Wound ulceration caused by diabetes is a typical chronic wound wherein healing the local tissue is difficult due to lack of blood vessels and tissue necrosis caused by the long-term accumulation of free radicals.Near-field electrospinning(NFES)is an innovative technology used to produce micro-nano-scaled,controllable sequencing fibers.In this study,we constructed a novel wound dressing based on the NFES polycaprolactone(PCL)fiber network and modified gelatin with methacrylic anhydride(GelMA)hydrogel to promote angiogenesis and the re-epithelialization of diabetic wounds.An angiogenic and antioxidant drug named deferoxamine(DFO)was encapsulated in a GelMA hydrogel to achieve a slow-release effect that is more suitable for chronic wounds.The cell adhesion experiment showed that the cells could attach to the fibers in the dressing group having a network of PCL fibers on the surface and grow along the direction of the fibers,which in turn,effectively regulates cell behavior from the physical structure.Additionally,the large pore size(~500μm)of the network allowed the cells to penetrate the pores and enter the surface of the hydrogel without being blocked out.Besides,the composite dressing had a notable effect on angiogenesis.Furthermore,antioxidation experiments confirmed that the DFO-loaded hydrogel exhibited antioxidant activity.Experimental animal models of diabetes showed that rats treated with the PCL-GelMA-DFO(PGD)hydrogel had faster ability of hemostasis,scab formation,and wound healing.In conclusion,the PGD hydrogel effectively promoted the repair of chronic wounds.

Electrospinning is a popular and effective method of producing porous nanofibers with a large surface area,superior physical and chemical properties,and a controllable pore size.Owing to these properties,electrospun nanofibers can mimic the extracellular matrix and some human tissue structures,based on the fiber configuration.Consequently,the application of electrospun nanofibers as biomaterials,varying from two-dimensional(2D)wound dressings to three-dimensional(3D)tissue engineering scaffolds,has increased rapidly in recent years.Nanofibers can either be uniform fiber strands or coaxial drug carriers,and their overall structure varies from random mesh-like mats to aligned or gradient scaffolds.In addition,the pore size of the fibers can be adjusted or the fibers can be loaded with disparate medicines to provide different functions.This review discusses the various structures and applications of 2D fiber mats and 3D nanofibrous scaffolds made up of different one-dimensional(1D)fibers in tissue engineering.In particular,we focus on the improvements made in recent years,especially in the fields of wound healing,angiogenesis,and tissue regeneration.

OBJECTIVE:To test the hypothesis that moxibustion may inhibit rheumatoid arthritis(RA)synovial inflammation by regulating the expression of macrophage migration inhibitory factor(MIF)/glucocorticoids(GCs).METHODS:Fifty male Sprague-Dawley rats were randomly divided into five groups(n=10 each):blank Control(CON)group,RA Model(RA)group,Moxibustion(MOX)group,MIF inhibitor(S,R)-3-(4-hydroxyphenyl)-4,5-dihydro-5-isoxazole acetic acid methyl ester(ISO-1)group,and Moxibustion+MIF inhibitor ISO-1(MOX+ISO-1)group.Rats in the ISO-1 group and ISO-1+MOX group were intraperitoneally injected with the inhibitor ISO-1.The rats in the RA group,ISO-1 group,MOX group,and ISO-1+MOX group were injected with Freund's complete adjuvant(FCA)in the right hind footpad to establish an experimental RA rat model.In the MOX group and MOX+ISO-1 group,rats were treated with Moxa.The thickness of the footpads of the rats in each group was measured at three-time points before,after modeling and after moxibustion treatment.The contents of serum MIF,corticosterone(CORT),tumor necrosis factor-α(TNF-α)and interleukin-1β(IL-1β)were detected by enzyme-linked immunosorbent assay;and the contents of synovial MIF were detected by Western blot.Hematoxylin-eosin(HE)staining method was used to observe the pathological changes of synovial tissue under a section light microscope,and pathological scoring was performed according to the grading standard of the degree of synovial tissue disease.RESULTS:Moxibustion was found to reduce the level of MIF and alleviate inflammation in RA rats in this study.In addition,after inhibiting the expression of MIF,the level of CORT increased,and the level of TNF-α decreased.Treating RA rats with inhibited MIF by moxibustion,the level of CORT was almost unchanged,but the level of TNF-α further decreased.The correlation analysis data suggested that MIF was positively related to the expression of TNF-α and negatively correlated with the expression of CORT.CONCLUSION:Reducing MIF to increase CORT and decrease TNF-α by moxibustion

