Severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)causes multi-organ damage,which includes hepatic dysfunction,as observed in over 50%of COVID-19 patients.Angiotensin I converting enzyme(peptidyl-dipeptidase A)2(ACE2)is the primary receptor for SARS-CoV-2 entry into host cells,and studies have shown the presence of intracellular virus particles in human hepatocytes that express ACE2,but at extremely low levels.Consequently,we asked if hepatocytes might express receptors other than ACE2 capable of promoting the entry of SARS-CoV-2 into cells.To address this question,we performed a genome-wide CRISPR-Cas9 activation library screening and found that Asialoglycoprotein receptor 1(ASGR1)promoted SARS-CoV-2 pseudovirus infection of HeLa cells.In Huh-7 cells,simultaneous knockout of ACE2 and ASGR1 prevented SARS-CoV-2 pseudovirus infection.In the immortalized THLE-2 hepatocyte cell line and primary hepatic parenchymal cells,both of which barely expressed ACE2,SARSCoV-2 pseudovirus could successfully establish an infection.However,after treatment with ASGR1 antibody or siRNA targeting ASGR1,the infection rate significantly dropped,suggesting that SARS-CoV-2 pseudovirus infects hepatic parenchymal cells mainly through an ASGR1-dependent mechanism.We confirmed that ASGR1 could interact with Spike protein,which depends on receptor binding domain(RBD)and N-terminal domain(NTD).Finally,we also used Immunohistochemistry and electron microscopy to verify that SARS-CoV-2 could infect primary hepatic parenchymal cells.After inhibiting ASGR1 in primary hepatic parenchymal cells by siRNA,the infection efficiency of the live virus decreased significantly.Collectively,these findings indicate that ASGR1 is a candidate receptor for SARS-CoV-2 that promotes infection of hepatic parenchymal cells.

Mpox(formerly Monkeypox)is a zoonotic infection caused by Monkeypox virus(MPXV).Since 2022,Mpox epidemics have occurred in many nonendemic countries and regions,leading the World Health Organization to declare a public health emergency of international concern.With the persistent transmission and evolution of MPXV,symptoms of Mpox have become milder,with some infections being asymptomatic.In addition,MPXV has become more contagious.Therefore,rapid and accurate diagnosis and screening of MPXV is vital to prevent and control MPXV epidemics.Here,we review and summarize the technical details,application scenarios,and the advantages and disadvantages of MPXV‐specific diagnostic methods.

Since the first reported case of monkeypox in the UK in May 2022,there has been an upward trend in monkeypox cases and a global outbreak.However,reports of severe cases are relatively limited.In this study,we report a case of severe monkeypox in a patient with HIV.The patient presented with skin lesions that started on his face and around the penis and persisted for several months.Throughout the course of the disease,he received systematic symp-tomatic supportive treatment,topical remedies,and special care for the rash.He also underwent cidofovir antiviral therapy and smallpox‐vaccinated healthy population‐derived plasma therapy in succession,with the condition ultimately showing improvement after plasma treatment.After more than 3 months of hospitalization,he fully recovered.To the best of our knowledge,this is the first reported use of smallpox‐vaccinated healthy population‐derived plasma in the treatment of severe monkeypox cases.

Cancer immunotherapy has emerged as the fourth most prevalent approach to tumor treatment,alongside surgery,radiotherapy,and chemotherapy.After several decades of development,chimeric antigen receptor T(CAR-T)cell therapy,a promising branch of adoptive T-cell therapy,has demonstrated superior efficacy and safety in comparison to other cell therapies in the treatment of cancer.At present,CAR-T cells are predominantly used to treat hematological malignancies,although their application in solid tumors is being readily investigated.Although numerous studies have examined the biomarkers associated with the safety of CAR-T cell therapy,few have evaluated predictors of CAR-T cell therapeutic efficacy.Thus,the primary objective of this review article was to provide a comprehensive overview of the factors predicting the efficacy of CAR-T cell therapy,with a particular focus on biomarkers and their detection methods.

