Evaluation of Expression Stability of Candidate References Genes among Green and Yellow Pea Cultivars (<i>Pisum sativum</i>L.) Subj.cted to Abiotic and Biotic Stress

Dry pea (Pisum sativum L.) is grown as human and animal feed throughout the world. Large yield losses in pea due to biotic and abiotic stresses compel an improved understanding of mechanisms of stress tolerance and genetic determinants conditioning these tolerances. The availability of stably expressed reference genes is a prerequisite for examining differential gene expression. The obj.ctive of this study was to examine the expression profile of several candidate reference genes across a broad range of commercial pea cultivars. Expression profiles of five candidate reference genes;18s rRNA, actin, TIF, β tubulin-2 and β tubulin-3 were examined. Relative quantifications of candidate reference genes were estimated from control plants, plants after 48 h of cold treatment, and plants 24 and 48 h after inoculation with Sclerotinia sclerotiorum, the causal agent of white mold disease of pea. rt-qPCR was performed on cDNA synthesized from three food grade spring peas, Ariel, Aragorn, and Sterling, and two spring yellow peas, Delta and Universal, which are used as animal feed. Analysis of variance (ANOVA) of CT values demonstrated significant variation between varieties and treatments under cold and disease conditions. The most abundant transcripts among tested reference genes were for 18s rRNA. Stability analysis indicated that TIF and β tubulin-3 genes were the most stably expressed candidate genes under both cold and disease stress and could serve as reference genes across a wide range of pea cultivars.

Surface-effects-dominated thermal and mechanical responses of zinc oxide nanobelts

t Molecular dynamics （MD） simulations are carried out to characterize the mechanical and thermal responses of [011^-1]-oriented ZnO nanobelts with lateral dimensions of 21.22A × 18.95 A, 31.02A× 29.42 A, and40.81A ×39.89A over the temperature range of 300-1000 K. The Young＇s modulus and thermal conductivity of the nanobelts are evaluated. Significant surface effects on properties due to the highsurface-to-volume ratios of the nanobelts are observed. For the mechanical response, surface-stress-induced internal stress plays an important role. For the thermal response, surface scattering of phonons dominates. Calculations show that the Young＇s modulus is higher than the corresponding value for bulk ZnO and decreases by -33% as the lateral dimensions increase from 21.22 A × 18.95A to 40.81 A × 39.89A. The thermal conductivity is one order of magnitude lower than the corresponding value for bulk ZnO single crystal and decreases with wire size. Specifically, the conductivity of the 21.22 A × 18.95 A belt is approximately （31-18）% lower than that of the 40.81 A × 39.89 A belt over the temperature range analyzed. A significant dependence of properties on temperature is also observed, with the Young＇s modulus decreasing on average by 12% and the conductivity decreasing by 50% as temperature increases from 300 K to 1000 K.

Studying Tuff Rings and Volcanic Hazards in a Tropical Setting: The Case of the Batoke Tuff Ring, Limbe, SW Region Cameroon

In subtropical volcanic environments, there are often few accessible outcrops. These are often highly weathered and of very poor quality. Soil development is rapid (1 cm/y) and small eruptions are unlikely to be preserved in the geological record. Reconstructing past eruptions and assessing hazards is a challenge. Here, we studied a poorly outcropping tuff ring (very poor, incomplete sections) with the best outcrop observed at a beach cliff (up to ca. 5 - 10 m high) at Batoke, to the south of Mt Cameroon volcano. Mt Cameroon has a few tuff rings, currently of unknown ages, near the SW coast of Cameroon. In the Batoke case, the sequence is dominated by gently dipping tuff beds varying in the proportion of lithics, j.venile clasts, and accretionary lapilli (acc-laps). Several beds are close-packed with acc-laps of up to 10 - 15 mm diameter. Part of the section is gullied by mud flow deposits. The rocks are highly weathered but differential weathering enhances relationships. Quantitative data can be extracted from a detailed study of outcrops’ external surfaces. The preserved section is close to where the deposits were initially thickest and where acc-laps were most abundant and largest. There is an empirical correlation between maximum acc-lap size in the thickest outcrop and eruption column height. This and the deposit features suggest that the Batoke eruption was pulsating but dominated by fallout, with a water and ice-rich eruption column reaching 10 - 15 km high. Recycling of water drops and ice-coated fine ash accumulated during eruption. At switch off, wholesale gravitational collapse of this material produced the mud flows, which gullied the previously-laid down deposits. Such ash fall and mud flows can represent a substantial hazard, e.g. they can gully down through towns and roads and cut evacuation routes. This study illustrates how, at subtropical tuff rings, it is possible to extract key data needed for hazard assessment from only 1 - 2 poor outcrops.

Using the win ratio to compare laparoscopic versus open liver resection for colorectal cancer liver metastases

Background:We sought to assess the overall benefit of laparoscopic versus open hepatectomy for treatment of colorectal liver metastases(CRLMs)using the win ratio,a novel methodological ***:CRLM patients undergoing curative-intent resection in 2001-2018 were identified from an international multi-institutional *** were paired and matched based on age,number and size of lesions,lymph node status and receipt of preoperative *** win ratio was calculated based on margin status,severity of postoperative complications,90-day mortality,time to recurrence,and time to ***:Among 962 patients,the maj.rity underwent open hepatectomy(n=832,86.5%),while a minority underwent laparoscopic hepatectomy(n=130,13.5%).Among matched patient-to-patient pairs,the odds of the patient undergoing laparoscopic resection“winning”were 1.77[WR:1.77,95%confidence interval(CI):1.42-2.34].The win ratio favored laparoscopic hepatectomy independent of low(WR:2.94,95%CI:1.20-6.39),medium(WR:1.56,95%CI:1.16-2.10)or high(WR:7.25,95%CI:1.13-32.0)tumor burden,as well as unilobar(WR:1.71,95%CI:1.25-2.31)or bilobar(WR:4.57,95%CI:2.36-8.64)*** odds of“winning”were particularly pronounced relative to short-term outcomes(i.e.,90-day mortality and severity of postoperative complications)(WR:4.06,95%CI:2.33-7.78).Conclusions:Patients undergoing laparoscopic hepatectomy had 77%increased odds of“winning”.Laparoscopic liver resection should be strongly considered as a preferred approach to resection in CRLM patients.
