Distinct effect of potassium 2-(l-hydroxypentyl) - benzoate on hippocampal neurons, synapses and dystrophic axons in APP/PS1 mice

OBJECTIVE To study the protective effect of potassium 2-(l-hydroxypentyl)-benzoate(PHPB) on hippocampal neurons,synapses and dystrophic axons in APP/PS1 *** Ten-month-old male APP/PS1 transgenic mice and age-matched wild-type mice were randomly divided into three groups:wild-type group(WT Con group,n=10),APP/PS1 group(Tg Con group,n=10) and PHPB treated APP/PS1 group(PHPB group,n=10).PHPB group received 30 mg · kg-1 PHPB by oral gavage once daily for 3 *** Con group and Tg Con group received the same volume of *** months later,mice were sacrificed for biochemical and pathological testing such as transmission electron microscopy,Golgi staining and Western boltting *** Under the transmission electron microscope,most hippocampal neurons and subcel ular organel es in WT Con group exhibited normal ***,the degenerative changes were observed in Tg Con group such as nuclear fragmentation,mitochondrial swelling,ribosomes detachment and autophagic vacuoles *** hippocampal synapses number and the thickness of postsynaptic density(PSD) were significantly decreased in Tg Con group compared with the WT Con group(P<0.05).After PHPB treatment,the degenerative changes in APP/PS1 mice were alleviated to some *** synapse number has been elevated significantly(P<0.05) and the PSD has been thickened as *** staining showed that the spine density of secondary and tertiary apical dendritic branches was significantly decreased in CA1 and DG areas of Tg Con group(P<0.05).Sholl analysis revealed a decrease of dendritic complexity in Tg Con group compared with WT Con group(P<0.05).These abnormalities were alleviated to some extent after PHPB *** blotting study showed that the protein levels of synaptic marker PSD-95 and synaptophysin were significantly decreased in the hippocampus of Tg Con group(P<0.05).A significant increase of PSD-95(P<0.05) and a slight increase of SYP were observed after the PHPB t

Transcriptome sequencing and metabolome analysis reveal the mechanism of Shuanghua Baihe Tablet in the treatment of oral mucositis

Oral mucositis(OM) caused by cancer therapy is the most common adverse reaction in the radiotherapy of head and neck tumors. In severe cases, it can lead to the interruption of treatment, which affects the control of the disease and the quality of *** Baihe Tablet(SBT) is a traditional Chinese medicine(TCM) formula, which is administerd to treat OM in China. It has been clinically effective for more than 30 years, but the underlying mechanism is not completely understood. With the development of multiple omics, it is possible to explore the mechanism of Chinese herbal compound prescriptions. Based on transcriptomics and metabolomics, we explored the underlying mechanism of SBT in the treatment of OM. An OM model of rats was established by 5-FU induction, and SBT was orally administered at dosages of 0.75 and 3 g·kg;·d;. In order to search for SBT targets and related metabolites, the dysregulated genes and metabolites were detected by transcriptomics and metabolomics. Immune related indicators such as interleukin-17(IL-17) and tumor necrosis factor-α(TNF-α) were detected by ELISA. Treg cell disorders was analyzed by flow cytometry. Our results showed that SBT significantly alleviated the symptoms of OM rats and the inflammatory infiltration of ulcer *** SBT administration, inflammatory related metabolic pathways including linoleic acid metabolism, valine, leucine and isoleucine biosynthesis were significantly altered. Furthermore, the production of proinflammatory factors like IL-17 and TNF-α, were also dramatically reduced after SBT administration. Besides, the infiltration degree of Treg cells in the spleen of OM modeling rats was significantly improved by SBT administration, thus maintaining the immune balance of the body. The current study demonstrates that SBT regulates inoleic acid metabolism, glycerophospholipid metabolism and amino acid metabolism, and inhibits IL-17/TNF signal transduction to restore Treg and Th17 cell homeostasis in OM

Factors influencing vaginal cuff dehiscence after laparoscopic hysterectomy and the psychological state of the patients

