Probiotics modulate the microbiota-gut-brain axis and improve memory deficits in aged SAMP8 mice

ProBiotic-4 is a probiotic preparation composed of Bifidobacterium lactis,Lactobacillus casei,Bifidobacterium bifidum,and Lactobacillus *** study aims to investigate the effects of ProBiotic-4 on the microbiota-gut-brain axis and cognitive deficits,and to explore the underlying molecular mechanism using senescence-accelerated mouse prone 8(SAMP8)***-4 was orally administered to 9-month-old SAMP8 mice for 12 *** observed that ProBiotic-4 significantly improved the memory deficits,cerebral neuronal and synaptic injuries,glial activation,and microbiota composition in the feces and brains of aged SAMP8 ***-4 substantially attenuated aging-related disruption of the intestinal barrier and blood-brain barrier,decreased interleukin-6 and tumor necrosis factor-αat both mRNA and protein levels,reduced plasma and cerebral lipopolysaccharide(LPS)concentration,toll-like receptor 4(TLR4)expression,and nuclear factor-κB(NF-κB)nuclear translocation in the *** addition,not only did ProBiotic-4 significantly decreased the levels ofγ-H2 AX,8-hydroxydesoxyguanosine,and retinoic-acid-inducible gene-I(RIG-I),it also abrogated RIG-I multimerization in the *** findings suggest that targeting gut microbiota with probiotics may have a therapeutic potential for the deficits of the microbiota-gut-brain axis and cognitive function in aging,and that its mechanism is associated with inhibition of both TLR4-and RIG-I-mediated NF-κB signaling pathway and inflammatory responses.

目的研究新型口腔凝胶对大鼠牙龈炎的治疗作用。方法通过临床正畸用钢丝结扎健康SD大鼠左上颌第二磨牙(M2)颈部,每天给予大鼠10%高糖水浸饲料,连续2周,诱导大鼠实验性牙龈炎。选取模型成功的大鼠,随机分为模型组和口腔凝胶组,另设不做处理的对照组。治疗7 d后,观察大鼠的探诊深度、龈沟出血指数和牙龈指数等临床指标,通过q-PCR检测白细胞介素(IL)-6、IL-1βmRNA的含量,HE染色观察牙龈组织病理学的改变。结果与模型组比较,治疗7 d后,口腔凝胶组大鼠的牙龈红肿明显减轻,探诊深度、龈沟出血指数和牙龈指数均有所下降,IL-6、IL-1βmRNA的含量也明显降低,牙龈组织水肿明显改善。结论新型口腔凝胶可改善大鼠实验性牙龈炎的临床指标,并缓解其症状。

目的研究维替泊芬(verteporfin,VP)对肺纤维化小鼠的治疗作用及其机制。方法给予C57BL/6小鼠气管内注射博来霉素(bleomycin,BLM)建立肺纤维化模型。造模后第8天开始使用VP(100 mg·kg-1)治疗,每3天腹腔注射一次,连续给药两周。于造模后第21天处死小鼠,计算体重变化率和肺系数;采用HE染色和Masson染色观察小鼠肺部的纤维化病变;羟脯氨酸(hydroxyproline,HYP)试剂盒测定肺组织的HYP水平;q-PCR检测α-平滑肌肌动蛋白(alpha-smooth muscle actin,α-SMA)、Yes相关蛋白(Yes-associated protein,YAP)和白细胞介素11(interleukin-11,IL-11)mRNA表达水平;Western blot法检测Ⅰ型胶原蛋白(CollagenⅠ)、α-SMA、YAP和核因子κB p65(nuclear factor kappa-B,NF-κB p65)的蛋白表达水平;ELISA试剂盒检测肺组织匀浆中IL-11水平。结果经过VP治疗后,小鼠的体重变化率和肺系数降低;HE染色和Masson染色中的纤维化病变及胶原沉积有改善;肺组织的HYP、CollagenⅠ和α-SMA水平降低;VP也降低了肺组织中YAP、NF-κB p65和IL-11的表达。结论VP可减轻BLM诱导的小鼠肺纤维化,其治疗作用与IL-11的下调相关。

