Regulatory elements and transcriptional control of chicken vasa homologue(CVH)promoter in chicken primordial germ cells

Background： Primordial germ cells（PGCs）, the precursors of functional gametes, have distinct characteristics and exhibit several unique molecular mechanisms to maintain pluripotency and germness in comparison to somatic cells. They express germ cel-specific RNA binding proteins（RBPs） by modulating tissue-specific cis-and trans-regulatory elements. Studies on gene structures of chicken vasa homologue（CVH）, a chicken RNA binding protein, involved in temporal and spatial regulation are thus important not only for understanding the molecular mechanisms that regulate germ cel fate, but also for practical applications of primordial germ cells. However, very limited studies are available on regulatory elements that control germ cel-specific expression in chicken. Therefore, we investigated the intricate regulatory mechanism（s） that governs transcriptional control of ***： We constructed green fluorescence protein（GFP） or luciferase reporter vectors containing the various 5′ flanking regions of CVH gene. From the 5′ deletion and fragmented assays in chicken PGCs, we have identified a CVH promoter that locates at-316 to ＋275 base pair fragment with the highest luciferase activity. Additional y, we confirmed for the first time that the 5′ untranslated region（UTR） containing intron 1 is required for promoter activity of the CVH gene in chicken PGCs. Furthermore, using a transcription factor binding prediction, transcriptome analysis and siR NA-mediated knockdown,we have identified that a set of transcription factors play a role in the PGC-specific CVH gene ***： These results demonstrate that cis-elements and transcription factors localizing in the 5′ flanking region including the 5′ UTR and an intron are important for transcriptional regulation of the CVH gene in chicken PGCs. Comparison of anterior segment optical coherence tomography findings in acanthamoeba keratitis and herpetic epithelial keratitis

This study is to investigate the characteristic features of Acanthamoeba keratitis（AK） that differentiating it from herpetic epithelial keratitis（HEK） using anterior segment optical coherence tomography（AS-OCT）. Medical records of three eyes of each AK and herpetic keratitis who had AS-OCT examination were reviewed in this study. Slitlamp biomicroscopy and AS-OCT was performed on the initial visit and on every follow-up visits in all patients. In all three AK cases, reflective bands in the corneal stroma that correspond to the area of radial keratoneuritis were observed. The depth of the reflective bands varied in each case. After AK treatment, slit-lamp biomicroscopy confirmed that radial keratoneuritis had resolved and AS-OCT confirmed that reflective bands in the corneal stroma had also disappeared in all patients. Unlike the AS-OCT results found in AK, highly reflective HEK lesions were observed only in the subepithelial area, not in the stroma. AS-OCT seems to be helpful analyzing the specific depth of the lesion which enables to distinguish AK from HEK.

In this work,we have prepared the hierarchically nanostructured core–shell NiCo layered double hydroxide(NiCo-LDH)nanosheets-and ZnFe_(2)O_(4) nanocubes-decorated polyacrylonitrile(PAN)/pitch-based carbon nanofibers(PPCNs)webs(NiCo-LDH@PPCNs as cathode and ZnFe_(2)O_(4)@PPCNs as anode materials)with the bonded network structure by a facile and scalable hydrothemal ***,the low-cost pitch with lower softening point(~90℃)as co-precursor was utilized to produce the PAN/pitch-based carbon nanofibers(PPCNs)with enhanced electrical *** obtained PPCNs with pitch content of 30%(PP30CNs)electrode material delivered higher specific capacitance(~67 F g^(-1))than that(~48 F g^(-1))of the PAN-based carbon nanofibers(PCNs)at 1 A g^(-1),due to the increased electrical conductivity and lower interfacial charge transfer resistance(RCT)of~0.16 ***,the NiCo-LDH-decorated PP30CNs(NiCoLDH@PP30CNs)as cathode material showed superior specific capacitance of 1162 F g^(-1) at 1.0 A g^(-1) and ultra-high retention rate of 91.56%at 10 A g^(-1).The ZnFe_(2)O_(4)@PP30CNs as anode material also showed higher specific capacitance of 282 F g^(-1) at 1 A g^(-1) and good rate capability with capacitance retention of 56.73%at 10 A g^(-1).The as-fabricated asymmetric NiCo-LDH@PP30CNs//ZnFe_(2)O_(4)@PP30CNs hybrid supercapacitor device delivered a specific capacitance of~98 F g^(-1) at 1 A g^(-1) and excellent capacitance retention of~88%after 5000 charge–discharge cycles.

