Introduction: The use of growth factors and cytokines in skin rejuvenation and reversal of photo ageing is a novel anti-ageing treatment. These factors provide a microenvironment that seems to favor tissue repair and regeneration. Methods: An open label, monocentric, single arm study to evaluate anti-ageing efficacy of conditioned medium obtained from adult human bone-marrow derived allogenic mesenchymal stromal cells (BMMSC) along with excipients formulated into a cosmetic serum, was conducted in 40 Indian female population aged 35 to 60 years, belonging to skin type IV - VI of Norwood Scale for a duration of 90 days. Parameters—fine lines, wrinkles, crow’s feet, evenness of skin tone, skin firmness/laxity, hydration of skin, homogeneity of age spots, and visible pores were used to evaluate the anti-ageing and rejuvenating properties of the test product. Results and Discussion: Improvement was seen in majority of parameters starting from 15 - 30 days. Product was safe and well tolerated as per the dermatologist and subjects’ self-assessment.

Escalation of drug resistant microbes (bacteria) had forced researchers to search new and improved therapeutic compounds from different possible sources, including metabolites secreted by the actinomycetes. The aim of this study was to evaluate the pattern of antimicrobial actinomycetes from physiologically distinct soil of different geographical locations. Forty five soil samples were collected from 5 districts of Gujarat including two sanctuaries as source of survey for bioactive actinomycetes. Crowded plate technique was used for isolation and Agar cylinder method was employed for the antimicrobial screening. A total of 171 actinomycetes were isolated and screened against eighteen pathogens responsible for causing diseases in plants and humans. Results indicate that 79% of the isolates were active against at least one of the eighteen tested pathogens. Some of the actinomycetes strain had shown strong antibacterial and antifungal activity which may be a good source of obtaining novel antimicrobials.

AIM:To investigate lipopolysaccharide(LPS) related signal transduction in interstitial cells of Cajal(ICCs) from mouse small ***:For this study,primary culture of ICCs was prepared from the small intestine of the *** was treated to the cells prior to measurement of the membrane currents by using whole-cell patch clamp *** was used to examine the expression of the proteins in ***:LPS suppressed the pacemaker currents of ICCs and this could be blocked by AH6809,a prostaglandin E2-EP2 receptor antagonist or NG-Nitro-Larginine Methyl Ester,an inhibitor of nitric oxide(NO) ***-like receptor 4,inducible NO synthase or cyclooxygenase-2 immunoreactivity by specific antibodies was detected on ***(antioxidant agent) had no action on LPS-induced action in *** actions were blocked by nuclear factor kB(NF-kB) inhibitor,actinomycin D(a gene transcription inhibitor),PD 98059(a p42/44 mitogen-activated protein kinases inhibitor) or SB 203580 [a p38 mitogen-activated protein kinases(MAPK) inhibitor].SB 203580 also blocked the prostaglandin E2-induced action on pacemaker currents in ICCs but not ***:LPS inhibit the pacemaker currents in ICCs via prostaglandin E2-and NO-dependent mechanism through toll-like receptor 4 and suggest that MAPK and NF-kB are implicated in these actions.

Despite more than 300 theories to explain the aging process, oxidative stress theory offers the best mechanism to explain aging and age related disorders. Several studies has shown the importance of oxidative stress during aging. Pub Med, Science Direct and Springer online data bases are taken into consideration to write this mini-review. Human erythrocytes are most abundant and specialized cells in the body. Erythrocytes were extensively studied due to their metabolism and gas transport functions. Recent studies on erythrocytes have provided us detailed information of cell membrane and its structural organization that may help in studying the aging and age associated changes. The susceptibility of an organism is associated with the antioxidant potential of the body. Erythrocytes have potent antioxidant protection consisting of enzymatic and nonenzymatic pathways that counteract with reactive oxygen species, thus maintaining the redox regulation in the body. The non-enzymatic and enzymatic antioxidants and other biomarkers associated with erythrocyte membrane transport functions are the main content of this review. Biomarkers of oxidative stress in erythrocytes and its membrane were taken into the consideration during human aging that will be the main subject of this minireview.

