20-Hydroxyecdysone from Dacrycarpus imbricatus bark inhibits the proliferation of acute myeloid leukemia cells

Objective:To investigate the anti-proliferative effects of 20-hydroxyecdysone isolated from the bark of Dacrycarpus imbricatus(Blume) de ***:Column chromatography was used for isolation of compounds from plant *** structure of the isolated compound was identified by mass spectrometry and nuclear magnetic resonance techniques,including HSQC,HMBC,NOE-difference *** isolated compound was tested for its anti-proliferative activity in acute myeloid leukemia(AML) and OCI-AML ***:Compound 1 was isolated from the ethyl acetate fraction of Dacrycarpus imbricatus barks by column *** chemical structure was identified as 20-hydroxyecdysone(20HE),a cholestane-type ecdysteroid,by a combination of mass spectrometry and nuclear magnetic resonance spectrometric *** goal was to test the anti-proliferative activity of 20 HE using the OCI-AML cell line.20 HE significantly decreased OCI cell number at a concentration of 1 mg/mL,whereas lower concentrations were ***,this decrease was due to partial blockage of the G_1/S phase of the cell cycle,with a reduction of cells in the G_2M phase,not due to increased ***:This indicates that 20 HE significantly decreases the number of cells in the G_1/S phase of the cell cycle in human AML *** is the first time that the anti-proliferative activity of 20 HE against a human tumor cell line has been reported.

Objective:To investigate the antitumor effect of maesopsin 4-O-β-glucoside(TAT2) isolated from the leaves of Artocarpus tonkinensis(***) *** ***:The antitumor activity of TAT2 was evaluated in Lewis lung carcinoma(LLC) tumor-bearing ***/c mice had tumors induced by implantation with 2× 10~6 LLC cells into the subcutaneous right posterior ***-bearing mice were treated orally with a range of doses of TAT2 and a standard drug,*** were observed for tumor growth and mortality *** was collected to determine hematological and biochemical *** TAT2 was isolated from an ethanolic extract of *** *** structure was determined by MS and NMR spectroscopy,and identified as *** compound did not show acute toxicity at the highest dose tested(2 OIK) mg/kg body weight).TAT2 exhibited antitumor activity by decreasing tumor growth,increasing the survival rale,and ameliorating some hematological and biochemical parameters at doses of 100 and 200 mg/kg body weight(Pvailability and low toxicity,and the fact that it could be isolated in a large scale from *** leaves,the compound shows promise as a potential anticancer drug.

Systemic therapy of hepatocellular carcinoma:Current status and future perspectives

The management of hepatocellular carcinoma (HCC) has substantially changed in the past few decades, the introduction of novel therapies (such as sorafenib) have improved patient survival. Nevertheless, HCC remains the third most common cause of cancer-related deaths worldwide. Decision-making largely relies on evidence-based criteria, as showed in the US and European clinical practice guidelines, which endorse five therapeutic recommendations:resection; transplantation; radiofrequency ablation; chemoembolization; and sorafenib. Many molecularly targeted agents that inhibit angiogenesis, epidermal growth factor receptor, and mammalian target of rapamycin are at different stages of clinical development in advanced HCC. Future research should continue to unravel the mechanism of hepatocarcinogenesis and to identify key relevant molecular targets for therapeutic intervention. Identification and validation of potential surrogate and predictive biomarkers hold promise to individualize patient’s treatment to maximize clinical benefit and minimize the toxicity and cost of targeted agents.

Exposure to ambient air particulate matter and non-alcoholic fatty liver disease

The present study was designed to alert the public opinion and policy makers on the supposed enhancing effects of exposure to ambient air particulate matter with aerodynamic diameters ver disease (NAFLD), the most common chronic liver disease in Western countries. For far too long literature data have been fixated on pulmonary diseases and/or cardiovascular disease, as consequence of particulate exposure, ignoring the link between the explosion of obesity with related syndromes such as NAFLD and air pollution, the worst characteristics of nowadays civilization. In order to delineate a clear picture of this major health problem, further studies should investigate whether and at what extent cigarette smoking and exposure to ambient air PM 2.5 impact the natural history of patients with obesity-related NAFLD,i.e. , development of non alcoholic steatohepatitis, disease characterized by a worse prognosis due its progression towards fibrosis and hepatocarcinoma.

Cardioprotective effects of glucagon-like peptide 1 receptor agonists in heart failure: Myth or truth?

Therapy with glucagon-like peptide 1(GLP1)receptor agonists has raised great interest for its beneficial cardiovascular effects in preventing atherosclerosis and heart failure-related ***,while evidence about atherosclerosis consistently suggests a cardioprotective potential with class effect,controversies remain on its impact on heart ***1 receptor agonists appear to prevent hospitalization for new-onset heart failure and reduce symptoms in heart failure with preserved ejection fraction(as demonstrated by the recent STEP-HFpEF Trial).Still,GLP1 agonism has resulted in neutral or even harmful effects in patients with established heart failure with reduced ejection fraction(the LIvE trial).GLP1 receptor agonists benefit the cardiovascular system indirectly through their marked metabolic effects(improved weight management,glycemic control,blood pressure,systemic and tissue inflammation),while direct effects on the heart have been ***,weight loss alone achieved through GLP1 receptor agonists has failed in improving left ventricular *** is a dual agonist of GLP1 and glucose-dependent insulinotropic polypeptide,representing an innovative treatment option in diabetes with a major impact on weight loss and promising cardiovascular *** this class of therapies is going to change the history of heart failure is an ongoing debate.

Experimental infection with Yersinia pseudotuberculosis in European brown hare(Lepus europaeus,Pallas)

Objective:To investigate clinicopathological,bacteriological and pathological aspects of an experimental infection with Yersinia pseudotuberculosis(***)in hares in order to verify the efficacy of serology for the in vivo ***,the pathogenicity of two *** strains was investigated in order to detect potential ***:Twelve European brown hares(Lepus europaeus,Pallas)were experimentally infected per os and via conjunctival mucosae with ***:six subjects were infected with a strain isolated from a naturally infected hare(YpH)and six subjects with a strain isolated from a naturally infected rabbit(YpR).Two hares were used as negative *** animals were subjected to clinical,bacteriological and serological examinations during 9 weeks following the infection and,at the end of the control period,subjects still alive were euthanized and submitted to a complete post mortem ***:All faecal samples collected during the control period were positive for bacteriological examinations and to a PCR for the inv gene of ***,while only one Yp H-infected hare showed a positive *** the 2nd to the 9th week post infection(pi),serological analysis revealed specific antibodies with titers ranging from 1:10 to 1:160 in all YpH-infected and two YpR-infected *** the Yp H-infected and two Yp R-infected hares scored positive for *** by means of bacteriological ***,suppurative multifocal lesions were detected in liver,spleen,kidney and sub-mandibular lymph nodes in both YpH-and YpR-infected hares and confirmed with *** lesions were observed only in Yp H-infected *** confirmed the presence of bacterial antigen in all infected ***:Results of this study revealed that YpH strain is more pathogenic for hares than the YpR strain;moreover the serological test performed in
