Covalent DNA–protein cross-links are toxic DNA lesions that interfere with essential biological processes,which can cause serious biological consequences,such as genomic instability and protein misexpression.5-Formyluracil(5 fU) as an important modification in DNA,which is mainly from oxidative damage,exists in a variety of cells and *** have reported that 5 fU mediated DNA–protein conjugates could exist in human cells [Zhou et *** Chem.2(2020) 54–63].We now aimed to explore its potential biological effects in vitro and in *** this paper,we firstly reported that 5 fU intermediated DNA–peptide or DNA–protein conjugates(both were called DPCs) could inhibit different polymerases bypass or cause *** we further investigated the functional impacts caused by 5 fU-mediated DPCs,which appeared in different gene expression components [in the promoter sequence or 50-untranslated regions(UTR)].These results together may contribute to a broader understanding of DNA–protein interactions as well as the biological functions associated with 5 fU.

The identification of hydroxylmethyl- and formylpyrimidines in genomic DNA was a landmark event in epigenetics. Numerous laboratories in related fields are investigating the biology of these and other nucleic acid modifications. However, limitations in the ability to detect and synthesize appropriate modifications are an impediment. Herein, we explored a remarkable development in the selective detection of 5-formyluracil in both single-stranded and double-stranded DNA under mild conditions. The ＂switch-on＂ specificity towards 5-formyluracil enabled a high signal-to-noise ratio in qualitatively and quantitatively detecting materials containing 5-formyluradl, which is not affected by the presence of abasic sites and 5-formylcytosine, the modified cytosine counterpart of 5-formyluracil. In summar~ the innoxiousness, convenience, and cost-effidency of the 5-formyluracil phosphoramidite synthetic routine would promote the understanding of the epi^enetic role of this natural thvmidine modification.

5-Formyluracil(5fU)is a vital DNA marker that is widely distributed in the cells of organisms.A unique feature of 5fU is the possession of a potentially reactive aldehyde group in its structure that could realize addition and condensation ***,the biological functional details of 5fU remain mostly elusive,especially,regarding its relatedness with *** this current study,we show that 5fU bases have a strong affinity toward nucleosome core particles,and that could yield regulable DNA–protein conjugates(DPCs)via chemical interactions between amino and aldehyde groups,and reductants could be applied to stabilize or dissociate the ***,we developed a photocaged method to exploit the relationship between 5fU and ***,by applying a combination of the existence of 5fU–histone interactions in vivo by ChIP analysis of histone H4 with liquid chromatography–mass spectrometry(LC–MS),we probed further,the DPCs’influence on nucleosome and ***,our results showed that the 5fU–protein interactions increase the occupancy and stability of nucleosomes,affect enzyme recognition,and block DNA *** might imply that,in vivo,the DPCs between 5fU and nucleosome core particles might play a key role in 5fU-associated pathways such as DNA repair,transcriptional regulation,or development.

Nd-Fe-B包晶合金高梯度定向凝固试验研究

在定向凝固试验中,采用全熔和区熔两种加热方式分别获得了180K/cm和250～360K/cm的温度梯度,在5～500μm/s的抽拉速率范围内对过包晶Nd13.5Fe79.75B6.75合金进行了定向凝固试验,考察了温度梯度和抽拉速率对此合金凝固组织及相含量的影响。结果表明,合金的凝固组织包含初生α-Fe枝晶、包晶Nd2Fe14B相及少量的富Nd相。随着抽拉速率的增加,包晶相含量先增加后减少;而在同一抽拉速率下,提高温度梯度可增加铁磁性Nd2Fe14B相的含量。当抽拉速率为50μm/s时,区熔试样中(G=300K/cm)Nd2Fe14B相的含量可达约90%,而相同速率下全熔试样(G=180K/cm)中其含量约为60%。