目的:研究西藏长叶松树皮乙醇提取物(alcoholic extract of bark of Pinus roxburghii Sarg.,AEPR)的抗惊厥作用。方法:用最大电休克和戊四唑诱发白化 Wistar大鼠癫痫。最大电休克模型经由大鼠耳电极给予150mA 电流刺激0.2s诱发癫痫,以后肢强直性伸展持续时间的变化作为结局指标衡量 AEPR的抗惊厥作用,即后肢强直性伸展持续时间减少或停止。戊四唑模型大鼠经腹膜内注射戊四唑诱发肌阵挛性发作和阵发性抽搐,以痉挛发作的延迟衡量 AEPR的抗惊厥作用。结果:在最大电休克惊厥模型中,AEPR剂量分别为300和500mg/kg体质量,均显著减轻所有阶段的大鼠惊厥发作(P<0.01);标准对照药物苯妥英钠组用量25mg/kg,能显著减轻屈曲阶段的发作(P<0.01),并抑制所有阶段的惊厥发作。戊四唑惊厥模型大鼠于注射戊四唑前30min分别给予 AEPR 300和500mg/kg,均能显著延迟阵挛性发作(P<0.01)。100mg/kg体质量的 AEPR在戊四唑诱发的癫痫模型中没有显著的抗惊厥作用;标准对照药物地西泮4mg/kg能大幅度延迟阵挛性发作。结论:本研究提示 AEPR能有效抑制普遍强直性肌阵挛和部分癫痫发作。因此,AEPR对最大电休克及戊四唑诱发的大鼠癫痫有抗惊厥作用。然而需要进一步的研究确认是 AEPR中的何种成分对这种抗惊厥作用起主导作用。

Feasibility and oncological outcomes of laparoscopic rectal resection following neo-adjuvant chemo-radiotherapy: A systematic review

AIM: To study the feasibility and oncological outcomesfollowing laparoscopic total mesorectal excision(LTME) in patients who have received Neo-adjuvant long course chemo-radiotherapy(LCRT). METHODS: A protocol driven systematic review of published literature was undertaken to assess the feasibility and oncological outcomes following LTME in patients receiving LCRT. The feasibility was assessed using peri-operative outcomes and short term results. The oncological outcomes were assessed using local recurrence, disease free survival and overall ***: Only 8 studies-1 randomized controlled trial, 4 Case Matched/Controlled Studies and 3 Case Series were identified matching the search criteria. The conversion rate was low(1.2% to 28.1%), anastomotic leak rates were similar to open total mesorectal excision(0%-4.1% vs 0%-8.3%). Only 3 studies reported on local recurrence rates(5.2%-7.6%) at median 34 mo follow-up. A single study described disease free survival and overall survival at 3 years as 78.8% and 92.1% respectively. CONCLUSION: LTME following LCRT is feasible in experienced hands, with acceptable short term surgical outcomes and with the usual benefits associated with minimally invasive procedures. The long term oncological outcomes of LTME after LCRT appear to be comparable to open procedures but need further investigation.

A simple, controlled, robust and scalable three-stage manufacturing process of Afatinib Dimaleate was assessed and optimized leading to improved yield and quality. The synthetic process involves sequence of reactions as nitro-reduction, amidation and salification. The developed and optimized route was demonstrated on 300 g scale with over all isolated yield of 84% for Afatinib free base. The developed process has the capability to control not only the process related impurities but also the degradation impurities. One new impurity was identified during the process development studies and characterized as acetamide impunity, chemically known as (S)-N-(4-((3-chloro-4-fluorophenyl) amino)-7-((tetrahydrofuran-3-yl) oxy) quinazolin-6-yl) acetamide. Other impurities were identified as degradation impurities, Process impurities and were labeled as 1-(4-((3-chloro-4-fluorophenyl) amino)-7-(((S)-tetrahydrofuran-3-yl) oxy) quinazoline-6-yl)-5-Hydroxypyrrolidin- 2-one (hydroxy impurity), Afatinib N-Oxide impurity and N4-(3-chloro-4-fluorophenyl)-7-[[(3S)-tetrahydro- 3-furanyl] oxy]-4,6-quinazolinediamine (Intermediate-1).
