Proteomic profile of human monocytic cells infected with dengue virus

Objective: To identify the changes in the proteome of U937 cells infected with dengue virus(DENV).Methods: In this study, differentiated U937 cultures were infected with two DENV-2strains, one of which was associated with dengue(DENV-2/NG) and the other one with severe dengue(DENV-2/16681), with the aim of determining the cellular proteomic profiles under different infection conditions. Cellular proteins were extracted and separated by two-dimensional electrophoresis, and those proteins with differential expression profiles were identified by mass spectrometry. The obtained results were correlated with cellular viability, the number of infectious viral particles, and the viral DNA/protein ***: In comparison with non-infected cultures, in the cells infected with the DENV-2/NG strain, nine proteins were expressed differentially(five were upregulated and four were downregulated); in those cultures infected with the DENV-2/16681 strain, six proteins were differentially expressed(two were downregulated and four were upregulated). The downregulated proteins included fatty acid-binding protein, heterogeneous nuclear ribonucleoprotein 1, protein disulfide isomerase, enolase 1, heat shock 70 k Da protein 9, phosphotyrosyl phosphatase, and annexin IV. The upregulated proteins included heat shock 90 k Da protein AA1, tubulin beta, enolase 1, pyruvate kinase,transaldolase and phospholipase ***: Because the monocyte/macrophage lineage is critical for disease pathogenicity, additional studies on these proteins could provide a better understanding of the cellular response to DENV infection and could help identify new therapeutic targets against infection.

Application of a new reagent for analysis of oxygen presence in a lowcarbon steel wire rod

A microstructural analysis was performed to determine the presence of oxygen in a wire rod section of AISI 1008 steel with a surface mechanical failure produced during wire *** failure zone was analyzed by comparison using three different attack reagents:alkaline sodium chromate(ASC),ASC with hydrogen peroxide and amyl alcohol,and a solution of nitric acid in ethyl *** reagents were applied in samples in the failure zone,showing different types of zones in regions with internal and superficial defects such as carbides,pores,cracks,deformation,and detachment zones,indicating the possible presence of *** areas identified were observed by means of an optical microscope and were correlated with the content of elements that were present in the region using a scanning electron microscope and a scattered X-ray energy spectrometer,which determined that the areas identified by chromate sodium correspond to regions with a high concentration of oxygen and slag-forming elements such as silicon,aluminum,and iron;this is associated with the presence of oxygen in the oxide *** was also observed that the ASC and the modified ASC solution with hydrogen peroxide and amyl alcohol could identify areas with a high presence of oxides,while the nitric acid solution only identified the steel *** modified ASC solution is an alternative to identify the presence of chemical variants of oxygen because the conventional formulation is unstable and has a too short shelf life,and therefore,its application must be carried out at the time of preparation.

Adaptive phase field modelling of crack propagation in orthotropic functionally graded materials

Photocatalytic performance of K2Ti6O13 whiskers to H2 evolution and CO2 photo-reduction

K2Ti6O13 whiskers were synthesized by conventional sol-gel method, sono-chemical assisted and microwave assisted sol-gel method in order to obtain catalysts with different particle sizes and to modify their optical, textural and electrochemical properties. These modifications improved their photocatalytic activity for H2 evolution and CO2 photo-reduction. long K2Ti6O13 whiskers prepared by ultrasound assisted sol-gel method are the most active photocatalysts for the hydrogen evolution reaction using pure water as reactant (U-SG, 10,065 μmol g^-1). In contrast, an opposite behavior was observed using a mixture of ethanol-water, where the highest activity was achieved by the shortest and less crystalline K2Ti6O13 whiskers (C-SG, 3,2871 μmol g^-1). In case of CO2 photo-reduction, long whiskers that were also prepared by the sono-chemical assisted sol-gel method were the most active to transform CO2 to formaldehyde, methane, methanol and hydrogen. The EFB value of this catalyst is located very close to the potential for formaldehyde production and favors the selectivity to this organic product.