A Highly Pathogenic Strain of Porcine Deltacoronavirus Caused Watery Diarrhea in Newborn Piglets

Porcine deltacoronavirus(PDCoV) is a newly identified virus that causes watery diarrhea in newborn piglets and results in significant economic losses to the pig industry. Since first reported in Hong Kong in 2012, PDCoV has been subsequently detected in USA, South Korea, Thailand, and China's Mainland. Here we isolated a strain of PDCoV, named CHN-GD-2016,from the intestinal content of a diseased newborn piglet with severe diarrhea in a pig farm in Guangdong, China. PDCoV CHN-GD-2016 could be identified by immunofluorescence with PDCoV specific rabbit antisera, and typical crown-shaped particles with spiky surface projections of this PDCoV were observed with electron microscopy. Genomic analysis showed that the PDCoV CHN-GD-2016 was closely related to other Chinese PDCoV strains, with the highest sequence similarity with the strain CHN/Tianjin/2016. Importantly, inoculation of newborn piglets with 1×10~5 TCID_(50) of CHN-GD-2016 by oral feeding successfully reproduced clear clinical symptoms, including vomiting, dehydration, and severe diarrhea in piglets. In addition, the virus RNA in rectal swabs from 1 to 7 days post inoculation was detected, macroscopic and microscopic lesions in small intestine were observed, and viral antigen was also detected in the small intestines with immunohistochemical staining. Collectively, the data show in this study confirms that PDCoV is present in Guangdong,China and is highly pathogenic in newborn piglets.

文本研究人流后关爱(PAC)在人工流产术后计划生育关爱中的应用。选择2011年1月—2013年1月,武汉市蔡甸区计划生育服务站门诊进行人工流产的260名病例为分析对象,根据其先后序号按照奇偶顺序分为关爱PAC组和对照组,每组130例,对PAC组进行流产后关爱指导,而对照组只进行常规处理。一年后,共收回有效问卷248份,其中关爱组126份,对照组122份。PAC组避孕知识和人流后保健知识掌握优良率均高于对照组,生活质量评分有明显优势,而重复流产率明显较低,育龄妇女满意率较高。其差异均有统计学意义(P<0.05)。高价值的PAC关爱,对于减少重复流产的概率和提高生活质量有着积极重要的意义。

进出港口的大型船舶需向港口申请拖轮协助以进行靠离泊作业。拖轮调度是港口重要的计划事项之一。针对拖轮调度过程中需要平衡完工时间和油耗量以提高港口服务水平和降低拖轮公司经营成本的问题,本文以最小化拖轮最大完工时间和最小化拖轮总油耗量为目标,构建了混合整数规划拖轮多目标优化调度模型。模型还考虑了潮汐港口大量船舶在潮水期间集中进出港的特点,并根据拖轮在调度过程中的不同状态分类计量其产生的油耗量,以使模型更接近实际状况。为求解模型,运用了带有精英策略的非支配排序遗传算法(NSGA-II),算法采用一维实数编码,以事件建模思想设置适应度函数,并结合拖轮调度特点设计了遗传算子,求得的Pareto前沿解和算法对比验证了该算法的有效性。最后,以广州港港口拖轮调度实际运作数据作为算例,验证了模型的可行性与有效性,为港口拖轮调度计划提供了决策依据。

