Large-scale frequent testing and tracing to supplement control of Covid-19 and vaccination rollout constrained by supply

Non-pharmaceutical interventions(NPI)were implemented all around the world in the fight against COVID-19:Social distancing,shelter-in-place,mask wearing,*** mitigate transmission,together with testing and contact-tracing to identify,isolate and treat the *** majority of countries have relied on the former measures,followed by a ramping up of their testing and tracing *** present here the cases of South Korea,Italy,Canada and the United States,as a look back to lessons that can be drawn for controlling the pandemic,specifically through the means of testing and *** fitting a disease transmission model to daily case report data in each of the four countries,we first show that their combination of social-distancing and testing/tracing have had a significant impact on the evolution of their first wave of pandemic *** then consider the hypothetical scenario where the only NPI measures implemented past the first pandemic wave consisted of isolating individuals due to repeated,country-scale testing and contact tracing,as a mean of lifting social distancing measures without a resurgence of *** give estimates on the average isolation rates needed to occur in each *** find that testing and tracing each individual of a country,on average,every 4.5 days(South Korea),5.7 days(Canada),6 days(Italy)and 3.5 days(US),would have been sufficient to mitigate their second pandemic *** also considered the situation in Canada to see how a frequent large-scale asymptomatic testing and contact tracing could have been used in combination with vaccination rollout to reduce the infection in the *** could offer an alternative approach towards preventing and controlling an outbreak when vaccine supply is limited,while testing capacity has been increasingly enhanced.

The contribution of Renewable Energy Resources(RER)in the process of power generation is signiﬁcantly high in the recent days since it paves the way for overcoming the issues like serious energy crisis and natural *** paper deals with the renewable energy based micro-grid as it is regarded as the apt solution for integrating the RER with the electrical *** theﬁxed droop coefﬁcients in conventional droop control approaches have caused various limitations like low power-sharing and sudden drops of grid voltage in the Direct Current(DC)side,the Harmonized Membership Fuzzy Logic(MFL)droop control is employed in this present *** proposed droop control for the hybrid PV-wind-battery system with MFL assists in achieving proper power-sharing and minimizing Total Harmonic Distortion(THD)in the emer-gency *** eradicates the deviations in voltage and frequency with itsﬂexible and robust *** THD is reduced and attains the value of 3.1%compared to the traditional droop *** simulation results of harmo-nized MFL droop control are analogized with the conventional approaches to vali-date the performance of the proposed *** addition,the experimental results provided by the Field Programmable Gate Array(FPGA)based laboratory setup built using a solar photovoltaic(PV)and wind Permanent Magnet Synchro-nous Generator(PMSG)reafﬁrms the design.

Mini-Review:Green sustainable processes using supercritical fluid carbon dioxide

Environmentally benign carbon dioxide offers significant potential in its supercritical fluid phase to replace current reliance on a range of hazardous, relatively expensive and environmentally damaging organic solvents that are used on an extensive global basis. The unique combination of the physical properties of supercritical fluids are being exploited and further researched to continue the development and establishment of high efficiency, compact plant to provide energy and water efficient manufacturing processes. This mini-review is focused on the use and potential applications of supercritical fluid carbon dioxide for a selected range of key and emerging industrial processes as a sustainable alternative to totally eliminate or greatly reduce the requirement of numerous conventional organic solvents. Examples of the industries include： chemical extraction and purification, synthetic chemical reactions including polymerization and inorganic catalytic processes. Biochemical reactions involving enzymes, particle size engineering, textile dyeing and advanced material manufacture provide further illustrations of vital industrial activities where supercritical fluid technology processes are being implemented or developed. Some aspects relating to the economics of sustainable supercritical fluid carbon dioxide processes are also considered.

Meaningful Contact Estimates among Children in a Childcare Centre with Applications to Contact Matrices in Infectious Disease Modelling

we present a mathematical model of a day care center in a developed country (such as Canada), in order to use it for the estimation of individual-to-individual contact rates in young age groups and in an educational group setting. In our model, individuals in the population are children (ages 1.5 to 4 years) and staff, and their interactions are modelled explicitly: person-to-person and person-to-environment, with a very high time resolution. Their movement and meaningful contact patterns are simulated and then calibrated with collected data from a child care facility as a case study. we present these calibration results as a first part in the further development of our model for testing and estimating the spread of infectious diseases within child care centers.

Regnase-1, a rapid response ribonuclease regulating nflammation and stress responses

RNA-binding proteins （RBPs） are central players in post-transcriptional regulation and immune homeostasis. The ribonuclease and RBP Regnase-1 exerts critical roles in both immune cells and non-immune cells. Its expression is rapidly induced under diverse conditions including microbial infections, treatment with inflammatory cytokines and chemical or mechanical stimulation. Regnase-1 activation is transient and is subject to negative feedback mechanisms including proteasome-mediated degradation or mucosa-associated lymphoid tissue 1 （MALT1） mediated cleavage. The major function of Regnase-1 is promoting mRNA decay via its ribonuclease activity by specifically targeting a subset of genes in different cell types. In monocytes, Regnase-1 downregulates IL-6 and IL-12B mRNAs, thus mitigating inflammation, whereas in T cells, it restricts T-cell activation by targeting c-Rel, Ox40 and 11-2 transcripts. In cancer cells, Regnase-1 promotes apoptosis by inhibiting anti-apoptotic genes including Bcl2L1, Bcl2A1, RelB and Bcl3. Together with up-frameshift protein-1 （UPF1）, Regnase-1 specifically cleaves mRNAs that are active during translation by recognizing a stem-loop （SL） structure within the 3＇UTRs of these genes in endoplasmic reticulum-bound ribosomes. Through this mechanism, Regnase-1 rapidly shapes mRNA profiles and associated protein expression, restricts inflammation and maintains immune homeostasis. Dysregulation of Regnase-1 has been described in a multitude of pathological states including autoimmune diseases, cancer and cardiovascular diseases. Here, we provide a comprehensive update on the function, regulation and molecular mechanisms of Regnase-1, and we propose that Regnase-1 may function as a master rapid response gene for cellular adaption triggered by microenvironmental changes.