Mutation of MED12 is not a frequent occurrence in prostate cancer of Korean patients

Prostate cancer is one of the major health care problems, but the molecular pathogenesis has been relatively insufficiently elucidated. Recently, whole exome sequencing of prostate cancer identified recurrent mutations involving MED12 in Caucasian patients, which finding was not reproduced in one subsequent study by Sanger sequencing. Thus, we investigated mutation status of MED12 in exons 2 and 26 by Sanger sequencing in 102 radical prostatectomy cases from Korean patients. The analysis found the mutation in none of the cases. Therefore, MED12 mutation does not appear to represent a significant molecular alteration in this cohort of patients according to the analysis by the traditional ＂gold standard.＂

3D-printing-assisted flexible pressure sensor with a concentric circle pattern and high sensitivity for health monitoring

In this study,a flexible pressure sensor is fabricated using polydimethylsiloxane(PDMS)with a concentric circle pattern(CCP)obtained through a fused deposition modeling(FDM)-type three-dimensional(3D)printer and poly(3,4-ethylenedioxythiophene):poly(styrenesulfonate)(PEDOT:PSS)as the active *** layer-by-layer additive manufacturing,the CCP surface is generated from a thin cone model with a rough surface by the FDM-type 3D printer.A novel compression method is employed to convert the cone shape into a planar microstructure above the glass transition temperature of a polylactic acid(PLA)*** endow the CCP surface with conductivity,PDMS is used to replicate the compressed PLA,and PEDOT:PSS is coated by *** size of the CCP is controlled by changing the printing layer height(PLH),which is one of the 3D printing *** sensitivity increases as the PLH increases,and the pressure sensor with a 0.16 mm PLH exhibits outstanding sensitivity(160 kPa^(-1)),corresponding to a linear pressure range of 0-0.577 kPa with a good linearity of R^(2)=0.978,compared to other *** pressure sensor exhibits stable and repeatable operation under various pressures and durability under 6.56 kPa for 4000 ***,monitoring of various health signals such as those for the wrist pulse,swallowing,and pronunciation of words is demonstrated as an *** results support the simple fabrication of a highly sensitive,flexible pressure sensor for human health monitoring.

The rapid development of emerging technologies observed in recent years,such as artificial intelligence, machine learning, mobile internet, big data,cloud computing, and the Internet of Everything, are generating escalatingdemands for expanding the capacity density, and speed in next-generation opticalcommunications. This poses a significant challenge to existing communicationtechniques. Within this context, the integration of near-infraredbroadband, tunable, and high-gain luminescent materials into silicon opticalcircuits or fiber architectures to transmit and modulate light shows enormouspotential for advancing next-generation communication techniques. Here, thisreview provides an overview of the recent breakthroughs in near-infrared luminescentepitaxial/colloidal quantum dots, and metal-active-center-doped materialsfor broadband optical amplifiers and tunable lasers. We also expound onefforts to enhance the bandwidth and gain of these materials-based amplifiersand lasers, exploring the challenges associate with developing ultra-broadbandand high-speed optical communication systems. Additionally, the potentialapplications in Fifth Generation Fixed Networks, integration with 5G and 6Gwireless networks, compensation for current Si electronic based CMOS forhigh computing capability, and the prospects of these light sources for nextgenerationoptoelectronic devices are discussed.

