Dysregulation of RNA modification systems in clinical populations with neurocognitive disorders

The study of modified RNA known as epitranscriptomics has become increasingly relevant in our understanding of disease-modifying mechanisms. methylation of N6 adenosine(m6A) and C5 cytosine(m5C) bases occur on mRNAs, tRNA, mt-tRNA, and rRNA species as well as non-coding RNAs. With emerging knowledge of RNA binding proteins that act as writer, reader, and eraser effector proteins, comes a new understanding of physiological processes controlled by these systems. Such processes when spatiotemporally disrupted within cellular nanodomains in highly specialized tissues such as the brain, give rise to different forms of disease. In this review, we discuss accumulating evidence that changes in the m6A and m5C methylation systems contribute to neurocognitive disorders. Early studies first identified mutations within FmR1 to cause intellectual disability Fragile X syndromes several years before FmR1 was identified as an m6A RNA reader protein. Subsequently, familial mutations within the m6A writer gene mETTL5, m5C writer genes NSUN2, NSUN3, NSUN5, and NSUN6, as well as THOC2 and THOC6 that form a protein complex with the m5C reader protein ALYREF, were recognized to cause intellectual development disorders. Similarly, differences in expression of the m5C writer and reader effector proteins, NSUN6, NSUN7, and ALYREF in brain tissue are indicated in individuals with Alzheimer’s disease, individuals with a high neuropathological load or have suffered traumatic brain injury. Likewise, an abundance of m6A reader and anti-reader proteins are reported to change across brain regions in Lewy bodies diseases, Alzheimer’s disease, and individuals with high cognitive reserve. m6A-modified RNAs are also reported significantly more abundant in dementia with Lewy bodies brain tissue but significantly reduced in Parkinson’s disease tissue, whilst modified RNAs are misplaced within diseased cells, particularly where synapses are located. Background Non-interventional large-scale research on real-world patients who had a stroke requires the use of multiple data sources ensuring access to longitudinal data from large populations with clinically-detailed *** sought to establish a framework for longitudinal research on patients hospitalised with stroke by linking information-rich,deidentified inpatient data from the Paul Coverdell National Acute Stroke Program(PCNASP)to commercial and medicare Advantage longitudinal claims *** All stroke admissions in PCNASP between 2008 and 2015 were evaluated for linkage to longitudinal claims from a commercial insurer using an algorithm based on six available common data fields(patient age,gender,admission date,discharge date,discharge diagnosis and state)and a hospital *** evaluated the linkage quality(via the percentage of unique records in the linked dataset)and the representativeness of the linked *** also described medical history,stroke severity and patterns of medication use among the PCNASP-claims linked *** The linkage produced uniqueness equal to 99.1%.We identified 5644 linked and 98896 unlinked patients who had an ischaemic stroke hospitalisation in claims *** patients were younger than unlinked(69.7 vs 72.5 years),but otherwise similar by medical history,prestroke medication use or lab *** severity was mild and most patients were discharged *** and discharge use of antihypertensive and statins in the PCNASP were greater than their use as measured by filled prescriptions in *** High-quality linkage between the PCNASP and commercial claims data is *** linkage identified differences between reported or recommended versus actual out-of-hospital medication utilisation,highlighting the importance of longitudinal data availability for research aimed to improve the care of patients who had a stroke.

Short-term effect of physical activity on sleep health:A population-based study using accelerometry

Background:Regular physical activity(PA) is an important behavior in improving sleep ***,the short-term effects of PA on sleep are still *** study aimed to verify the effect of different intensities of PA practiced in different periods of the day on the subsequent sleep night in a population-based cohort of young ***::Prospective analyses were conducted for PA performed during the day and its effect on the following sleep night using data from the22-year follow-up of the 1993 Pelotas Birth Cohort in Brazil(mean age of participants-22.6 years).Wrist-worn accelerometry was usedto measure both PA and sleep *** intensity,we analyzed the sleep effect of light PA(LPA),moderate PA,and vigorous PA,stratified by *** variables were sleep time window(STW;the difference between sleep onset and sleep end),total sleep time(TST;the sum of minutes classified as sleep in STW),and sleep percent(SP;SP=(TST/STW);expressed in percentage).We performed generalized estimating equations using Stata ***::The means of STW,TST,and SP were 443.6 min/day,371.1 min/day,and 84%,*** spent in moderate PA and vigorous PA in the morning and afternoon was not associated with sleep *** men,10 min/day of morning LPA increased TST by2.56 min/*** women,10 min/day of morning LPA increased SP by 0.15 percentage *** LPA also increased SP by 0.09 percentage points for *** PA seems to have an inverse effect on sleep variables for any intensity and both ***::The effect of PA on sleep health is intrinsically related to the period of the day in which it is *** effect magnitude is different between *** better sleep health,it is preferable that PA be performed during the day.

