Acute effects of human protein S administration after traumatic brain injury in mice

Despite years of effort,no effective acute phase treatment has been discovered for traumatic brain *** impediment to successful drug development is entangled secondary injury *** we show that protein S,a natural multifunctional protein that regulates coagulation,inflammation,and apoptosis,is able to reduce the extent of multiple secondary injuries in traumatic brain injury,and therefore improve *** subjected to controlled cortical impact were treated acutely(10–15 minutes post-injury)with a single dose of either protein S(1 mg/kg)or vehicle phosphate buffered saline via intravenous *** 24 hours post-injury,compared to the non-treated group,the protein S treated group showed substantial improvement of edema and fine motor coordination,as well as mitigation of progressive tissue *** and western blot targeting caspase-3,B-cell lymphoma 2(Bcl-2)along with terminal deoxynucleotidyl transferase dUTP nick end labeling(TUNEL)assay revealed that apoptosis was suppressed in treated *** targeting CD11 b showed limited leukocyte infiltration in the protein S-treated ***,protein S treatment increased the ipsilesional expression of aquaporin-4,which may be the underlying mechanism of its function in reducing *** results indicate that immediate intravenous protein S treatment after controlled cortical impact is beneficial to traumatic brain injury *** Use Protocols(AUPs)were approved by the University Committee on Animal Resources(UCAR)of University of Rochester Medical Center(approval ***-2008-102 R)on November 12,2013.

Multiple description coding with spatial-temporal hybrid interpolation for video streaming in peer-to-peer networks

In this paper, we present an innovative design of multiple description coding with spatial-temporal hybrid interpola- tion (MDC-SThI) for peer-to-peer (P2P) video streaming. MDC can be effective in P2P networks because the nature of overlay routing makes path diversity more feasible. however, most MDC schemes require a redesign of video coding systems and are not cost-effective for wide deployment. We base our work on multiple state video coding, a form of MDC that can utilize standard codecs. Two quarter-sized video bit streams are generated as redundancies and embedded in the original-sized streams. With MDC-SThI, the nodes in P2P network can adjust the streaming traffic to satisfy the constraints of their devices and network environment. By design, the redundancies are used to compensate for missing frames, and can also be streamed independently to fulfill certain needs of low rate, low resolution applications. For better error concealment, optimal weights for spatial and temporal interpolation are determined at the source, quantized, and included in redundancies.

Exercise Catheterization for hemodynamic Evaluation of Adults with Coarctation of the Aorta

Background: Coarctation of the aorta (CoA) is associated with a generalized arteriopathy and long-term complicationsdespite repair. Data on invasive exercise hemodynamics in this population are lacking. Accordingly, wereviewed adults with CoA undergoing exercise catheterization to assess 1. hemodynamic profile;2. feasibilityfor assessment of CoA severity. Methods: Twenty patients undergoing exercise cardiac catheterization (12 armadduction and 8 supine cycle ergometry) at a quaternary care center between 2004 and 2021 were identified. Restingand exercise hemodynamic data were abstracted from the procedure logs. Results: Mean age was 43.6 ±12.0 years. Eleven patients (55%) had resting pulmonary arterial wedge pressure (PAWP) >15 mmhg;amongthose undergoing arterial catheterization, left ventricular end-diastolic pressure was >15 mmhg in 63%. Elevenpatients (55%) had pulmonary hypertension: 7 (35%) combined and 4 (20%) isolated post-capillary. At peak exercise,ΔPAWP/Δcardiac output (CO) ≥2 and Δmean pulmonary artery pressure/ΔCO ≥3 mmhg/l/min were foundin 7 (78%) and 6 (67%) patients, respectively;the composite of exercise PAWP ≥25 mmhg or ΔPAWP/ΔCO>2 was seen in 12 (86%). CoA peak-to-peak gradients at baseline (n = 14) and during exercise (n = 9) were 12(3–16) and 16 mmhg (9–28), respectively. Only 2 patients had an increase in CoA gradient to >20 mmhg withexercise. Conclusions: Diastolic dysfunction and pulmonary hypertension were highly prevalent, with exerciseunmasking abnormal diastolic and pulmonary vascular reserve in some individuals. Most patients failed to showsignificant increases in CoA peak-to-peak gradients with exercise. Further studies are warranted to establish thebest diagnostic method for CoA severity assessment.

