SARS-CoV-2 main protease(M^(pro))is one of the most extensively exploited drug targets for *** disparate compounds have been reported as M^(pro) inhibitors,raising the question of their target *** elucidate the target specificity and the cellular target engagement of the claimed M^(pro) inhibitors,we systematically characterize their mechanism of action using the cell-free FRET assay,the thermal shift-binding assay,the cell lysate Protease-Glo luciferase assay,and the cell-based FlipGFP ***,our results have shown that majority of the M^(pro) inhibitors identified from drug repurposing including ebselen,carmofur,disulfiram,and shikonin are promiscuous cysteine inhibitors that are not specific to M^(pro),while chloroquine,oxytetracycline,montelukast,candesartan,and dipyridamole do not inhibit M^(pro) in any of the assays ***,our study highlights the need of stringent hit validation at the early stage of drug discovery.

Many-body interactions in condensed matter could lead to emergent phenomena spanning superconductivity,ferromagnetism,exciton condensation,*** emergence of these phenomena often requires highly ordered spatial arrangements of the interacting species to enforce specific space symmetries and interacting ***−organic frameworks(MOFs),crystalline materials formed by self-assembly of metal ions and organic ligands,allow for precise design of their crystal structures and sophisticated tuning of Coulombic interaction or magnetic coupling among lattice *** atomic-level designability combined with high crystallinity and versatile types of lattices(e.g.,kagome and honeycomb lattices)render MOFs as a great platform to investigate emergent *** this Emerging Topic,we summarize recent studies evidencing emergent phenomena in MOFs including strong correlations,superconductivity,charge density wave,long-range magnetic order,and quantum spin *** highlight the great potential of MOFs as quantum materials and discuss challenges including growth of high-quality single crystals and in-depth physical characterizations to reveal insights into the nature of physical properties of MOFs.

Using biodegradable material derived from renewable resources as petroleum-based plastics replacement is a promising way towards sustainable ***,the insufficient mechanical properties and complex manufacturing process of bioplastics still need to be improved for high-quality food ***,we report a top-down strategy to transform natural wood into a clear wood packaging film through scalable delignification and polyvinyl alcohol(PVA)*** wood packaging film demonstrates a laminated structure with completely collapsed cell walls and PVA intertwined together after energy-saving air drying,resulting in high light transmittance with low haze,good mechanical performance,and high barrier performance for oxygen and water *** dynamics simulations reveal the underlying fracture mechanism between cellulose and PVA,which effectively enhances the Young’s modulus and strength of the wood packaging *** findings contribute to the development of biodegradable and strong packaging materials,as well as other food-related applications,using sustainable wood.

Bird flocking is a paradigmatic case of self‐organised collective behaviours in *** camera systems are employed to observe flocks of starlings,jackdaws,and chimney swifts,mainly on a spot‐fixed basis.A portable non‐fixed stereo vision‐based flocking observation system,namely FlockSeer,is developed by the authors for observing more species of bird flocks within field *** portable flocking observer,FlockSeer,responds to the challenges in extrinsic calibration,camera synchronisation and field movability compared to existing spot‐fixed observing systems.A measurement and sensor fusion approach is utilised for rapid calibration,and a light‐based synchronisation approach is used to simplify hardware *** has been implemented and tested across six cities in three provinces and has accomplished diverse flock‐tracking tasks,accumulating behavioural data of four species,including egrets,with up to 300 resolvable *** authors reconstructed the trajectories of a flock of egrets under disturbed conditions to verify the practicality and *** addition,we analysed the accuracy of identifying nearest neighbours,and then examined the similarity between the trajectories and the Couzin *** results demonstrate that the developed flocking observing system is highly portable,more convenient and swift to deploy in wetland‐like or coast‐like *** observation process is reliable and practical and can effectively support the study of understanding and modelling of bird flocking behaviours.

