Recombinant adenovirus-p53(Gendicine) sensitizes a pancreatic carcinoma cell line to radiation

Objective： In this study, we examine the effects of recombinant adenovirus-p53 （rAd-p53） on the pancreatic carcinoma cell line Sw1990. Specifically, we determine if expression of rAd-p53 sensitizes these cells to radiation. Methods： Following transfection of Sw1990 cells with rAd-p53, we measured expression of P53, P21 and Bax by immunocytochemistry. Both transfected and control cell lines were irradiated with a range of doses, and the survival fractions （SF） were calculated. Dose survival cttrves were constructed and modeled for comparison. Results： Transfection of Sw1990 cells with rAd-p53 resulted in increased expression of P53, P21 and Bax in a time-dependent manner. At 96 h after transfection, 89.92% of cells expressed P53, 56.8% expressed P21, and 76.50% expressed Bax. The SF following radiation was lower in the rAd-p53 transfected cells compared to the control cells, suggesting that rAd-p53 sensitizes Sw1990 cells to radiation （Do for the experimental and control groups was 2.199 and 2.462, respectively）. Conclusions： Use of the adenoviral vector is an effective means of transfecting Sw1990 cells with wild-type P53, and this sensitizes the cell line to irradiation. This work suggests that combining rAd-p53 with radiation therapy in pancreatic cancer may be therapeutically beneficial.

Objectives:To investigate the efficacy of high-dose radiation therapy(RT) to the primary and regional disease in combination with systemic chemotherapy and local treatment to metastatic foci in patients with newly diagnosed metastatic nasopharyngeal carcinoma(NPC).Methods: One hundred and five consecutive patients with pathologically confirmed NPC with distant metastasis at diagnosis seen between 1995 and 2002 were *** were offered cisplatin-based chemotherapy, high-dose radiation therapy(>30 Gy) to the head and neck region and active treatment to the metastatic ***:Patients’ median age was 46 years,and all had a karnofsky Performance Score(kPS) of≥***-nine patients(85%) had metastases confined to one ***-six patients(91%) received at least one cycle of chemotherapy,and 71(68%) received >65 Gy of radiation to the head and neck *** a median follow-up time of 22 months(range 2-142 months),90 patients had deceased,and the median survival time of the entire group was 25 *** 2-and 5-year estimated overall survival (OS) rates were 50%and 17%,*** dose of > 65 Gy to the primary region(p=0.05) and number of organs with metastases(single versus multiple)(p=0.002) were independent predictive factors for OS on log-rank *** moderately severe acute toxicities,such as Radiation Therapy Oncology Group(RTOG) grade 3 mucositis,skin desquamation,and leucocytopenia were *** patient experienced Grade 4 acute ***:High dose radiation therapy is indicated for local disease control in patients with metastatic nasopharyngeal carcinoma,and may improve survival when actively used with systemic chemotherapy and local treatment for metastatic *** with single organ metastases have a better prognosis as compared to those with more widespread metastases.

AbTechnology has always driven advances in radiotherapy *** this review,we describe the main technological advances in radiotherapy over the past decades for the treatment of nasopharyngeal cancer(NPC)and highlight some of the pressing issues and challenges that *** aim to identify emerging trends in radiation *** include advances in personalized medicine and advanced imaging modalities,standardization of planning and delineation,assessment of treatment response and adaptive re-planning,impact of particle therapy,and role of artificial intelligence or automation in clinical *** conclusion,we expect significant improvement in the therapeutic ratio of radiotherapy treatment for NPC over the next decade.
