Polyvinylpyrrolidone K-30(PVP) was introduced into the preparation of nanozero-valent iron(n ZVI) and the traditional liquid-phase reduction was improved. The introduction of PVP simplified the traditional *** n ZVI prepared with this new approach showed excellent surface characters and high performance on the removal of cadmium. TEM results showed that the aggregates of n ZVI can reach to several micrometers in length but less than 100 nm in diameter. The iron particles that were enclosed by a layer of oxide film that is less than10 nm, demonstrated that the n ZVI possesses a core–shell structure. BET results indicate that the specific surface area of the n ZVI was 20.3159 m^2g^(-1). A three factor and three level orthogonal experiment was employed to find out the dominant factor that affects the removal rate of cadmium by n ZVI. Based on the range values, the prominence order of each factor was: initial p H of the solution N initial concentration of cadmium N dosage of n ZVI, the range was 96.453, 3.294 and 1.747, respectively. A simulation was performed under the same condition and a same conclusion was derived, this consistence confirmed the validity of the conclusion that p H is the most significant factor that affects the adsorption efficiency.

Ultrasensitive protein identification is of paramount importance in basic research and clinical diagnostics but remains extremely challenging. A key bottleneck in preventing single-molecule protein sequencing is that, unlike the revolutionary nucleic acid sequencing methods that rely on the polymerase chain reaction(PCR) to amplify DNA and RNA molecules, protein molecules cannot be directly amplified. Decoding the proteins via amplification of certain fingerprints rather than the intact protein sequence thus represents an appealing alternative choice to address this formidable challenge. Herein, we report a proof-of-concept method that relies on residue-resolved DNA barcoding and composition code counting for amplifiable protein fingerprinting(AmproCode). In AmproCode, selective types of residues on peptides or proteins are chemically labeled with a DNA barcode, which can be amplified and quantified via quantitative PCR. The operation generates a relative ratio as the residue-resolved ‘composition code’ for each target protein that can be utilized as the fingerprint to determine its identity from the proteome database. We developed a database searching algorithm and applied it to assess the coverage of the whole proteome and secretome via computational simulations, proving the theoretical feasibility of AmproCode. We then designed the residue-specific DNA barcoding and amplification workflow, and identified different synthetic model peptides found in the secretome at as low as the fmol/L level for demonstration. These results build the foundation for an unprecedented amplifiable protein fingerprinting method. We believe that, in the future,AmproCode could ultimately realize single-molecule amplifiable identification of trace complex samples without further purification, and it may open a new avenue in the development of next-generation protein sequencing techniques.

Based on hourly precipitation from national surface stations,persistent heavy rainfall events(PHREs)over the Sichuan Basin(SCB)are explored during the warm season(May to September)from 2000 to 2015 to compare synoptic circulations and maintenance mechanisms between different PHRE *** are two main types of PHREs:one is characterized by a rain belt west of 106°E over the SCB(WSB-PHREs),and the other features a rain belt east of 106°E over the SCB(ESB-PHREs).In total,there are 18 ESB-PHREs and 10 WSB-PHREs during the study ***,the rain belts of WSB-PHREs are along the terrain distribution east of the Tibetan Plateau,while the precipitation intensity of ESB-PHREs is *** the two types of PHREs,the shortwave trough over the SCB and the western Pacific subtropical high act as their favorable background environments,particularly for *** water vapor of WSB-PHREs is mainly transported from the South China Sea,whereas for ESB-PHREs the South China Sea and Bay of Bengal are their main moisture *** composite vorticity budgets of southwest vortices during their mature stage indicate that the convergence effect is a dominant factor for maintaining the two types of PHREs,and the strong vertical vorticity advection is also favorable,but the relative contribution of vertical advection is larger for WSB-PHREs.

