Telomeric DnA breaks in human induced pluripotent stem cells trigger ATR-mediated arrest and telomerase-independent telomere damage repair

Although mechanisms of telomere protection are well-def.ned in differentiated cells,how stem cells sense and respond to telomere dysf.nction,in particular telomeric double-strand breaks(DSBs),is poorly ***,we report the DnA damage signaling,cell cycle,and transcriptome changes in human induced pluripotent stem cells(iPSCs)in response to telomere-internal *** engineer human iPSCs with an inducible TRf1-fokI fusion protein to acutely induce DSBs at *** this model,we demonstrate that TRf1-fokI DSBs activate an ATR-dependent DnA damage response,which leads to p53-independent cell cycle arrest in *** CRISPR–Cas9 to cripple the catalytic domain of telomerase reverse transcriptase,we show that telomerase is largely dispensable for survival and lengthening of TRf1-fokI-cleaved telomeres,which instead are effectively repaired by robust homologous recombination(HR).In contrast to HR-based telomere maintenance in mouse embryonic stem cells,where HR causes ZSCAn4-dependent extension of telomeres beyond their initial lengths,HR-based repair of telomeric breaks is sufficient to maintain iPSC telomeres at a normal length,which is compatible with sustained survival of the cells over several days of TRf1-fokI *** f.ndings suggest a previously unappreciated role for HR in telomere maintenance in telomerase-positive iPSCs and reveal distinct iPSC-specific responses to targeted telomeric DnA damage.

The hydrothermal synthesis of In_(2)O_(3)and CeO_(2)–In_(2)O_(3)is investigated as well as the properties of sensor layers based on these *** the synthesis of In_(2)O_(3),intermediate products In(OH)_(3)and InOOH are formed,which are the precursors of stable cubic(c-In_(2)O_(3))and metastable rhombohedral(rh-In_(2)O_(3))phases,respectively.A transition from c-In_(2)O_(3)to rh-In_(2)O_(3)is observed with the addition of CeO_(2).The introduction of cerium into rh-In_(2)O_(3)results in a decrease in the sensor response to hydrogen,while it increases in composites based on c-In_(2)O_(3).The data on the sensor activity of the composites correlate with XPS results in which CeO_(2)causes a decrease in the concentrations of chemisorbed oxygen and oxygen vacancies in rh-In_(2)O_(3).The reverse situation is observed in composites based on c-In_(2)O_(3).Compared to In_(2)O_(3)and CeO_(2)–In_(2)O_(3)obtained by other methods,the synthesized composites demonstrate maximum response to H_(2)at low temperatures by 70–100℃,and have short response time(0.2–0.5 s),short recovery time(6–7 s),and long-term stability.A model is proposed for the dependence of sensitivity on the direction of electron transfer between In_(2)O_(3)and CeO_(2).

Memory deficit,which is often associated with aging and many psychiatric,neurological,and neurodegenerative diseases,has been a challenging issue for *** till now,all potential drug candidates have failed to produce satisfa ctory ***,in the search for a solution,we found that a treatment with the gene corresponding to the RGS14414protein in visual area V2,a brain area connected with brain circuits of the ventral stream and the medial temporal lobe,which is crucial for object recognition memory(ORM),can induce enhancement of *** this study,we demonstrated that the same treatment with RGS14414in visual area V2,which is relatively unaffected in neurodegenerative diseases such as Alzheimer s disease,produced longlasting enhancement of ORM in young animals and prevent ORM deficits in rodent models of aging and Alzheimer’s ***,we found that the prevention of memory deficits was mediated through the upregulation of.neuronal arbo rization and spine density,as well as an increase in brain-derived neurotrophic factor(BDnf).A knockdown of BDnf gene in RGS14414-treated aging rats and Alzheimer s disease model mice caused complete loss in the upregulation of.neuronal structural plasticity and in the prevention of ORM *** f.ndings suggest that BDnf-mediated neuronal structural plasticity in area V2 is crucial in the prevention of memory deficits in RGS14414-treated rodent models of aging and Alzheimer’s ***,our f.ndings of RGS14414gene-mediated activation of.neuronal circuits in visual area V2 have therapeutic relevance in the treatment of memory deficits.

Immune response to inactivated bacterial vector carrying the recombinant K39 antigen of Leishmania inf.ntum in mice

Assessment of CO_(2)storage prospect based on physical properties of Rio Bonito formation rock units