Concentrations and sources of an emerging pollutant, decabromodiphenylethane (DBDPE), in sewage sludge for land application

The presence of an emerging brominated flame retardant, decabromodiphenylethane （DBDPE）, has been confirmed in Spanish sewage sludge. Thirty one samples from different urban wastewater treatment plants （WWTPs） were analyzed for this brominated flame retardant. DBDPE was positively identified and quantified in all samples at lower concentrations （47.0 ± 29.7 ng/g dry weight （dw）; mean ± SD） than those obtained for decabromodiphenyl ether （BDE-209）（290 ± 236 ng/g dw; mean ± SD） in a previous study. Influence of the WWTP characteristics in the pollutant levels was evaluated. No significant correlations were obtained between DBDPE concentrations and the population or sewage sludge production rate associated with the plants, neither wastewater treatment method. Sources of DBDPE in the sludge were also evaluated. Data indicate a common origin for DBDPE and BDE-209, which may be related to leaching processes during the use and disposal of consumer products containing these chemicals. Nevertheless, DBDPE contents are not influenced by industrial activities, which suggests that the infusion of DBDPE commercial mixture is not a source of this chemical into the environment, and indicates that the use of DBDPE in the Spanish industry is still low compared to deca-BDE.

The relationship between DNA fragmentation and the intensity of morphologically abnormal human spermatozoa

Abdominal aortic aneurysm (AAA) is a common degenerative vascular disease with a prevalence up to 8% in males over 60 years of age.1 This is a complex disorder characterized by permanent and focal dilation of abdominal aorta that exceeds at least 50% of the normal diameter. In these patients, the aortic diameter progressively expands boosted by smoking and aging as major risk factors. Unfortunately, aneurysm growth increases the risk of aortic rupture, a life-threatening emergency that carries a mortality rate of 80%. Despite the increasing effort of the scientific community to identify therapeutic strategies for AAA, there are currently no pharmacological tools that ameliorate aneurysm expansion, while the underlying mechanisms involved in this disease are not completely understood.

Unraveling the factors that determine variation of diversity in tropical mountain systems is a topic for debate in plant *** is especially true in areas where topography is complex due to volcano elevational gradients and where forests are vulnerable to human *** this study we used a set of climatic(temperature,rainfall,and radiation solar),topographic(elevation,slope aspect,and slope orientation)and human disturbance variables to determine their effect on diversity and composition patterns of a tree community,considering three slope aspects of a tropical volcano in southeastern *** sampled trees in seventy 0.1-ha plots distributed on three slope aspects of the Tacanávolcano along an elevational gradient of 1500 to 2500 *** determined diversity patterns(general tree richness,exponential of Shannon index,and pioneer species richness)with linear regression models,and for beta diversity,we used a dissimilarity index(within and between elevational bands 100 m wide).The effect of a set of environmental and human disturbance variables on tree diversity and community composition was analyzed with general linear models and multivariate analyses,*** registered 2,949 individual trees belonging to 176 species and *** average species richness and alpha diversity per plot were 13(standard deviation±6)and 9(±5),*** tree richness and alpha diversity increased in the middle part(unimodal patterns)of the elevational gradient,but pioneer species richness decreased linearly with *** variance explained by general linear models was greater in richness(32%)than in alpha diversity(25.3%).The most important predictor variables were temperature(elevational gradient),which explained the unimodal pattern(richness and alpha diversity increase at intermediate levels of temperature),and slope orientation,which explained the increase in richness and alpha diversity toward the geographic *** temperature had a signific

Parametrization of the Teichmüller Space of Bordered Surface NEC Groups

A non-Euclidean crystallographic group F （NEC group, for short） is a discrete subgroup of isometries of the hyperbolic plane H, with compact quotient space H/Г. These groups uniformize Klein surfaces, surfaces endowed with dianalytic structure. These surfaces can be seen as a generalization of Riemann surfaces. Fundamental polygons play an important role in the study of parametrizations of the Teichmuller space of NEC groups. In this work we construct a class of right-angled polygons which are fundamental regions of bordered surface NEC groups. The free parameters used in the construction of the polygons give a parametrization of the Teichmuller space. From the parameters we obtain explicit matrices of the generators of the groups. Finally, we give examples to exhibit how different relations between the parameters reflect the existence of automorphisms on the quotient surfaces.