The Omicron variant of SARS-CoV-2(or Omicron)is extremely contagious and has swept the world in a short period.Shenzhen—a new international city in China with a permanent population of 17.56 million and covering nearly 2000 square kilometers—was attacked by Omicron in 2022.However,the pandemic was controlled in a very short time as a result of prompt government reaction that prevented the variant’s further spread.The total number of cases in this wave of the pandemic was more than 2600.Shenzhen’s successful experience in tackling the Omicron wave deserves in-depth discussion.Proposed by American scholar Robert Heath,the 4Rs model aims to reduce the harm from a crisis through the measures of reduction,readiness,response,and recovery.This article presents the successful experience of Shenzhen’s local government and the Third People’s Hospital of Shenzhen,the only hospital in Shenzhen designated to use the 4Rs emergency management model for the treatment of COVID-19 during the 2022 Omicron wave of the pandemic.

Human immunodeficiency virus(HIV)infection of the central nervous system(CNS)has attracted significant attention because it contributes to severe complications of acquired immunodeficiency syndrome(AIDS)and seriously impairs the life quality of infected patients.In this review,we briefly describe the latent infection of HIV in CNS and focus on the role of the important immune cells,such as T cells,in the formation and maintenance of the HIV reservoir in CNS.This review explores the mechanisms by which T cells enter CNS and establish latent infection of HIV in the CNS.In conclusion,we summarize the role of these cells in the interaction between HIV and CNS.With our better understanding of the underlying mechanisms,we propose future directions for the development of novel strategies to eliminate HIV reservoirs in the CNS based on cellular components.

Monkeypox has a very prominent regional epidemic.It has been confined to Western and Central African countries.Sporadic cases found in countries outside Africa generally have a history of sojourn in endemic areas.However,the recent multinational outbreak of monkeypox cases in Europe in early May 2022 has revealed a changing epidemiological trend,those confirmed cases had no sojourn history in endemic areas and with a high proportion of cases involving men who have sex with men(MSM).Among the MSM cases,many of them presented atypical clinical manifestations of monkeypox and with other sexually transmitted diseases co-infection.Combined with the high social interactivity in this community,there is likely a higher risk of monkeypox transmission in this population.Establishing an infectious disease surveillance system,maintaining highly vigilant regarding the transmission of monkeypox in MSM,and responding promptly are necessary and effective measures to contain the outbreak.

Dear Editor,The Omicron variant of severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)has rapidly replaced the Delta variant as a dominating SARS-CoV-2 variant because of natural selection,which favors the variant with higher infectivity and stronger vaccine breakthrough ability.1 Among three lineages BA.1(B.1.1.529.1),BA.2(B.1.1.529.2),and BA.3(B.1.1.529.3)of Omicron,BA.2 is rising rapidly worldwide since January 2022 and become a new dominating“variant of concern”in China.2 With the current surge in Omicron in China,both the absolute number of children hospitalized with COVID-19 and the percentage of total COVID-19 hospitalizations have been boosting recently.Understanding the clinical characteristics of SARS-CoV-2 Omicron subvariant BA.2 in children and the protective effect of vaccines is of great help for epidemic prevention strategies.However,so far no sufficient experimental data have been reported about infectivity,disease severity,vaccine breakthrough capability,and antibody resistance from pediatric BA.2.

The newly emerged Omicron(B.1.1.529)variant of SARS-CoV-2 is quickly overtaking the Delta variant and becoming the dominant strain around the world due to its enhanced transmissibility and high immune escape potential[1,2].Compared to the Wuhan strain,the Omicron variant carries 37 spike mutations,of which 15 are within the receptor-binding domain(RBD)(Fig.1A).A high mutation rate is associated with remarkable resistance to current vaccines and RBD-specific antibody therapeutics[2,3,4].
