Effect of parameters on Si plasma emission in collinear double-pulse laser-induced breakdown spectroscopy

Collinear dual-pulse laser-induced breakdown spectroscopy was carried out on Si crystal by using a pair of nanosecond Nd：YAG laser sources emitting at 1064 nm. The spectral intensities and signal- to-noise ratios of selected Si atomic and ionic lines were used to evaluate the optical emission. The optical emission intensity was recorded while varying the interpulse delay time and energy ratio of the two pulsed lasers. The effects of the data acquisition delay time on the line intensity and signal-to-noise ratio have been investigated as well. Based on the results, the optimal interpulse delay time, energy ratio of the two pulsed lasers, and data acquisition delay time for achieving the maximum atomic and ionic line intensities were found for generation of Si plasma with the collinear dual-pulse laser approach. The dominant mechanism for the observed line intensity variation was also discussed. In addition, the plasma temperature and electron number density at different gate delay times and different interpulse delay times were derived. A significant influence of plasma shielding on the electron temperature and electron number density at shorter interpulse delay times was observed.

Upregulation of Neural Precursor Cell Expressed Developmentally Downregulated 4-1 is Associated with Poor Prognosis and Chemoresistance in Lung Adenocarcinoma

Background： The E3 ubiquitin ligasc neural precursor cell expressed developmentally downregulated 4-1（N EDD4-1） negatively regulates phosphatase and tensin homolog deleted on chromosome 10 （PTEN） protein levels through polyubiquitination and proteolysis, but its significance in lung cancer is still unclear. This study investigated the expression and the role of NEDD4-1 in tumor developnaent and chemosensitivity of lung adenocarcinoma （ADC）. Methods： We retrospectively investigated the expression and significance ofNEDD4-1, PTEN, and p-Akt proteins in 135 paired A DC and adjacent noncancerous tissue specimens using immunohistochemistry. Furthemaore, we evaluated the relationship between NEDD4-1 expression and clinicopathologic characteristics and prognosis. The effects of small interfering RNA against NEDD4-1 on proliferation and chemosensitivity were examined in A549 cells in vitro using 3- （4,5-dimethylthiazol-2-yl） -5-（3-carboxymethoxyphenyl） -2-（4-sulfophenyl）- 2H-tetrazoliunl method. The ability of migration and invasion ofA549 cells was tested by transwell assay. Moreover, reverse-transcription quantitative polymerase chain reaction and Western blotting analyses were used to determine the expression of NEDD4-1, PTEN, phosphoinositide 3-kinase （PI3K）/Akt activity, and its downstream target proteins. Results： NEDD4-1 protein was significantly upregulated in lung ADC tissues, whereas it was weak or negative in normal lung epithelial cells. The expression ofNEDD4-1 in ADC （78.5%, 106/135） was significantly much higher than that in adjacent normal lung tissue （ 13.3%, 29/135, P 〈 0.01）, and it was associated with lymph node metastasis, tumor-node-metastasis （TNM） stage, and chemotherapy resistance. PTEN expression was downregulated in lung ADC （60.7% vs. 100.0% in noncancerous specimens, P - 0.007）, and was negatively correlated with lymph node metastasis, histological variants, clinical stage, chemoresistance. Decadal expansion and contraction of permafrost in the Three-River Source Region,Qinghaie-Tibet Plateau(1901-2020)

Permafrost and its spatiotemporal variation considerably influence the surface and sub-surface hydrological processes,biogeochemical cycles,fauna and flora growth and cold region engineering projects in the Three-River Source Region(TRSR),Qinghai–Tibet ***,the dynamics of permafrost over a relatively long term duration(e.g.>100 years)in the TRSR is not well ***,the spatial and temporal variations of the temperature at the top of the perennially frozen/unfrozen ground(TTOP),active layer thickness(ALT)in permafrost regions and the maximum depth of frost penetration(MDFP)in the seasonally frozen ground of the TRSR during 1901–2020 were simulated using the TTOP model and Stefan equation driven by the widely used reanalysis Climatic Research Unit 4.05 *** revealed that the permafrost in the TRSR over the past 120 years did not degrade monotonically but experienced considerable fluctuations in area with the decadal oscillations of climate warming and cooling:shrinking from 263.9×10^(3)km^(2)in the 1900s to 233.3×10^(3)km^(2)in the 1930s,expanding from 232.3×10^(3)km^(2)in the 1940s to 260.9×10^(3)km^(2)in the 1970s and shrinking again from 254.1×10^(3)km^(2)in the 1980s to 228.9×10^(3)km^(2)in the *** regional average TTOP increased from−1.34±2.74℃in the 1910s to−0.48±2.69℃in the 2010s,demonstrating the most noticeable change for the extremely stable permafrost(TTOP3.0 m by 12%from 1901 to ***,minor changes were observed for the regional average MDFP,probably due to the increase in the area proportion of MDFP3.5 m(owing to the transformation of permafrost to seasonally frozen ground)by 7.39%and 4.77%,*** findings can facilitate an in-depth understanding of permafrost dynamics and thus provide a scientific reference for eco-environment protection and s