The physical properties of the rock units associated with the Rio Bonito formation are presented in this study with the focus on modelling reservoir quality based on petrophysics-derived parameters to evaluate CO_(2)storage *** involves the modelling of the reservoir depths,thicknesses,flow zone indicators(fZI),and effective permeability(Keff)and presenting the CO_(2)storage efficiency factors peculiar to the rock units of the study *** results presented by this study for the stated objectives are not quite common in the *** values range from 200 mD to higher than 2000 mD,and fZI values are generally above 1.0 mm and up to 13.0 mm within the portions covered by the drilled *** sandstone units recorded are up to 20 m thick in some *** Keff and fZI models indicate the sandstone reservoirs as permeable units to support the injection and circulation of CO_(2)within the potential reservoir units of the Rio Bonio formation across São Paulo *** from some points in the southeastern part of the study location,where the Rio Bonito are delineated at depths less than 800 m(minimum CO_(2)storage depth based on best practices),other portions are deeper,ranging from 950 m to 3500 ***-bedded layers will affect the integrity of the rocks as CO_(2)storage tanks or reservoir seals/traps/overburden within the *** bed thicknesses are up to 20 m in some ***,hybrid CO_(2)reservoir units are feasible,especially in portions where thin siltstone layers are sandwiched between sandstone units to provide considerable thicknesses based on CO_(2)storage *** current study shows that useable areas considering reservoir thickness,depth,and other physical qualities will significantly control the CO_(2)storage efficiency of the study *** studies featuring a detailed geophysical exploration of the site to confirm the availability and saturations of preexisting fluid(hydrocarbon and water)are encouraged

Localized-states quantum conf.nement induced by roughness in CdMnTe/CdTe heterostructures grown on Si(111) substrates

This work shows that despite a lattice mismatch of almost 20%, CdMnTe/CdTe/CdMnTe heterostructures grown directly on Si(111) have surprisingly good optical emission properties. The investigated structures were grown by molecular beam epitaxy and characterized by scanning transmission electron microscopy, macro-and micro-photoluminescence. Low temperature macro-photoluminescence experiments indicate three emission bands which depend on the CdTe layer thickness and have different conf.nement characteristics. Temperature measurements reveal that the lower energy emission band (at 1.48 eV)is associated to defects and bound exciton states, while the main emission at 1.61 eV has a weak 2D character and the higher energy one at 1.71 eV has a well-def.ned (zero-dimensional, 0D) 0D nature. Micro-photoluminescence measurements show the existence of sharp and strongly circularly polarized (up to 40%) emission lines which can be related to the presence of Mn in the heterostructure. This result opens the possibility of producing photon sources with the typical spin control of the diluted magnetic semiconductors using the low-cost silicon technology.

Predicting uniaxial compressive strength of tuff after accelerated freeze-thaw testing: Comparative analysis of regression models and artificial neural networks

Ignimbrites have been widely used as building materials in many historical and touristic structures in the Kayseri region of Türkiye. Their diverse colours and textures make them a popular choice for modern construction as well. However, ignimbrites are particularly vulnerable to atmospheric conditions, such as freeze-thaw cycles, due to their high porosity, which is a result of their formation process. When water enters the pores of the ignimbrites, it can freeze during cold weather. As the water freezes and expands, it generates internal stress within the stone, causing micro-cracks to develop. Over time, repeated freeze-thaw (f-T) cycles lead to the growth of these micro-cracks into larger cracks, compromising the structural integrity of the ignimbrites and eventually making them unsuitable for use as building materials. The determination of the long-term f-T performance of ignimbrites can be established after long f-T experimental processes. Determining the long-term f-T performance of ignimbrites typically requires extensive experimental testing over prolonged freeze-thaw cycles. To streamline this process, developing accurate predictive equations becomes crucial. In this study, such equations were formulated using classical regression analyses and artificial neural networks (Ann) based on data obtained from these experiments, allowing for the prediction of the f-T performance of ignimbrites and other similar building stones without the need for lengthy testing. In this study, uniaxial compressive strength, ultrasonic propagation velocity, apparent porosity and mass loss of ignimbrites after long-term f-T were determined. following the f-T cycles, the disintegration rate was evaluated using decay f.nction approaches, while uniaxial compressive strength (UCS) values were predicted with minimal input parameters through both regression and Ann analyses. Radar Absorbing Materials(RAM)are a class of composites that can attenuate incident electromagnetic waves to avoid radar *** carbon allotropes that have the potential to be used as RAM are either carbon nano-tubes(CnTs),graphene,carbon black(CB)and ultimately,sustainable porous carbon(SPC).Here,black wattle bark waste(following tannin extraction)was used as a sustainable source to produce SPC made from biomass *** was characterized and used as aﬁller for a silicone rubber matrix to produce aﬂexible *** elec-tromagnetic performance of this composite was compared with composites made with commercial CB and CnT through reﬂection loss(RL),where-10 dB is equivalent to 90%of *** composites were evaluated in single-layer,double-layer,and as radar absorbing structures(RAS)with the aim of improving their effective absorption bandwidth(EAB)performances and a reduction in *** CnT composite presented a RL of-26.85 dB at 10.89 GHz and an EAB of 2.6 GHz with a 1.9 mm thickness,while the double-layer structures using CnT and SPC provided a RL of-19.74 dB at 10.75 GHz and an EAB of 2.51 ***,the double-layer structures are~42%cheaper than the composite using only CnT since less material is ***,the largest EAB was achieved with a RAS using SPC,reaching~2.8 GHz and a RL of-49.09 dB at 10.4 ***,SPC made of black wattle bark waste can be a competitive,alternative material for use as RAM and RAS since it is cheaper,sustainable,and suitable for daily life uses such as absorbers for anechoic chambers,sensors,and elec-tromagnetic interference shields for electronics,wallets,vehicles,and others.
