Alternative relay regulates the adenosine triphosphatase activity of Locusta migratoria striated muscle myosin

Locust(Locusta migratoria)has a single striated muscle myosin heavy chain(Mhc)gene,which contains 5 clusters of alternative exclusive exons and 1 differently included penultimate exon.The alternative exons of Mhc gene encode 4 distinct regions in the myosin motor domain,that is,the N-terminal SH3-like domain,one lip of the nucleotide-binding pocket,the relay,and the converter.Here,we investigated the role of the alternative regions on the motor function of locust muscle myosin.Using Sf9-baculovirus protein expression system,we expressed and purified 5 isoforms of the locust muscle myosin heavy meromyosin(HMM),including the major isoform in the thorax dorsal longitudinal flight muscle(FL1)and 4 isoforms expressed in the abdominal intersegmental muscle(AB1 to AB4).Among these 5 HMMs,FL1-HMM displayed the highest level of actin-activated adenosine triphosphatase(ATPase)activity(hereafter referred as ATPase activity).To identify the alternative region(s)responsible for the elevated ATPase activity of FL1-HMM,we produced a number of chimeras of FL1-HMM and AB4-HMM.Substitution with the relay of AB4-HMM(encoded by exon-14c)substantially decreased the ATPase activity of FL1-HMM,and conversely,the relay of FL1-HMM(encoded by exon-14a)enhanced the ATPase activity of AB4-HMM.Mutagenesis showed that the exon-14a-encoded residues Gly474 and Asn509 are responsible for the elevated ATPase activity of FL1-HMM.Those results indicate that the alternative relay encoded by exon-14a/c play a key role in regulating the ATPase activity of FL1-HMM and AB4-HMM.

Effect of different bottom blowing elements on stirring characteristics of molten bath in converter

The bottom blowing element is the key equipment to ensure the bottom blowing effect of the converter.Three types of bottom blowing elements,dispersive type(D1),double circular seam(D2)and straight cylinder type(D3),were built,and the effects of bottom blowing element type on molten bath flow,wall erosion and furnace bottom erosion were simulated.It was found that when the bottom blowing elements of dispersive type(D1)and double circular seam(D2)were used,the dead zone area in the lower part of the molten bath was smaller,and the high-speed zone area was larger;therefore,the stirring effect on the bottom melt was better.When the straight cylinder type(D3)bottom blowing element was used,the gas penetrated the molten bath at a faster rate to reach the surface of molten bath and failed to disperse in the bottom molten bath,and the wall shear stress near the nozzle outlet was larger.When argon was blown by three different bottom blowing elements,the area of the wall shear stress greater than 3 Pa was 4.8,5.6 and 8.7 cm2,respectively,within 0.2 m of the bottom blowing nozzle outlet.

Neuroprotective effects of acteoside in a glaucoma mouse model by targeting Serta domain-containing protein 4

AIM:To explore the therapeutic effect and main molecular mechanisms of acteoside in a glaucoma model in DBA/2J mice.METHODS:Proteomics was used to compare the differentially expressed proteins of C57 and DBA/2J mice.After acteoside administration in DBA/2J mice,anterior segment observation,intraocular pressure(IOP)monitoring,electrophysiology examination,and hematoxylin and eosin staining were used to analyze any potential effects.Immunohistochemistry(IHC)assays were used to verify the proteomics results.Furthermore,retinal ganglion cell 5(RGC5)cell proliferation was assessed with cell counting kit-8(CCK-8)assays.Serta domain-containing protein 4(Sertad4)mRNA and protein expression levels were measured by qRT-PCR and Western blot analysis,respectively.RESULTS:Proteomics analysis suggested that Sertad4 was the most significantly differentially expressed protein.Compared with the saline group,the acteoside treatment group showed decreased IOP,improved N1-P1 wave amplitudes,thicker retina,and larger numbers of cells in the ganglion cell layer(GCL).The IHC results showed that Sertad4 expression levels in DBA/2J mice treated with acteoside were significantly lower than in the saline group.Acteoside treatment could improve RGC5 cell survival and reduce the Sertad4 mRNA and protein expression levels after glutamate injury.CONCLUSION:Sertad4 is differentially expressed in DBA/2J mice.Acteoside can protect RGCs from damage,possibly through the downregulation of Sertad4,and has a potential use in glaucoma treatment.

