by integrating the shrinkage factor and adaptive inertia *** the algorithm,particles are encoded according to the fixture *** dimension of the particle is assigned to a sub-region by constraining the optional position range of each fixture to improve the optimization ***,a case study on ship curved panel assembly is provided to prove that our method can optimize the number of fixtures while meeting the assembly quality *** research proposes a method to optimize the number of fixtures,which can reduce the number of fixtures and achieve deformation control at the same time.

Background The priority effect of plant arrival is a key driver of community assembly and ecosystem succession during the restoration of degraded plant communities. However, the significance of the arrival order of different plant functional groups and their interactions with community assemblies remains unclear. Using a phytotron experiment with three fully crossed factors, we investigated the underlying mechanisms of priority effects and their relationships with the biomass and biodiversity effects in mixed plant communities by manipulating the order of arrival of species, isolation of roots, and removal of specific *** The results showed that the strength and direction of priority effects were influenced by arrival order, root interactions, asymmetric competition among species, and their interactions. The identities of early and late-sown species also determined the magnitude of priority effects. The priority effects were stronger in grass-first(24.76%) and legume-first communities(24.48%) than in forb-first communities. The pot biomass of the different priority treatments was highest in grass-first(5.85 g), followed by legume-first(3.94 g) and forb-first(2.48 g). The order of arrival in the mixture significantly affected the net biodiversity effects(P key determinant in successfully establishing a highly diverse community in the early stages of restoration. Simulation of gas-solid flow characteristics of the circulating fluidized bed boiler under pure-oxygen combustion conditions

Under the pressure of carbon neutrality,many carbon capture,utilization and storage technologies have witnessed rapid development in the recent years,including oxy-fuel combustion(OFC)***,the conventional OFC technology usually depends on the flue gas recirculation system,which faces significant investment,high energy consumption,and potential low-temperature corrosion *** these deficiencies,the direct utilization of pure oxygen to achieve particle fluidization and fuel combustion may reduce the overall energy consumption and CO_(2)-capture *** this paper,the fundamental structure of a self-designed 130 t·h^(-1) pure-oxygen combustion circulating fluidized bed(CFB)boiler was provided,and the computational particle fluid dynamics method was used to analyze the gas-solid flow characteristics of this new-concept boiler under different working *** results indicate that through the careful selection of design or operational parameters,such as average bed-material size and fluidization velocity,the pure-oxygen combustion CFB system can maintain the ideal fluidization state,namely significant internal and external particle ***,the contraction section of the boiler leads to the particle backflow in the lower furnace,resulting in the particle suspension concentration near the wall region being higher than that in the center ***,the upper furnace still retains the classic core-annulus flow *** addition to increasing solid circulation rate by reducing the average bed-material size,altering primary gas ratio and bed inventory can also exert varying degrees of influence on the gas-solid flow characteristics of the pure-oxygen combustion CFB boiler.

As a common hyperglycemic disease,type 1 diabetes mellitus(T1DM)is a complicated disorder that requires a lifelong insulin supply due to the immunemediated destruction of pancreaticβ*** it is an organ-specific autoimmune disorder,T1DM is often associated with multiple other autoimmune *** most prevalent concomitant autoimmune disorder occurring in T1DM is autoimmune thyroid disease(AITD),which mainly exhibits two extremes of phenotypes:hyperthyroidism[Graves'disease(GD)]and hypothyroidism[Hashimoto's thyroiditis,(HT)].However,the presence of comorbid AITD may negatively affect metabolic management in T1DM patients and thereby may increase the risk for potential diabetes-related ***,routine screening of thyroid function has been recommended when T1DM is ***,first,we summarize current knowledge regarding the etiology and pathogenesis mechanisms of both ***,an updated review of the association between T1DM and AITD is ***,we provide a relatively detailed review focusing on the application of thyroid ultrasonography in diagnosing and managing HT and GD,suggesting its critical role in the timely and accurate diagnosis of AITD in T1DM.

The glass-forming ability and mechanical properties of metallic glasses and their composites are well known to be sensitive to the preparation conditions and are highly deteriorated by industrial preparing conditions such as low-purity raw materials and low ***,we showed that a series of in-situ bulk metallic glass composites(BMGCs)which exhibit excellent ductility and segmental work hardening were successfully developed utilizing a high vacuum high-pressure die casting(HV-HPDC)technology along with industrial-grade raw *** tensile properties of these BMGCs are systematically investigated and correlated with the alloy *** compared with the copper mold suction casting method,the volume fraction difference of the dendrite phase for the BMGCs with the same composition is not significant when fabricated by the HV-HPDC,whereas the size of theβ-phase is generally *** BMGCs with the composition of Ti_(48)Zr_(20)(V_(12/17)Cu_(5/17))19 Be 13 obtained by the HV-HPDC process show ductility up to 11.3%under tension at room temperature and exhibit a certain amount of work *** conditions need to be met to enable the BMGCs,which are prepared by vacuum die-casting to retain favorable ductility:(1)The volume fraction ofβphase stays below 62%±2%;(2)The equiaxed crystals with a more uniform size in the range of 5-10μ***,the results of the present study provided guidance for developing BMGCs with good ductile properties under industrial conditions.

Inspired by the excellent adhesion performances of setae structure from organisms,micro/nano-pillar array has become one of the paradigms for adhesive *** micropillar arrays are composed of the resin pillars for adhesion and the substrate with different elastic modulus for *** stress singularity at the bi-material corner between the pillars and the substrate can induce the failure of the micropillar-substrate corner and further hinder the fabrication and application of micropillar arrays,yet the design for the stability of the micropillar array lacks systematical and quantitative *** this work,we develop a semi-analytical method to provide the full expressions for the stress distribution within the bi-material corner combining analytical derivations and numerical *** predictions for the stress within the singularity field can be obtained based on the full expressions of the *** good agreement between the predictions and the FEM results demonstrates the high reliability of our *** adopting the strain energy density factor approach,the stability of the pillar-substrate corner is assessed by predicting the failure at the *** the elastic mismatch between the pillar and substrate given in this paper,the stability can be improved by increasing the ratio of the shear modulus of the substrate to that of the *** study provides accurate predictions for the stress distribution at the bi-material corner and can guide the optimization of material combinations of the pillars and the substrate for more stable bioinspired dry adhesives.

The bound states around a vortex in anisotropic superconductors is a longstanding yet important *** this work,we develop a variational theory on the basis of the Andreev approximation to obtain the energy levels and wave functions of the low-energy quantized bound states in superconductors with anisotropic pairing on arbitrary Fermi *** the case of circular Fermi surface,the effective Schr¨odinger equation yielding the bound state energies gets back to the theory proposed by Volovik and Kopnin many years *** generalization here enables us to prove the equidistant energy spectrum inside a vortex in a broader class of *** importantly,we are now able to obtain the wave functions of these bound states by projecting the quasiclassical wave function on the eigenmodes of the effective Schr¨odinger equation,going beyond the quasiclassical Eilenberger results,which,as we find,are sensitive to the scattering *** the case of isotropic Fermi surface,the spatial profile of the low-energy local density of states is dominated near the vortex center and elongates along the gap antinode directions,in addition to the ubiquitous Friedel oscillation arising from the quantum inteference neglected in the Eilenberger ***,as a consequence of the pairing anisotropy,the quantized wave functions develop a peculiar distribution of winding number,which reduces stepwise towards the vortex *** work provides a flexible way to study the vortex bound states in the future.