Visualization of atomic scale reaction dynamics of supported nanocatalysts during oxidation and ammonia synthesis using in-situ environmental(scanning) transmission electron microscopy

SH-Mode Seismic-Reflection Imaging of Earthfill Dams

Assessing subsurface characteristics and imaging geologic features （e.g., faults, cavities, low-velocity lay- ers, etc.） are typical problems in near-surface geophysics. These questions often have adverse geotechni-cal engineering implications, and can be especially acute when associated with high-hazard structures such as large earthen flood-control dams. Dam-related issues are becoming more frequent in the United States, because a large part of this major infrastructure was designed and constructed in the early- to mid-twentieth century; these dams are thus passing into the latter stages of their design life, where minute flaws that were overlooked or thought to be insignificant in design/construction are now proving problematic. The high-hydraulic heads associated with these structures can quicken degra-dation of weak areas and compromise long-term integrity. Addressing dam-related problems solely with traditional invasive drilling techniques is often inadequate （i.e., lack of lateral resolution） and]or econom- ically exorbitant at this scale. However, strategic geotechnical drilling integrated with the broad utility of near-surface geophysics, particularly the horizontally polarized shear-wave （SH-mode） seismic-reflection technique for imaging the internal structural detail and geological foundation conditions of earthfill embankment dams can cost-effectively improve the overall subsurface definition needed for remedial engineering. Demonstrative evidence for this supposition is provided in the form of SH-wave seismic-reflection imaging of in situ and engineered as-built components of flood-control embankment dams at two example sites in the central United States.

Utilization of longitudinal ultrasound to quantify joint soft-tissue changes in a mouse model of posttraumatic osteoarthritis

To assess the utility of longitudinal ultrasound （US） to quantify volumetric changes in joint soft tissues during the progression of posttraumatic osteoarthritis （PTOA） in mice, and validate the US results with histological findings. A longitudinal cohort of 3-month-old wild-type C57BL/6 male mice received the Hulth-Telhag surgical procedure on right knee to induce ~OA, and sham surgery on their left knee as control. US scans were performed on both knees before, 2, 4, 6, and 8 weeks post-surgery. Joint space volume and Power-Doppler （PD） volume were obtained from US images via Amira software. A paraUel cross-sectional cohort of mice was killed at each US time point, and knee joints were subjected to histological analysis to obtain synovial soft-tissue area and OARSI scores. The correlation between US joint space volume and histological synovial soft-tissue area or OARSI score was assessed via linear regression analysis. US images indicated increased joint space volume in PTOA joints over time, which was associated with synovial inflammation and cartilage damage by histology. These changes started from 2 weeks post-surgery and gradually became more severe. No change was detected in sham joints. Increased joint space volume was significantly correlated with increased synovial soft-tissue area and the OARSI score （P 〈 0.001）. PD signal was detected in the joint space of PTOA joints at 6 weeks post-surgery, which was consistent with the location of blood vessels that stained positively for CD31 and alpha-smooth muscle actin in the synovium. This study indicates that US is a cost-effective longitudinal outcome measure of volumetric and vascular changes in joint soft tissues during PTOA progression in mice, which positively correlates with synovial inflammation and cartilage damage.

Sustainable oil-in-water analysis using a supercritical fluid carbon dioxide extraction system directly interfaced with infrared spectroscopy

A direct aqueous supercritical fluid extraction （SFE） system using carbon dioxide provides a sustainable means by which a vast range of industries may continue to depend on well established infrared （IR） techniques to determine oil-in-water. The SFE-IR method provides an environmentally friendly substitute for current national standard IR reference methods for measuring oil-in-water that rely on using increasingly restricted ozone depleting solvents whose manufacture is being phased out in accordance with international law. The SFE-IR analysis of a 500 mL water sample can be accomplished in 15 min. A rapid on-line SFE-IR calibration method has been implemented. with this calibration method, SFE-IR accuracy for determining diesel oil in 500 mL spiked water samples using single wave number measurement was 86.0%-98.8% with precision （RSD） ranging from 2.5%-7.0%. Using a general purpose calculation which involves measuring infrared absorbance values at three different wave numbers, SFE-IR method accuracy for determining diesel oil in 500 mL spiked water samples was 83.7%-92.2% with RSD 1.0%-9.3%. Data is presented that indicates current long established national standard IR reference methods involving three wave number calculations should be reviewed since, without careful consideration, the inclusion of calculated aromatic hydrocarbon species contributions to final oil-in-water concentration values may provide less accurate results.