BACKGROUND Despite laparoscopic total hysterectomy provided more benefits to patients and has become one of the most commonly used surgical methods for total ***,vaginal cuff dehiscence is a serious complication after laparoscopic total hysterectomy and so far,there are limited studies on vaginal cuff dehiscence after a total *** To investigate the factors influencing vaginal cuff dehiscence after laparoscopic hysterectomy and the psychological state of the *** This study retrospectively,univariately,and multivariately analyzed the clinical data of patients who experienced vaginal cuff dehiscence after laparoscopic hysterectomy in Yuyao People’s Hospital from January 2015 to December *** regression was utilized to analyze the high-risk factors of vaginal cuff *** Among 1459 cases with laparoscopic hysterectomy,9 reported vaginal cuff dehiscence postoperatively,with an incidence of 0.617%.Univariate logistic regression analysis revealed that preoperative hemoglobin value[odds ratio(OR)=5.12,P=0.016],vaginal cuff suture method(OR=0.26,P=0.048),and postoperative first sexual lifetime(OR=15.86,P=0.002)were associated with vaginal cuff dehiscence following laparoscopic *** logistic regression analysis revealed that preoperative hemoglobin value of<90 g/L(OR=5.17,P=0.015)and time interval between postoperative first sexual life and operation of<3 months(OR=54.00,P=0.004)are independent risk factors for vaginal cuff dehiscence after laparoscopic *** Postoperative hemoglobin and the time interval between postoperative first sexual life and operation were the independent factors of developing vaginal cuff dehiscence after laparoscopic hysterectomy.

Energy storage devices have been extensively owing to their critical role in addressing the energy and environment *** trivalent metal batteries are promising due to their unique three-electron redox reactions for high reversible capacity and high safety [1]. Especially, aqueous aluminum-based batteries have attracted significant attention [2]. However, severe water decomposition occurs due to their low redox potential (Al3+/Al,-1.66 V vs. SHE)[3]. In contrast, the metallic indium (In)/or In3+has a more suitable redox potential (-0.34 V vs. SHE), which is within the water stability window, minimizingwater decomposition [4]. Recently, a study on trivalent indium metal batteries was firstly reported by Wu’s group in Journal of the American Chemical Society [5].

Facilitation of microbialite development by continental weathering in the Cambrian Zhangxia Formation,southern North China Block

We report geochemical data for carbonate rocks from the Cambrian Zhangxia Formation,southern North China Block,including trace-element contents and C,O,and Sr isotopic compositions,to determine the controls on the development of microbialites in this *** sedimentary environment of the Zhangxia Formation evolved from carbonate-slope through platform-margin-reef to ooid-shoal,and then to open-platform settings,corresponding to the development of micrites,microbialites,and oolitic *** data show that δ13C values decrease upwards through the Zhangxia Formation,from 0.99‰to-1.59‰,whereas δ18O values increase from-9.91‰to-7.10‰.Total rare earth element （REE）contents decrease upwards through the stratigraphy,and the contents of Th and Sc show a similar trend.87Sr/86Sr ratios reach a maximum of 0.710544 in the horizons of microbialite development,Member 2 of the Zhangxia Formation,above which the ratios are uniform,with a mean value of *** these geochemical data with the inferred sedimentary evolution,the Zhangxia Formation is interpreted to have formed in a depositional environment that changed from transgression to *** weathering played a crucial role in promoting the input of terrigenous materials,which in turn favored the development of microbialites during the early stage of deposition of the Zhangxia Formation.

Anisotropy characterization of upper shanghai clays: Shear strength and small-strain stiffness

Comprehensive investigations have been conducted to study the structure and overconsolidation of upper Shanghai clays, i.e. Layers 2–6 clays, typically located at depths of 30–40 m. However, limited information is available on their anisotropy, and even less is known about the correlation between structure, overconsolidation, and anisotropy. In this study, the undrained anisotropy characteristics of shear strength and small-strain shear stiffness in upper Shanghai Layers 2–6 clays were thoroughly assessed using a series of K0-consolidated undrained triaxial compression (TC) and triaxial extension (TE) tests (K0 is the coefficient of lateral earth pressure at rest). The effective stress paths, shear strength, and small-strain shear stiffness from the undrained TC and TE tests demonstrate the anisotropic behaviors in upper Shanghai clays. Analyses of data from upper Shanghai clays and other clays worldwide indicate that the shear strength anisotropy ratio (Ks) converges at 0.8 as the overconsolidation ratio (OCR) and plasticity index (Ip) increase, while the small-strain shear stiffness anisotropy ratio (Re) converges at 1.0. The influence of OCR on Ks and Re is more pronounced than that of Ip and sensitivity (St). Nevertheless, no clear correlation between Ks and Re is observed in upper Shanghai clays.