Internet of Things (IoT) is transforming the technical setting ofconventional systems and finds applicability in smart cities, smart healthcare, smart industry, etc. In addition, the application areas relating to theIoT enabled models are resource-limited and necessitate crisp responses, lowlatencies, and high bandwidth, which are beyond their abilities. Cloud computing (CC) is treated as a resource-rich solution to the above mentionedchallenges. But the intrinsic high latency of CC makes it nonviable. The longerlatency degrades the outcome of IoT based smart systems. CC is an emergentdispersed, inexpensive computing pattern with massive assembly of heterogeneous autonomous systems. The effective use of task scheduling minimizes theenergy utilization of the cloud infrastructure and rises the income of serviceproviders by the minimization of the processing time of the user job. Withthis motivation, this paper presents an intelligent Chaotic Artificial ImmuneOptimization Algorithm for Task Scheduling (CAIOA-RS) in IoT enabledcloud environment. The proposed CAIOA-RS algorithm solves the issue ofresource allocation in the IoT enabled cloud environment. It also satisfiesthe makespan by carrying out the optimum task scheduling process with thedistinct strategies of incoming tasks. The design of CAIOA-RS techniqueincorporates the concept of chaotic maps into the conventional AIOA toenhance its performance. A series of experiments were carried out on theCloudSim platform. The simulation results demonstrate that the CAIOA-RStechnique indicates that the proposed model outperforms the original version,as well as other heuristics and metaheuristics.

Chick chorioallantoic membrane model to investigate role of migrasome in angiogenensis

The development of the vascular system is essential for embryonic development, including processes such as angiogenesis. Angiogenesis plays a critical role in many normal physiological and pathological processes. It is driven by a set of angiogenic proteins, including angiogenic growth factors, chemokines,and extracellular matrix proteins. Among various animal model systems, the chorioallantoic membrane(CAm), a specialized and highly vascularized tissue of the avian embryo, has proven to be a valuable tool for analyzing the angiogenic potential of candidate cells or factors. In this protocol, we provide detailed procedures for establishing the CAm model to evaluate the function and mechanism of migrasomes in embryonic angiogenesis. This includes the CAm nylon mesh assay and CAm ex vivo sprouting assay to assess CAm angiogenesis, as well as the observation, purification, and delivery of migrasomes. Additionally, we describe the generation of T4-KO-m Cherry-KI embryos using the CRISPR system within the CAm tissue to investigate the role of migrasomes in angiogenesis.

Extraction of protein from churpi of yak milk origin:Size reduction,nutraceutical potential and as a wall material for resveratrol

Resveratrol is a known anti-oxidant and anti-cancer *** low bioavailability and poor water solubility of resveratrol is a major barrier that restricts its usage in food *** overcome these problems,the study presents micro and nano encapsulation of resveratrol in casein micelles extracted from Himalayan cheese(Churpi).Resveratrol loaded casein microparticles(CS-rm)were obtained using emulsion-freeze ***,resveratrol loaded casein nanoparticles(CS-rn)were obtained using *** were characterized using dynamic light scattering(DLS),scanning electron microscopy(SEm)and attenuated total reflectance-fourier transform infrared spectroscopy(ATR-FTIR).DLS revealed hydrodynamic diameter of 71.24μm and 387.023 nm and zeta potential of-22.62 and-28.08 mV for CS-rm and *** confirmed the characteristic peaks of resveratrol at 965.4,1380.86,1586.66 and 1607 cm-1 confirmed successful micro and ***-rm displayed high encapsulation efficiency(52.32%)and swelling power(77.90 at pH 3 and 68.65 at pH 7.5)than *** micro and nanoencapsulation protected resveratrol in gastric conditions,however,CS-rn showed high release of resveratrol then CS-rm in simulated intestinal *** anti-oxidant and anti-cancer activity of resveratrol under simulated gastric and intestinal conditions(SGID)was significantly higher(p≤0.05)for CS-rn,showing that nanoencapsulation improved release and nutraceutical profile of *** loaded nano and micro casein particles can be used a source of fortified protein supplement in growing food and nutraceutical industry.
