Achieving increasingly finely targeted drug delivery to organs,tissues,cells,and even to intracellular biomacromolecules is one of the core goals of *** the delivery destination is refined to cellular and subcellular targets,it is essential to explore the delivery of nanomedicines at the molecular ***,due to the lack of technical methods,the molecular mechanism of the intracellular delivery of nanomedicines remains unclear to ***,we develop an enzyme-induced proximity labeling technology in nanoparticles(nano-EpL)for the real-time monitoring of proteins that interact with intracellular ***(lactic-co-glycolic acid)nanoparticles coupled with horseradish peroxidase(hRp)were fabricated as a model(hRp(+)-pNps)to evaluate the molecular mechanism of nano delivery in *** adding the labeling probe biotin-phenol and the catalytic substrate h_(2)O_(2)at different time points in cellular delivery,nano-EpL technology was validated for the real-time in situ labeling of proteins interacting with ***-EpL achieves the dynamic molecular profiling of 740 proteins to map the intracellular delivery of hRp(+)-pNps in macrophages over *** on dynamic clustering analysis of these proteins,we further discovered that different organelles,including endosomes,lysosomes,the endoplasmic reticulum,and the golgi apparatus,are involved in delivery with distinct participation *** importantly,the engagement of these organelles differentially affects the drug delivery efficiency,reflecting the spatial–temporal heterogeneity of nano delivery in *** summary,these findings highlight a significant methodological advance toward understanding the molecular mechanisms involved in the intracellular delivery of nanomedicines.

the Cretaceous was a significant greenhouse period in Earth's history with higher atmospheric CO_(2) levels and temperatures than today. Although evidence of combustion has been widely described from the Cretaceous deposits, our understanding of the spatiotemporal diversification pattern and process of the Cretaceous wildfires is still limited. In this study, we comprehensively synthesize a total of 271 published Cretaceous wildfire occurrences based on the by-products of burning, including fossil charcoal, pyrogenic inertinite(fossil charcoal in coal), and pyrogenic polycyclic aromatic hydrocarbons(pAhs). spatially, the dataset shows a distinctive distribution of reported wildfire evidence characterized by high concentration in the middle latitudinal areas of the Northern hemisphere(30°N-60°N) over the Cretaceous. temporally, an overall increasing trend of the reported wildfire data from the Early Cretaceous to the Late Cretaceous is coincident with higher atmospheric O_(2) levels. however, the spatial and temporal patterns may result from many types of factors, such as taphonomy, preservation, and researcher biases, instead of a real picture of the Cretaceous wildfire evolution. to better understand the spatiotemporal diversification of the Cretaceous wildfire, more investigations on the record of wildfire occurrences during this period would be necessary in the future.

AIM:to evaluate the clinical efficacy of small-diameter acellular porcine corneal stroma(sAps)for the treatment of peripheral corneal ulceration(pCU).MEthODs:this retrospective clinical study included 18 patients(18 eyes)with pCU between April 2018 and December *** patients had pCU and underwent lamellar keratoplasty with *** indicators included preoperative and postoperative best-corrected visual acuity(BCVA)and transparency of *** infection control rate in the surgical eye-lesion area was also ***:Eighteen patients underwent lamellar keratoplasty with sAps to treat *** of the patients experienced rejection after 6mo(18/18)and 12mo(16/16)of *** BCVA(0.47±0.30)at the 6mo followup after operation was significantly improved compared with the baseline(0.99±0.80),and the difference was statistically significant(Z=-3.415,pthe BCVA at the 12mo follow-up after operation was not statistically significant compared to the 6mo(Z=0,p=1).With time,the sAps graft gradually became *** the 6mo(18/18)and 12mo(16/16)follow-up,none of the patients had recurrent corneal ***:sAps is clinically effective in the treatment of pCU,improving the patient’s BCVA and reducing the incidence of rejection after keratoplasty.

Due to the depletion of conventional energy reserves,there has been a global shift towards non-conventional energy *** oil and gas have emerged as key *** resources have dense and heterogeneous reservoirs,which require hydraulic fracturing to *** process depends on identifying optimal fracturing layers,also known as‘sweet spots’.however,there is currently no uniform standard for locating these sweet *** paper presents a new model for evaluating fracturability that aims to address the current gap in the *** model utilizes a hierarchical analysis approach and a mutation model,and is distinct in its use of original logging data to generate a fracturability evaluation *** this paper’s shale fracturing sweet spot evaluation method based on a two-step mutation model,four wells in different blocks of Fuling and Nanchuan Districts in China were validated,and the results showed that the proportion of high-yielding wells on the sweet spot line could reach 97.6%,while the proportion of low-producing wells was only 78.67%.Meanwhile,the evaluation results of the model were compared with the microseismic data,and the matching results were consistent.