Implantable biomedical devices require an anti-biofouling,mechanically robust,low friction surface for a prolonged lifespan and improved ***,there exist no methods that could provide uniform and effective coatings for medical devices with complex shapes and materials to prevent immune-related side effects and thrombosis when they encounter biological ***,we report a lubricant skin(L-skin),a coating method based on the application of thin layers of bio-adhesive and lubricant-swellable perfluoropolymer that impart anti-biofouling,frictionless,robust,and heat-mediated self-healing *** demonstrate biocompatible,mechanically robust,and sterilization-safe L-skin in applications of bioprinting,microfluidics,catheter,and long and narrow medical *** envision that diverse applications of L-skin improve device longevity,as well as anti-biofouling attributes in biomedical devices with complex shapes and material compositions.

Pretransplant absolute monocyte count in peripheral blood predicts posttransplant tumor prognosis in patients undergoing liver transplantation for hepatocellular carcinoma

BACKGROUND: Preoperative absolute monocyte count in peripheral blood(AMCPB) is closely associated with prognoses in not only various malignancies but also hepatocellular carcinoma(HCC). The purpose of this study was to evaluate whether pretransplant AMCPB predicts posttransplant outcomes in patients with HCC undergoing liver transplantation(LT).METHOD: We retrospectively analyzed relationships between clinicopathologic factors involving pretransplant AMCPB and tumor recurrence or survival in 256 patients who had undergone LT for HCC between January 2005 and April ***: ROC curve analysis showed that AMCPB >200/mm3was a risk factor for tumor recurrence; 43 patients showed higher AMCPB(>200/mm3), whereas 213 showed lower AMCPB(≤200/mm3) at the time of LT. On multivariate analysis,pretransplant high AMCPB, positive findings in pretransplant18F-FDG PET/CT, pathological maximal tumor size >5 cm,intrahepatic metastasis, moderately or poorly differentiated tumor and microvascular invasion were independent factors affecting recurrence-free survival. When we performed subgroup analysis based on the Milan criteria, high AMCPB was an independent factor for predicting HCC recurrence in patients with tumor beyond the Milan criteria(P=0.004), and not for patients within the ***: This study demonstrated that pretransplant AMCPB could predict tumor recurrence after LT for HCC,especially in patients with tumor beyond the Milan criteria.

Graft-to-recipient weight ratio lower to 0.7% is safe without portal pressure modulation in right-lobe living donor liver transplantation with favorable conditions

BACKGROUND: The low graft-to-recipient weight ratio(GRWR) in adult-to-adult living donor liver transplantation(LDLT) is one of the major risk factors affecting graft survival. The goal of this study was to evaluate whether the lower limit of the GRWR can be safely reduced without portal pressure modulation in right-lobe LDLT. METHODS: From 2005 to 2011, 317 consecutive patients from a single institute underwent LDLT with right-lobe grafts without portal pressure modulation. Of these, 23 had a GRWR of less than 0.7%(group A), 27 had a GRWR of ≥0.7%, <0.8%(group B), and 267 had a GRWR of more than and equal to 0.8%(group C). Medical records, including recipient, donor, operation factors, laboratory findings and complications were reviewed retrospectively. RESULTS: The baseline demographics showed low model for end-stage liver disease score(mean 16.3±8.9) and high percentage of hepatocellular carcinoma(231 patients, 72.9%). Three groups by GRWR demonstrated similar characteristics except recipient body mass index and donor gender. For smallforsize syndrome, there were 3(13.0%) in group A, 1(3.7%) in group B, and 2 patients(0.7%) in group C(P<0.001). Hepatic artery thrombosis was more frequently observed in group A than in groups B and C(8.7% vs 3.7% vs 1.9%, P=0.047). However, among the three groups, graft survival rates at 1 year(100% vs 96.3% vs 93.6%) and 3 years(91.7% vs 73.2% vs 88.1%) were not different(P=0.539). In laboratory measurements,there was no group difference in total bilirubin and albumin. However, prothrombin time was longer in group A within postoperative 1 week and platelet count was lower in groups A and B within postoperative 1 month. CONCLUSION: A GRWR lower to 0.7% is safe and does not need to modulate portal pressure in adult-to-adult LDLT using the right-lobe in favorable conditions including low model for end-stage liver disease score.
