Preventive effect of a pectic polysaccharide of the common cranberry Vaccinium oxycoccos L. on acetic acid-induced colitis in mice

AIM： To study isolation and chemical characterization of pectin derived from the common cranberry Vaccin/um oxycoccos L. （oxycoccusan OP） and the testing of its preventive effect on experimental colitis. METHODS： Mice were administrated orally with OP two days prior to a rectal injection of 5% acetic acid and examined for colonic damage 24 h later. Colonic inflammation was characterized by macroscopical injury and enhanced levels of myeloperoxidase activity measured spectrophotometrically with o-phenylene diamine as the substrate. The mucus contents of the colon were determined by the Alcian blue dye binding method. Vascular permeability was estimated using 4% Evans blue passage after i.p. injection of 0.05 mol/L acetic acid. RESULTS： In the mice treated with OP, colonic macroscopic scores （1.1 ± 0.4 vs 2.7, P 〈 0.01） and the total square area of damage （10 ± 2 vs 21 ± 7, P 〈 0.01） were significantly reduced when compared with the vehicle-treated colitis group. OP was shown to decrease the tissue myeloperoxidase activity in colons （42 ± 11 vs 112 ± 40, P 〈 0.01） and enhance the amount of mucus of colitis mice （0.9 ± 0.1 vs 0.4 ± 0.1, P 〈 0.01）. The level of colonic malondialdehyde was noted to decrease in OP-pretreated mice （3.6 ± 0.7 vs 5.1 ± 0.8, P 〈 0.01）. OP was found to decrease the inflammatory status of mice as was determined by reduction of vascular permeability （161 ± 34 vs 241 ± 21, P 〈 0.01）. Adhesion of peritoneal neutrophils and macrophages was also shown to decrease after administration of OP （141 ± 50 vs 235 ± 37, P 〈 0.05）. CONCLUSION： Thus, a preventive effect of pectin from the common cranberry, namely oxycoccusan OP, on acetic acid-induced colitis in mice was detected. A reduction of neutrophil infiltration and antioxidant action may be implicated in the protective effect of oxycoccusan.

AIM: To investigate the inhibitory action of diet-derived phenolic compound gallic acid(GA) against HCT-15 colon cancer ***: The antiproliferative effect of GA against colon cancer cells was determined by performing thiazolyl blue tetrazolium bromide(MTT) assay. The colony forming ability of GA treated colon cancer cells was evaluated using the colony forming assay. The cell cycle changes induced by GA in HCT-15 cells were analyzed by propidium iodide staining. Levels of reactive oxygen species(ROS) and mitochondrial membrane potential of HCT-15 exposed to GA was assessed using 2',7'-dichlorfluorescein-diacetate and rhodamine-123 respectively, with the help of flow cytometry. Morphological changes caused by GA treatment in the colon cancer cells were identified by scanning electron microscope and photomicrograph examination. Apoptosis was confirmed using flow cytometric analysis of GA treated HCT-15 cells after staining with ***: MTT assay results illustrated that GA has an inhibitory effect on HCT-15 cells with IC50 value of 740 μmol/L. A time-dependent inhibition of colony formation was evident with GA treatment. Cell cycle arrest was evident from the accumulation of GA treated HCT-15 cells at sub-G1 phase(0.98 ± 1.03 vs 58.01 ± 2.05)with increasing exposure time. Flow cytometric analysis of GA treated HCT-15 cells depicted early events associated with apoptosis like lipid layer breakage and fall in mitochondrial membrane potential apart from an increase in the generation of ROS which were in a time dependent manner. SEM and photomicrograph images of the GA-treated cells displayed membrane blebbing and cell shrinking characteristics of apoptosis. Further apoptosis confirmation by Yo-Pro-1 staining also showed the time-dependent increase of apoptotic cells after ***: These results show that GA induced ROS dependent apoptosis and inhibited the growth of colon cancer cells.

This paper deals with gold extraction from a refractory concentrate by chlorine *** process considers a pre-treatment of refractory materials by low temperature oxidation carried out with low oxygen *** oxidized material is treated by leaching with *** gold adsorption/reduction onto activated carbon,iron and base metals can be precipitated by *** tests show the necessity to carry out a thermal pre-treatment at least at 550°C to obtain a reduction of sulfur and mercury in the material(50%and 90%,respectively).Highest gold extraction yield(around 93%)is obtained in the leaching test performed with material sample treated at 650°*** result confrms the necessity to optimize the thermal pre-treatment to improve Au recovery and to reduce chlorine consumption.A comparison with conventional cyanidation confrms that chlorination could be an useful alternative:in fact,gold extraction yield is quite low:57%in non-pre-treated material and 25%in pre-treatment material.