目的探究足部距骨骨折合并脱位的早期康复护理效果。方法在该院展开相关研究工作,研究的开展时间在2018年6月—2019年6月,该研究共选取患者80例,将其随机分为实验组和对照组,每组有患者40例,均为足部距骨骨折合并脱位患者,分别给予其常规护理配合康复护理和单纯的常规护理,对其护理效果进行观察分析。结果实验组患者的踝关节功能恢复优良率达到90.00%,而对照组患者的优良率则为72.00%,实验组患者的优良率较高,对两组数据差异进行比较,结果显示差异有统计学意义(P<0.05)。实验组患者和对照组患者的护理满意度分别为97.50%和82.50%,对其数据差异进行比较,差异有统计学意义(P<0.05)。结论针对足部距骨骨折合并脱位患者,给予其康复护理,能够实现对患者踝关节的改善,将患者护理满意度进行提高,值得临床加强重视程度,展开积极推广。

Chondrocyte differentiation is a critical process for endochondral ossification,which is responsible for long bone development and fracture repair.Considerable progress has been made in understanding the transcriptional control of chondrocyte differentiation;however,epigenetic regulation of chondrocyte differentiation remains to be further studied.NSD1 is a H3K36(histone H3 at lysine 36)methyltransferase.Here,we showed that mice with Nsd1 deficiency in Prx1+mesenchymal progenitors but not in Col2+chondrocytes showed impaired skeletal growth and fracture healing accompanied by decreased chondrogenic differentiation.Via combined RNA sequencing(RNA-seq)and chromatin immunoprecipitation sequencing(ChIP-seq)analysis,we identified sex determining region Y box 9(Sox9),the key transcription factor of chondrogenic differentiation,as a functional target gene of NSD1.Mechanistically,NSD1 regulates Sox9 expression by modulating H3K36me1 and H3K36me2 levels in the Sox9 promoter region,constituting a novel epigenetic regulatory mechanism of chondrogenesis.Moreover,we found that NSD1 can directly activate the expression of hypoxia-inducible factor 1α(HIF1α),which plays a vital role in chondrogenic differentiation through its regulation of Sox9 expression.Collectively,the results of our study reveal crucial roles of NSD1 in regulating chondrogenic differentiation,skeletal growth,and fracture repair and expand our understanding of the function of epigenetic regulation in chondrogenesis and skeletal biology.

目的探讨后外侧小切口微创半髋关节置换术治疗老年股骨颈骨折患者的临床疗效。方法选取我院2013年9月至2016年9月收治的老年股骨颈骨折患者76例,随机分为研究组和对照组,每组38例。研究组采用后外侧小切口微创半髋关节置换术,对照组采用常规后外侧入路半髋关节置换术。对比两组患者围手术期的各项指标。结果研究组的术中出血量、手术时间和下床活动时间均优于对照组(P<0.05)。治疗后,研究组的Harris评分和VAS评分均优于对照组(P<0.05)。结论后外侧小切口微创半髋关节置换术可有效改善老年股骨颈骨折患者的髋关节功能,且术中出血量少,安全性高,值得临床推广应用。

基于Petri网的拖车到达调度优化模型与仿真

为解决内河集装箱码头大量等待作业的拖车引起港口外围交通要道阻塞的问题,通过优化拖车到达调度,采用非平稳泊松过程,模拟进港拖车到达规律,建立了适合珠三角内河集装箱码头进港拖车到达作业流程的混合Petri网(hybrid Petri net,HPN)模型和优化调度方案.根据HPN模型对佛山市三水港建立了Arena仿真模型,使用过程分析器(process analyzer,PAN)确定拖车的最优到达批量并检验优化效果,仿真结果表明:拖车到达调度优化后,拖车等待时间可减少87%,等待数量减少了26%,调度优化有效地缓解了港口拖车排队导致的交通阻塞问题.