Leaf area index （LAI） is a key parameter for describing vegetation structures and is closely associated with vegetative photosynthesis and energy balance. The accurate retrieval of LAI is important when modeling biophysical processes of vegetation and the productivity of earth systems. The Random Forests （RF） method aggregates an ensemble of deci- sion trees to improve the prediction accuracy and demonstrates a more robust capacity than other regression methods. This study evaluated the RF method for predicting grassland LAI using ground measurements and remote sensing data. Parameter optimization and variable reduction were conducted before model prediction. Two variable reduction methods were examined： the Variable Importance Value method and the principal component analysis （PCA） method. Finally, the sensitivity of RF to highly correlated variables was tested. The results showed that the RF parameters have a small effect on the performance of RF, and a satisfactory prediction was acquired with a root mean square error （RMSE） of 0.1956. The two variable reduction methods for RF prediction produced different results; variable reduction based on the Variable Importance Value method achieved nearly the same prediction accuracy with no reduced prediction, whereas variable re- duction using the PCA method had an obviously degraded result that may have been caused by the loss of subtle variations and the fusion of noise information. After removing highly correlated variables, the relative variable importance remained steady, and the use of variables selected based on the best-performing vegetation indices performed better than the vari- ables with all vegetation indices or those selected based on the most important one. The results in this study demonstrate the practical and powerful ability of the RF method in predicting grassland LAI, which can also be applied to the estimation of other vegetation traits as an alternative to conventional empirical regression models an

BACKGROUND An in-depth study of the pathogenesis and biological characteristics of ampullary carcinoma is necessary to identify appropriate treatment strategies. To date, only eight ampullary cancer cell lines have been reported, and a mixed-type ampullary carcinoma cell line has not yet been *** To establish a stable mixed-type ampullary carcinoma cell line originating from *** Fresh ampullary cancer tissue samples were used for primary culture and subculture. The cell line was evaluated by cell proliferation assays, clonal formation assays, karyotype analysis, short tandem repeat(STR) analysis and transmission electron microscopy. Drug resistances against oxaliplatin, paclitaxel, gemcitabine and 5-FU were evaluated by cell counting kit-8 assay. Subcutaneous injection 1 × 106 cells to three BALB/c nude mice for xenograft studies. The hematoxylin-eosin staining was used to detect the pathological status of the cell line. The expression of biomarkers cytokeratin 7(CK7), cytokeratin 20(CK20), cytokeratin low molecular weight(CKL), Ki67 and carcinoembryonic antigen(CEA) were determined by immunocytochemistry *** DPC-X1 was continuously cultivated for over a year and stably passaged for more than 80 generations;its population doubling time was 48 h. STR analysis demonstrated that the characteristics of DPC-X1 were highly consistent with those of the patient’s primary tumor. Furthermore, karyotype analysis revealed its abnormal sub-tetraploid karyotype. DPC-X1 could efficiently form organoids in suspension culture. Under the transmission electron microscope, microvilli and pseudopods were observed on the cell surface, and desmosomes were visible between the cells. DPC-X1 cells inoculated into BALB/C nude mice quickly formed transplanted tumors, with a tumor formation rate of 100%. Their pathological characteristics were similar to those of the primary tumor. Prediction of the globally ecological suitability of Panax quinquefolius by the geographic information system for global medicinal plants(GMPGIS)

American ginseng(Panax quinquefolius L.) is a well-known Asian traditional herbal medicine with a large market demand. The plant is native to eastern North America, and its main producing areas worldwide are decreasing due to continuous cropping obstacles and environmental changes. Therefore, the identification of maximum similarities of new ecological distribution of ***, and prediction of its response to climate change in the future are necessary for plant introduction and cultivation. In this study, the areas with potential ecological suitability for P. quinquefolius were predicted using the geographic information system for global medicinal plants(GMPGIS) based on 476 occurrence points and 19 bioclimatic variables. The results indicate that the new ecologically suitable areas for P. quinquefolius are East Asia and the mid-eastern Europe, which are mainly distributed in China, Russia, Japan, Ukraine, Belarus, North Korean, South Korea, and Romania. Under global climate change scenarios, the suitable planting areas for P. quinquefolius would be increased by 9.16%–30.97%, and expanding north and west over the current ecologically suitable areas by 2070. The potential increased areas that are ecologically suitable include northern Canada, Eastern Europe, and the Lesser Khingan Mountains of China, and reduced regions are mainly in central China, the southern U.S., and southern Europe. Jackknife tests indicate that the precipitation of the warmest quarter was the important climatic factor controlling the distribution of P. *** findings can be used as a useful guide for P. quinquefolius introduction and cultivation in ecologically suitable areas.