After synthesis of an asymmetric tetradentate ONN＇O＇ Schiff base ligand （H2L） followed by reaction of the synthesized H2L with an equimolar mixture of methanolic solutions of the VO（acac）2, a new oxidovanadium（IV） Schiff base complex （VOL） was synthesized. The Schiff base ligand and its complex were characterized by FT-IR and UV-vis spectra and C, H, N analysis, The crystal structure of VOL was also determined by single crystal X-ray analysis. The VOL complex crystallizes in monoclinic space group Cc. The Schiffbase ligand acts as a tetradentate ligand through its two＇iminic nitrogens and two phenolic and acetylacetonate oxygens. Thermogravimetric analysis of the VOL showed that it decomposes in two steps and converts to mixed vanadium oxides at 477℃. In addition, thermal decomposition of the VOL complex in air at 660 ℃ leads to formation of V2O5 nanoparticles with the average size estimated from XRD 49 nm. The catalytic activity of the VOL complex was investigated in the epoxidation reaction and different reaction parameters were optimized. The results showed that the cyclic alkenes were efficiently converted to the corresponding epoxides, whereas the VOL did not appreciably convert the linear alkenes.

The distributions of two ubiquitous fouling cheilostome bryozoans,Cryptosula pallasiana(Moll,1803)and W atersipora sp.,on a ship moored for almost six years in Qingdao Bay show diff erences with respect to illumination,C ryptosula being dominant on the side of the ship which was exposed to the sun and Watersipora dominating on the other side which was in shadow for most of the *** interactions for substrate space were nearly always won by Watersipora,which succeeded in overgrowing the edges of C ryptosula colonies regardless of the side of the *** for the superiority of W atersipora in spatial competition with Cryptosula could include faster growth rate and the stronger feeding currents created by the larger lophophores of Watersipora.

Dating of fracture-filling calcitewith U-Pb geochronology is becoming a rapidly adopted technique for determining the absolute timing of brittle deformation in the upper *** calcite is a desirable target,as it precipitates between individual fault slip displacement events,and provides additional kinematic *** we present a case study of slickenfibres formed on the Očkov thrust in the Lower Palaezoic Prague Basin,Bohemian Massif,utilising a combination of petrographic and in situ *** demonstrate that slickenfibre external textures can be preserved,whilst internally primary textures are removed by fluid infiltration and recrystallization,leading to variable U and Pb *** slickenfibre yielded a date of ca.250 Ma,which we interpret as recording fault slip along the Očkov *** cross-cutting slickenfibre yielded more scattered U-Pb data,with an imprecise apparent age around ca.95 *** slickenfibre is recrystallised,destroying the primary textures,and exhibits element *** meaning of this younger apparent age is therefore questionable;whereas it likely reflects Cretaceous U and Pb mobility assisted by fluid-flow along the fault plane,it may not reflect a period of fault *** results demonstrate that slickenfibre-based U-Pb dates do not unequivocally relate to fault motion,and that petrographic and elemental analyses are important requirements for interpreting calcite U-Pb data.

Detection of H pylori infection by ELISA and Western blot techniques and evaluation of anti CagA seropositivity in adult Turkish dyspeptic patients

AIM: To detect H pylori infection and to evaluate the anti CagA seropositivity in adult Turkish dyspeptic patients. METHODS: We evaluated anti-H pylori IgA, IgG and anti-CagA antibodies using commercial enzyme-linked immunoassay (ELISA) and Western blot in dyspeptic Turkish patients. H pylori status was determined by histology and rapid urease testing. RESULTS: Fifty-six patients were entered. Forty-eight (85.7%) out of the 56 patients were positive for H pylori. H pylori IgG seropositivity was 82.1%, IgA seropositivity 48.2%. CagA ELISA showed that IgG was positive in 50% and IgA in 30.4% of those with H pylori infections. Western blot showed that IgG seropositivity was 80.4% and IgA seropositivity 33.9%. Western blot detected IgG antibodies with reactivity to CagA in 50%, VacA in 62.5%, UreB in 87.5%, UreA in 80.4%, and OMP in 57.1%. None of the tests had a sensitivity and specifi city above 80%. CONCLUSION: None of these commercial tests seems clinically useful for H pylori detection in adult dyspeptic patients, while Western blot can give seropositivity and determine anti-CagA, VacA virulence factor status of Turkish dyspeptic patients in the Izmir region.

Autophagy is an intracellular degradation mechanism involved in antiviral defense,but the strategies employed by plant viruses to counteract autophagy-related defense remain unknown for the majority of the ***,we describe how the Chinese wheat mosaic virus(CWMV,genus Furovirus)interferes with autophagy and enhances its infection in Nicotiana *** two-hybrid screening and in vivo/in vitro assays revealed that the 19 k Da coat protein(CP19 K)of CWMV interacts with cytosolic glyceraldehyde-3-phosphate dehydrogenases(GAPCs),negative regulators of autophagy,which bind autophagy-related protein 3(ATG3),a key factor in ***19 K also directly interacts with ATG3,possibly leading to the formation of a CP19 K–GAPC–ATG3 ***19 K–GAPC interaction appeared to intensify CP19 K–ATG3 ***,CP19 K expression upregulated GAPC gene transcripts and reduced autophagic ***,the silencing of GAPC genes in transgenic *** reduced CWMV accumulation,whereas CP19 K overexpression enhanced ***,our results suggest that CWMV CP19 K interferes with autophagy through the promotion and utilization of the GAPC role as a negative regulator of autophagy.
