Genome organization and phylogenetic tree analysis of Garlic virus E,a new member of genus Allexivirus

The complete sequence of an Allexivirus iso-lated from garlic plants in Yuhang City, Zhejiang Province, china had been determined. The single-strand, positive RNA genome was 8451 nucleotides in length excluding poly(A) tail. The genome organization of this virus was similar to that of the other Allexiviruses but only with 62.8%-64.8% nucleo-tide acid identities. The amino acid sequences of proteins encoded by ORFl-6 shared 67.6%-78.5%, 55.4%-66.2%, 56.7%-66.4%,40.3%-55.6%,66.3%-79.7%and 52.2%-68.8% identities with those of the others respectively. The homology range between it and the other Allexiviruses was similar to that between the other distinct species in this genus. A more comprehensive comparison using all available CP amino acid sequences showed that it shared only 63.9%-79.8% amino acids identical with the others. Therefore, it had been considered as a new member of the genus, named as garlic virus E (GarV-E). Phylogenetic analysis confirmed GarV-E as a distinct member and the correct names

Use of Controlled Release Fertilizer for Increasing N Efficiency of Direct Seeding Rice

Field trials on a silt-loamy paddy soil derived from shallow-sea deposit in direct seeding rice fields were conducted in Zhejiang, china, in 1996 to compare N efficiency of controlled release fertilizers (LP fertilizers) with the conventional urea. Six treatments including CK (no N fertilizer), conventional urea and different types of LP fertilizers at different rates were designed for two succeeding crops of early and late rice. A blend of different types of LP fertilizers as a single preplant "co-situs" application released N in a rate and amount synchronizing with uptake pattern of direct seeding rice. A single preplant application of the LP fertilizers could meet the N requirement of rice for the whole growth period without need of topdressing. Using LP fertilizer blends, equivalent grain yields could be maintained even if the N fertilization rates were reduced by 25%～50% compared with the conventional urea. Agronomic efficiency of the LP fertilizers was 13.6%～ 86.4% higher than that of the conventional urea in early rice and 100%～164.1% in late rice, depending on the amounts of the LP fertilizers applied. N fertilizer recovery rate increased from 27.4% for the conventional application of urea to 41.7%～54.l% for the single preplant "co-situs" application of the LP fertilizers. Use of the LP fertilizers was promising if the increase in production costs due to the high LP fertilizer prices could be compensated by increase in yield and N efficiency, reduction in labor costs and improvement in environment.

Polymyxa graminis is a eukaryotic obligate biotrophic parasite of plant roots that belongs to a poorly studied discrete taxonomic unit informally called “plasmodiophorids”. P. graminis is nonpathogenic, but has the ability to acquire and transmit nine plant viruses which belong to genera Bymovirus and Furovirus and cause serious diseases in cereal crop species and also result in significant yield reductions in china and elsewhere. Genus Bymovirus contains barley yellow mosaic virus (BaYMV), barley mild mosaic virus (BaMMV), wheat yellow mosaic virus (WYMV), wheat spindle streak mosaic virus (WSSMV), and oat mosaic virus (OMV), and genus Furovirus contains soil-borne wheat mosaic virus (SBWMV), oat golden stripe virus (OGSV), and newly identified Chinese wheat mosaic virus (CWMV) and soil-borne cereal mosaic virus (SBCMV). All these viruses have been sequenced and their worldwide distributions have been studied. The viruses are protected by the environment within P. graminis resting spores that may remain dormant but viable for decades (probably until a suitable host plant is encountered). Spontaneous deletion mutants of SBWMV, OGSV and OMV are detected, and these deletion mutants are not transmissible by the fungus. The persistent, soil-borne nature of these diseases makes the use of virus-resistant crop varieties currently the only practical and environmentally friendly means to control them, and a large number of disease resistant germplasms have been screened.

The transformation rate of three different strains of silkworm Bombyx mori was comparedafter the introduction of enhanced green fluorescence protein （EGFP）-encoding genes into the silkwormeggs by microinjection of a mixture of piggyBac vector and helper plasmid containing a *** there were no significant differences among the three strains in the percentages offertile moths in microinjected eggs （P=0.1258）,the percentages of G0transformed moths in fertile mothsand injected eggs were both significantly different （P=0.01368 and P=0.02398, respectively）.Thetransformation rate of the Nistari strain （Indian strain） was significantly higher than that of the other twostrains,Golden-yellow-cocoon （Vietnamese strain） and Jiaqiu （Chinese strain）,which had similar rate. Theseresults indicate that the transformation efficiency of the piggyBac-based system might vary with silkwormstrains with different genetic *** presence of endogenous piggyBac-like elements might bean important factor influencing the transformation efficiency of introduced piggyBac-derived vectors,andthe diverse amount and activation in different silkworm strains might account for the significant differences.

N availability is one of the most important factors limiting crop yield *** recovery of applications of 15 N-labeled fertilizer and crop residues in a rice-wheat cropping system was determined for up to 6 consecutive growing *** crop residues from the previous season were either incorporated or removed as two different *** results showed that 16.55%-17.79% (17.17% on average) of the fertilizer N was recovered in the crop during the first growing season,suggesting that more than 80% of crop N was not directly from the N *** 15 N-labeled residues were applied,12.01% was recovered in the crop in the first growing *** average recoveries of fertilizer N and crop residue N in the soil after the first growing season were 33.46% and 85.64%,respectively.N from soil organic matter contributed approximately 83% of the N in the crop when 15 N fertilizer was applied or 88% when crop residues were *** was a larger difference in the total 15 N recovery in plant and soil between N applications in the forms of fertilizer and crop *** of crop residues following the 15 N fertilizer application did not significantly promote 15 N recovery in the crop or *** average,only additional 1.94% of N for the fertilizer-applied field or 5.97% of N for the crop residue-applied field was recovered by the crops during the 2nd and 3rd growing *** total recoveries of 15 N in crop and soil were approximately 64.38% for the fertilizer-applied field after 6 growing seasons and 79.11% for the crop residue-applied field after 5 growing *** fertilizer N appeared to be more readily available to crops than crop residue N,crop residue N replenished soil N pool,especially N from soil organic matter,much more than fertilizer ***,crop residue N was a better source for sustaining soil organic *** results suggested that the long-term effect of fertilizer or crop residues on N recovery were differen

In this study, two DNA fragments encoding amino acid (141–160)-(21–40)-(141–160) of the VP1 of FMDV (foot-and-mouth disease virus) serotype O and (138–160)-(21–40)-(138–160) of the serotype A FMDV were chemically synthesized. These two tandem-repeat fragments were ligated and transfected into prokaryotic expression vector pTrcHis A to construct pTH-O-A. The other vector called pTH-O-scIgG-A was constructed similarly only that the two tandem-repeat DNA fragments were linked by the bovine- IgG heavy chain coding sequence. Guinea pigs immunized with the two bivalent vaccines pTH-O-A and pTH-O-scIgG-A showed both specific antibody activity and T cell proliferation responses. FMDV challenge tests showed that 85% and 70% of guinea pigs vaccinated twice with 200 μg of the fusion protein of pTH-O-A were protected from FMDV serotype O and serotype A infection respectively. 70% and 57% of the guinea pigs immunized with the fusion protein of pTH-O-scIgG-A were protected from FMDV serotype O and serotype A infection respectively.