Migratable Power System Transient Stability AssessmentMethod Based on Improved XGBoost

The data-driven transient stability assessment(TSA)of power systems can predict online real-time prediction by learning the temporal features before and after faults.However,the accuracy of the assessment is limited by the quality of the data and has weak transferability.Based on this,this paper proposes a method for TSA of power systems based on an improved extreme gradient boosting(XGBoost)model.Firstly,the gradient detection method is employed to remove noise interference while maintaining the original time series trend.On this basis,a focal loss function is introduced to guide the training of theXGBoostmodel,enhancing the deep exploration of minority class samples to improve the accuracy of the model evaluation.Furthermore,to improve the generalization ability of the evaluation model,a transfer learning method based on model parameters and sample augmentation is proposed.The simulation analysis on the IEEE 39-bus system demonstrates that the proposed method,compared to the traditional machine learning-based transient stability assessment approach,achieves an average improvement of 2.16%in evaluation accuracy.Specifically,under scenarios involving changes in topology structure and operating conditions,the accuracy is enhanced by 3.65%and 3.11%,respectively.Moreover,the model updating efficiency is enhanced by 14–15 times,indicating the model’s transferable and adaptive capabilities across multiple scenarios.

Tongxinluo Activates PI3K/AKT Signaling Pathway to Inhibit Endothelial Mesenchymal Transition and Attenuate Myocardial Fibrosis after Ischemia-Reperfusion in Mice

Objective: To investigate the potential role of Tongxinluo(TXL) in attenuating myocardial fibrosis after myocardial ischemia-reperfusion injury(MIRI) in mice. Methods: A MIRI mouse model was established by left anterior descending coronary artery ligation for 45 min. According to a random number table, 66 mice were randomly divided into 6 groups(n=11 per group): the sham group, the model group, the LY-294002 group, the TXL group, the TXL+LY-294002 group and the benazepril(BNPL) group. The day after modeling, TXL and BNPL were administered by gavage. Intraperitoneal injection of LY-294002 was performed twice a week for 4 consecutive weeks. Echocardiography was used to measure cardiac function in mice. Masson staining was used to evaluate the degree of myocardial fibrosis in mice. Qualitative and quantitative analysis of endothelial mesenchymal transition(EndMT) after MIRI was performed by immunohistochemistry, immunofluorescence staining and flow cytometry, respectively. The protein expressions of platelet endothelial cell adhesion molecule-1(CD31), α-smoth muscle actin(α-SMA), phosphatidylinositol-3-kinase(PI3K) and phospho protein kinase B(p-AKT) were assessed using Western blot. Results: TXL improved cardiac function in MIRI mice, reduced the degree of myocardial fibrosis, increased the expression of CD31 and inhibited the expression of α-SMA, thus inhibited the occurrence of EndMT(P0.05). In addition, the use of the PI3K/AKT pathway-specific inhibitor LY-294002 to block this pathway and combination with TXL intervention, eliminated the protective effect of TXL, further supporting the protective effect of TXL. Conclusion: TXL activated the PI3K/AKT signaling pathway to inhibit EndMT and attenuated myocardial fibrosis after MIRI in mice.

Magnetic resonance imaging of extraocular rectus muscles abnormalities in acute acquired concomitant esotropia

AIM:To investigate the difference of medial rectus(MR)and lateral rectus(LR)between acute acquired concomitant esotropia(AACE)and the healthy controls(HCs)detected by magnetic resonance imaging(MRI).METHODS:A case-control study.Eighteen subjects with AACE and eighteen HCs were enrolled.MRI scanning data were conducted in target-controlled central gaze with a 3-Tesla magnetic resonance scanner.Extraocular muscles(EOMs)were scanned in contiguous image planes 2-mm thick spanning the EOM origins to the globe equator.To form posterior partial volumes(PPVs),the LR and MR cross-sections in the image planes 8,10,12,and 14 mm posterior to the globe were summed and multiplied by the 2-mm slice thickness.The data were classified according to the right eye,left eye,dominant eye,and non-dominant eye,and the differences in mean cross-sectional area,maximum cross-sectional area,and PPVs of the MR and LR muscle in the AACE group and HCs group were compared under the above classifications respectively.RESULTS:There were no significant differences between the two groups of demographic characteristics.The mean cross-sectional area of the LR muscle was significantly greater in the AACE group than that in the HCs group in the non-dominant eyes(P=0.028).The maximum cross-sectional area of the LR muscle both in the dominant and non-dominant eye of the AACE group was significantly greater than the HCs group(P=0.009,P=0.016).For the dominant eye,the PPVs of the LR muscle were significantly greater in the AACE than that in the HCs group(P=0.013),but not in the MR muscle(P=0.698).CONCLUSION:The size and volume of muscles dominant eyes of AACE subjects change significantly to overcome binocular diplopia.The LR muscle become larger to compensate for the enhanced convergence in the AACE.