目的分析糖化血红蛋白(HbA1c)水平正常的2型糖尿病(T2DM)肾病患者外周血单核细胞自噬相关蛋白微管相关蛋白水平与血糖变化的关系。方法选取2020年10月至2021年11月宜春市人民医院收治的110例T2DM肾病患者作为研究对象,按照血糖波动幅度(MAGE)分为对照组(MAGE<3mmol)与观察组(MAGE≥3mmol/L),每组55例。两组均进行外周血单核细胞自噬相关蛋白微管相关蛋白水平、血糖血脂指标、炎症因子及人单核细胞趋化蛋白(MCP-1)、尿游离8-异前列腺素F2α(8-isoPGF2α)水平检测,比较两组血糖血脂指标及炎症因子、人MCP-1、尿游离8-isoPGF2α、外周血单核细胞自噬相关蛋白微管相关蛋白水平[微管相关蛋白1轻链3(LC3)B-Ⅰ、LC3B-Ⅱ、LC3B-Ⅱ/LC3B-Ⅰ],采用多元线性回归分析T2DM肾病患者血糖水平波动的影响因素。结果观察组三酰甘油(TG)、空腹胰岛素(FINS)、肿瘤坏死因子-α(TNF-α)、人MCP-1、尿游离8-isoPGF2α水平均高于对照组,MAGE明显大于对照组,差异有统计学意义(P<0.05);两组总胆固醇(TC)、高密度脂蛋白胆固醇(HDL-C)、低密度脂蛋白胆固醇(LDL-C)、HbA1c水平比较差异无统计学意义。观察组LC3B-Ⅰ水平明显高于对照组,LC3B-Ⅱ水平及LC3B-Ⅱ/LC3B-Ⅰ均明显低于对照组,差异有统计学意义(P<0.05)。多元线性回归分析显示,人MCP-1、尿游离8-isoP-GF2α、MAGE、LC3B-Ⅰ、LC3B-Ⅱ水平均为T2DM肾病患者血糖水平波动的影响因素(P<0.05)。结论HbA1c水平正常的T2DM肾病患者持续的血糖波动可通过持续的氧化应激反应造成外周血单核细胞的自噬缺陷,外周血单核细胞自噬相关蛋白微管相关蛋白水平与其血糖变化有关。

目的分析肠道菌群调节治疗男性2型糖尿病(T2DM)合并骨质疏松(OP)患者的临床意义。方法选取2020年1月至2022年1月宜春市人民医院内分泌科收治的60例男性T2DM-OP患者作为研究对象,按照随机数字表法分为基础组与联药组,每组30例。基础组给予基础治疗,联药组在基础组基础上给予双歧杆菌四联活菌片治疗。比较两组血清胰岛素样生长因子-1(IGF-1)、胰高血糖素样肽1(GLP-1)、骨膜蛋白(PN)水平,Ward三角区、股骨颈、腰椎L2~4体积骨密度(vBMD),及视觉模拟评分法(VAS)评分、不良反应发生率。结果治疗后,两组联合组血清IGF-1、GLP-1水平均高于基础组,PN水平低于基础组,差异有统计学意义(P<0.05)。治疗后,联药组Ward三角区、股骨颈、腰椎L2~4 vBMD均高于基础组,VAS评分均低于低于基础组,差异有统计学意义(P<0.05)。两组不良反应发生率比较差异无统计学意义。结论双歧杆菌四联活菌片可有效改善男性T2DM-OP患者骨代谢,提高骨密度,且不良反应较少,疗效显著。

Demonstration of 60 GHz millimeter-wave short-range wireless communication system at 3.5 Gbps over 5 m range

Millimeter-wave communication has received considerable attention for use in new-generation broadband wireless communication because of the scarcity of microwave band spectrum resources. The unlicensed frequency band at 60 GHz is suitable for indoor short-range broadband communication. In this paper, we introduce a 60 GHz millimeter-wave short-range wireless communication demo system adhering to the IEEE 802.11 ad standard and targeting high-definition video streaming transmission. The system uses single-carrier transmission with frequency-domain equalization. The hardware prototype consists of a 65-nm CMOS radio-frequency front-end and a baseband transceiver for the physical layer functions. Important baseband technologies such as carrier synchronization, phase noise compensation, frequency-domain equalization, and low-density parity-check decoding based on probability calculation are also discussed. The system performance is demonstrated in an experiment on high-definition video transmission, where a data rate of 3.52 Gbps is achieved with a quadrature phase-shift keying signal over a distance of 5.109 m.

Effect of different carbon sources on performance of an A2N- MBR process and its microbial community structure

Effect of different carbon sources on purification performance and change of microbial community structure in a novel AzN-MBR process were investigated, The results showed that when fed with acetate, propionate or acetate and propionate mixed （ 1 ： 1 ） as carbon sources, the effluent COD, NH4＋- N, TN and TP were lower than 30, 5, 15 and 0.5 mg-L-1, respectively. However taken glucose as carbon source, the TP concentration of effluent reached 2.6 1 mg.L-1. Process analysis found that the amount of anaerobic phosphorus release would be the key factor to determine the above effectiveness. The acetate was beneficial to the growth of Candidatus Accumulibacter associated with biological phosphorus removal, which was the main cause of high efficiency phosphorus removal in this system. In addition, it could eliminate the Candidatus Competibacter associated with glycogen-accumulating organisms and guarantee high efficiency phosphorus uptake of phosphorus accumulating organisms in the system with acetate as carbon source.