Sorption of inorganic nanoparticles in woven cellulose fabrics

Titanium dioxide was deposited from aqueous suspension onto cellulosic surfaces. Titania was sourced from Degussa （P25TM, 70：30 anatase：rutile）. Dry uptake of particles was shown to be rapid and dominant with one-third of the deposition occurring in less than 30 s and over one-half in the first minute. Isotherms were recorded to compare the rate of titanium deposition on dry and pre-wetted cotton. In the dry case uptake reached a maximum in 30 min whereas in the pre-wetted case the uptake was seen to continue beyond 180 min. A broad trend of higher deposition occurring at lower ph was seen, corresponding to the region where surface charges were opposite and thus attractive. Dry pickup was less significant at high ph. The response to varying ionic strength was complex and was attributed to the combined effect of charge screening, particle aggregation and consequent particle entrapment or occlusion. Titania deposition into the interstices of woven cotton sheets resulted in the formation of inorganic, nanoparticulate skeletons which could be isolated by controlled combustion of the cellulose and thus cotton was suggested to have potential for the templated synthesis of high surface area semiconductor materials.

An update–tissue engineered nerve grafts for the repair of peripheral nerve injuries

Peripheral nerve injuries（PNI） are caused by a range of etiologies and result in a broad spectrum of disability. While nerve autografts are the current gold standard for the reconstruction of extensive nerve damage, the limited supply of autologous nerve and complications associated with harvesting nerve from a second surgical site has driven groups from multiple disciplines, including biomedical engineering, neurosurgery, plastic surgery, and orthopedic surgery, to develop a suitable or superior alternative to autografting. Over the last couple of decades, various types of scaffolds, such as acellular nerve grafts（ANGs）, nerve guidance conduits, and non-nervous tissues, have been filled with Schwann cells, stem cells, and/or neurotrophic factors to develop tissue engineered nerve grafts（TENGs）. Although these have shown promising effects on peripheral nerve regeneration in experimental models, the autograft has remained the gold standard for large nerve gaps. This review provides a discussion of recent advances in the development of TENGs and their efficacy in experimental models. Specifically, TENGs have been enhanced via incorporation of genetically engineered cells, methods to improve stem cell survival and differentiation, optimized delivery of neurotrophic factors via drug delivery systems（DDS）, co-administration of platelet-rich plasma（PRP）, and pretreatment with chondroitinase ABC（Ch-ABC）. Other notable advancements include conduits that have been bioengineered to mimic native nerve structure via cell-derived extracellular matrix（ECM） deposition, and the development of transplantable living nervous tissue constructs from rat and human dorsal root ganglia（DRG） neurons. Grafts composed of non-nervous tissues, such as vein, artery, and muscle, will be briefly discussed.

Aptamers,short single DNA or RNA oligonucleotides,have shown immense application potential as molecular probes for the early diagnosis and therapy of ***,conventional cell-SELEX technologies for aptamer discovery are time-consuming and *** we discovered a new aptamer BC-3 by using an improved rapid X-Aptamer selection process for human bladder carcinoma,for which there is no specific molecular probe *** show that BC-3 exhibited excellent affinity in bladder cancer cells but not normal *** demonstrate that BC-3 displayed high selectivity for tumor cells over their normal counterparts in vitro,in mice,and in patient tumor tissue *** endocytosis pathway analysis revealed that BC-3 internalized into bladder cancer cells via clathrin-mediated ***,we identified ribosomal protein S7(RPS7)as the binding target of BC-3 via an integrated methodology(mass spectrometry,colocalization assay,and immunoblotting).Together,we report that a novel aptamer BC-3 is discovered for bladder cancer and its properties in the disease are *** findings will facilitate the discovery of novel diagnostic and therapeutic strategies for bladder cancer.

The purpose of this review and commentary was to provide an historical and evidence-based account of organic acids and the biochemical and organic chemistry evidence for why cells do not produce metabolites that are *** scientific study of acids has a long history dating to the 16th and 17th centuries,and the definition of an acid was proposed in 1884 as a molecule that when in an aqueous solution releases a hydrogen ion(h^(+)).There are three common ionizable functional groups for molecules classified as acids:1)the carboxyl group,2)the phos-phoryl group and 3)the amine *** propensity by which a cation will associate or dissociate with a negatively charged atom is quantified by the equilibrium constant(K_(eq))of the dissociation constant(K_(d))of the ionization(K_(eq)=K_(d)),which for lactic acid(hLa)***(La^(-))is expressed as:K_(eq)=K_(d)=[h^(+)][La^(-)]/[hLa]=4677.3514(ionic strength=0.01 Mol⋅L^(-1),T=25℃).The negative log10 of the dissociation pKd reveals the ph at which half of the molecules are ionized,which for hLa=***,knowing the pKd and the ph of the solution at question will reveal the extent of the ionization *** of molecules that are classified as acids.

Astrocytic modulation of potassium under seizures