Leakages from subsea oil and gas equipment cause substantial economic losses and damage to marine ecosystem,so it is essential to locate the source of the leak.However,due to the complexity and variability of the marine environment,the signals collected by hydrophone contain a variety of noises,which makes it challenging to extract useful signals for localization.To solve this problem,a hydrophone denoising algorithm is proposed based on variational modal decomposition(VMD)with grey wolf optimization.First,the average envelope entropy is used as the fitness function of the grey wolf optimizer to find the optimal solution for the parameters K andα.Afterward,the VMD algorithm decomposes the original signal parameters to obtain the intrinsic mode functions(IMFs).Subsequently,the number of interrelationships between each IMF and the original signal was calculated,the threshold value was set,and the noise signal was removed to calculate the time difference using the valid signal obtained by reconstruction.Finally,the arrival time difference is used to locate the origin of the leak.The localization accuracy of the method in finding leaks is investigated experimentally by constructing a simulated leak test rig,and the effectiveness and feasibility of the method are verified.

Numerous studies have shown that cell replacement therapy can replenish lost cells and rebuild neural circuitry in animal models of Parkinson’s disease.Transplantation of midbrain dopaminergic progenitor cells is a promising treatment for Parkinson’s disease.However,transplanted cells can be injured by mechanical damage during handling and by changes in the transplantation niche.Here,we developed a one-step biomanufacturing platform that uses small-aperture gelatin microcarriers to produce beads carrying midbrain dopaminergic progenitor cells.These beads allow midbrain dopaminergic progenitor cell differentiation and cryopreservation without digestion,effectively maintaining axonal integrity in vitro.Importantly,midbrain dopaminergic progenitor cell bead grafts showed increased survival and only mild immunoreactivity in vivo compared with suspended midbrain dopaminergic progenitor cell grafts.Overall,our findings show that these midbrain dopaminergic progenitor cell beads enhance the effectiveness of neuronal cell transplantation.

Objective:In this study,we screened for therapeutic targets of the Xian-Lian-jie-Du decoction(XLJDD)for colorectal cancer(CRC)and explored the underlying mechanisms using network pharmacology techniques.Methods:Genes associated with CRC were collected from the Gene Cards database.The names of the active compounds of XLJDD were used as keywords in the“chemical name”in the Traditional Chinese Medicine Systems Pharmacology(TCMSP)database to search the targets.The protein-protein interaction(PPI)network was constructed using Cytoscape 3.8.1.Gene Ontology functional enrichment and Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway enrichment analyses were performed to identify key target proteins.Results:A total of 234 XLJDD-related targets and 250 cross-targets between XLJDD and CRC were collected based on the TCMSP and HIT 2.0 databases.A PPI network constructed based on the STRING database revealed interactions for all 250 proteins.The network results revealed TP53,MYC,CCND1,AKT1,CASP3,and STAT3 as core potential targets.KEGG pathway analysis of the 250 potential XLJDD targets for CRC in the Metascape database was performed using RStudio software.The top 12 gene ratio aggregated analysis results were visualized in bubble charts.The interleukin(IL)-17 signaling pathway had the highest correlation with the tumor necrosis factor(TNF)signaling pathway.Conclusions:XLJDD may be effective in ameliorating CRC by controlling inflammatory factors related to the IL-17 and TNF pathways and targeting proto-oncogenes and tumor suppressor genes,including MYC,CCND1,CTNNB1,and TP53.