In the Kaiping Coal field,mining of five coal seams,located within 80 m in the Kailuan Group,#5,#7,#8,#9 and#12 coal seam,is difficult due to small interburden thickness,concentrated stress distributions,high coal seam metamorphism,and complex geological *** using the ZTR12 geological penetration radar(GPR)survey combined with borehole observations,the overburden caving due to mining of the five coals seams was *** development characteristics of full-cover rock fractures after mining were obtained from the GPR scan,which provides a measurement basis for the control of rock strata in close multiple coal seam *** the first time,it was found that the overburden caving pattern shows a periodic triangular caved ***,it is proposed that an upright triangular collapsed pile masonry and an inverted triangular with larger fragments piled up alternately appear in the lower *** research results show that the roof structure formed in the gob area can support the key overlying strata,which is beneficial to ensure the integrity and stability of the upper coal seams in multiple-seam mining of close coal seams.

Carboxylate-based metal–organic frameworks(CMOFs)have received considerable attentions for their high stability,catalytic activity,and ***,synthesis of CMOFs requires high temperature,pressure,and long reaction ***,we explored the activity of 2-methylimidazole(2-MIM)for ultrafast synthesis of CMOF nanostructures(CMOFNs),in aqueous medium at room temperature and reaction time of 10 *** CMOFNs have been synthesized by using Al3+,Cr3+,Cu2+,Fe3+,In3+,or Cd2+salt and1,4-bezenedicarboxylic acid,or 1,3,5-benzenetricarboxylic *** this technique,the CMOFs with space time yield 181–501 kgcrystal sizes of ca.200–700 nm was obtained.

Prospective observational study on the prognosis of ureteral lesions caused by impacted stones via dual-energy spectral computed tomography

Objective Ureteral lesions caused by impacted ureteral stones are likely to result in postoperative ureteral *** this basis,the study aimed to investigate if dual-energy spectral computed tomography can predict ureteral hardening caused by impacted stones and to explore the relationship between different types of ureteral lesions and the risk of ureteral *** This prospective study collected data of 93 patients with impacted stones from hospital automation system during January 2018 to October *** underwent an abdominal scan on a dual-energy spectral computed *** surgery,the operator used ureteroscopy to identify ureteral lesions,which were classified into four categories:edema,polyps,pallor,and *** months later,90 patients were reviewed for the degree of *** Endoscopic observations revealed 38(41%)cases of ureteral edema,20(22%)cases of polyps,13(14%)cases of pallor,and 22(24%)cases of *** were significant differences in hydronephrosis,the period of impaction,the calcium concentration of the ureter,and the slope of the spectral Hounsfield unit curve between the four *** that,we evaluated the factors associated with ureteral hardening and found that the calcium concentration of the ureter and hydronephrosis remained independent predictors of ureteral *** operating characteristic curve analysis showed that 5.3 mg/cm^(3)calcium concentration of the ureter is an optimal cut-off value to predict ureteral *** result of follow-up showed that 80 patients had complete remission of hydronephrosis,with a complete remission rate of 61.9%(13/21)in the hardening group and 97.1%(67/69)in the non-hardening group(p<0.001).Conclusion Calcium concentration of the ureter is an independent predictor of ureteral *** with ureteral hardening have more severe hydronephrosis after ureteroscopic *** the calcium concentration of the ureter is less

Osteosarcoma is usually resistant to immunotherapy and,thus primarily relies on surgical resection and high-dosage ***,less invasive or toxic therapies such as photothermal therapy(PTT)and chemodynamic therapy(CDT)generally failed to show satisfactory *** multimodal therapies with proper safety profiles may provide better solutions for ***,a simple nanocomposite that synergistically combines CDT,PTT,and chemotherapy for osteosarcoma treatment was *** this composite,small 2D Ni Fe-LDH flakes were processed into 3D hollow nanospheres via template methods to encapsulate 5-Fluorouracil(5-FU)with high loading *** nanospheres were then adsorbed onto larger 2D Ti3C2MXene monolayers and finally shielded by bovine serum albumin(BSA)to form 5-FU@Ni Fe-LDH/Ti3C2/BSA nanoplatforms(5NiTiB).Both in vitro and in vivo data demonstrated that the 5-FU induced chemotherapy,Ni Fe-LDH driven chemodynamic effects,and MXene-based photothermal killing collectively exhibited a synergistic“all-in-one”anti-tumor effect.5NiTiB improved tumor suppression rate from<5%by 5-FU alone to~80.1%.This nanotherapeutic platform achieved higher therapeutic efficacy with a lower agent dose,thereby minimizing side ***,the composite is simple to produce,enabling the fine-tuning of dosages to suit different ***,the platform is versatile and efficient,with potential for further development.