Effects of <i>Streptococcus salivarius</i>K12 on Experimental Periodontitis and Oral Microbiota in Mice

Background: Periodontal diseases comprise a wide range of inflammatory conditions that affect the supporting structures of the teeth, and may lead to severe periodontal destruction and even tooth loss.?Streptococcus salivarius K12 (S. salivarius K12), one of oral probiotics, has been reported to be able to inhibit various potentially deleterious bacteria. This study was the first time to investigate the effects of S. salivarius K12 on ligature-induced periodontitis in mice. Methods: A silk ligature was tied around the second left maxillary molar to establish the model of periodontitis, and then, mice in group S. salivarius K12 were administered with S. salivarius K12 (2 x 109 CFU) twice daily for 30 d, the others were treated with isopyknic water. Results: Administration with S. salivarius K12 markedly decreased the anaerobic bacteria accumulation on the teeth, and it also considerably alleviated periodontitis-induced alveolar bone resorption and attachment loss. Moreover, S. salivarius K12 administration increased the relative species abundance of Gram-positive bacterium in oral cavity while decreased Gram-negative bacterium, as well as the diversity of the bacterial community. Conclusion: Oral administration of the probiotic S. salivarius K12 may mitigate the alveolar bone resorption and attachment loss of periodontitis by modulating the oral microbiota.

为探讨丹参酮ⅡＡ对人血管平滑肌细胞增殖的影响与机制，用四甲基偶氮唑兰比色法测定细胞线粒体脱氢酶活性，以评价该药的细胞毒性；在碱性纤维母细胞生长因子诱导的血管平滑肌细胞增殖模型上，用氚标胸腺嘧啶脱氧核苷掺入法观察该药对ＤＮＡ合成的影响；用细胞计数法测定该药对平滑肌细胞增殖的影响。结果发现，在无细胞毒性的浓度范围内，丹参酮ⅡＡ剂量依赖性地抑制碱性纤维母细胞生长因子诱导的人血管平滑肌细胞ＤＮＡ的合成，ＩＣ５０为２．８μｍｏｌ／Ｌ；丹参酮ⅡＡ１５μｍｏｌ／Ｌ孵育６天，显著抑制细胞增殖（Ｐ＜０．０５）。提示丹参酮ⅡＡ对生长因子为始因引起的血管成形术后再狭窄、慢性高血压。

Kanggan Granule Ameliorates Dexamethasone-Induced Immunosuppression in Mice

Objective: This study was to investigate the effect of the Chinese herbal compound Kanggan granule (KG) on immune function in a mouse model of immunosuppression and its possible mechanism of action. Method: ICR mice were randomly divided into a normal control group (untreated non-immunosuppressed, Control), untreated immunosuppressed group (Model), positive control group (immunosuppressed and treated with 1.6 g/kg astragalus granule [AG]), high-dose KG group (immunosuppressed and treated with 24 g/kg, KG-24), and low-dose KG group (immunosuppressed and treated with 6 g/kg, KG-6). Each group received intragastric administration once daily for 7 days. Immunosuppression was induced by an intraperitoneal injection of dexamethasone (25 mg/kg) once daily beginning on day 1 for 3 days. To illuminate the mechanism of immunomodulatory, we studied the effects of KG on nonspecific immunity, humoral immunity and cellular immunity in mice respectively. Results: KG improved organ weights and improved the phagocytic ability of mononuclear macrophages in immunosuppressed mice (p p < 0.05). The proliferation of spleen lymphocytes and number of peripheral blood leukocytes were enhanced after KG treatment in immunosuppressed mice (p +/CD8+ ratio in immunosuppressed mice (p p Conclusions: KG can improve immune function in immunosuppressed mice. Nonspecific immunity, humoral immunity, and cellular immunity were all enhanced.
