Resveratrol modifies biliary secretion of cholephilic compounds in sham-operated and cholestatic rats

AIM To investigate the effect of resveratrol on biliary secretion of cholephilic compounds in healthy and bile duct-obstructed rats. METHODS Resveratrol(RSV) or saline were administered to rats by daily oral gavage for 28 d after sham operation or reversible bile duct obstruction(BDO). Bile was collected 24 h after the last gavage during an intravenous bolus dose of the Mdr1/Mrp2 substrate azithromycin. Bile acids,glutathione and azithromycin were measured in bile to quantify their level of biliary secretion. Liver expression of enzymes and transporters relevant for bile production and biliary secretion of major bile constituents and drugs were analyzed at the m RNA and protein levels using q RT-PCR and Western blot analysis,respectively. The TR-FRET PXR Competitive Binding Assay kit was used to determine the agonism of RSV at the pregnane X receptor. RESULTS RSV increased bile flow in sham-operated rats due to increased biliary secretion of bile acids(BA) and glutathione. This effect was accompanied by the induction of the hepatic rate-limiting transporters for bile acids and glutathione,Bsep and Mrp2,respectively. RSV also induced Cyp7 a1,an enzyme that is crucial for bile acid synthesis; Mrp4,a transporter important for BA secretion from hepatocytes to blood; and Mdr1,the major apical transporter for xenobiotics. The findings were supported by increased biliary secretion of azithromycin. The TR-FRET PXR competitive binding assay confirmed RSV as a weak agonist of the human nuclear receptor PXR,which is a transcriptional regulator of Mdr1/Mrp2. RSV demonstrated significant hepatoprotective properties against BDO-induced cirrhosis. RSV also reduced bile flow in BDO rats without any corresponding change in the levels of the transporters and enzymes involved in RSV-mediated hepatoprotection. CONCLUSION Resveratrol administration for 28 d has a distinct effect on bile flow and biliary secretion of cholephilic compounds in healthy and bile duct-obstructed rats.

We report mid-infrared Ge-on-Si waveguide-based PIN diode modulators operating at wavelengths of 3.8 and8 μm. Fabricated 1-mm-long electro-absorption devices exhibit a modulation depth of >35 dB with a 7 V forward bias at 3.8 μm, and a similar 1-mm-long Mach–Zehnder modulator has a Vπ· L of 0.47 V · cm. Driven by a 2.5 Vpp RF signal, 60 MHz on-off keying modulation was demonstrated. Electro-absorption modulation at 8 μm was demonstrated preliminarily, with the device performance limited by large contact separation and high contact resistance.

Aging is linked to the deterioration of many physical and cognitive abilities and is the leading risk factor for Alzheimer’s disease. The growing aging population is a significant healthcare problem globally that researchers must investigate to better understand the underlying aging processes. Advances in microarrays and sequencing techniques have resulted in deeper analyses of diverse essential genomes(e.g., mouse, human, and rat) and their corresponding cell types, their organ-specific transcriptomes, and the tissue involved in aging. Traditional gene controllers such as DNA-and RNA-binding proteins significantly influence such programs, causing the need to sort out long non-coding RNAs, a new class of powerful gene regulatory elements. However, their functional significance in the aging process and senescence has yet to be investigated and identified. Several recent researchers have associated the initiation and development of senescence and aging in mammals with several well-reported and novel long non-coding RNAs. In this review article, we identified and analyzed the evolving functions of long non-coding RNAs in cellular processes, including cellular senescence, aging, and age-related pathogenesis, which are the major hallmarks of long non-coding RNAs in aging.

Contamination of Pond Water and Sediment in Coal Burning Area

The pond water is used by a variety of animals i.e. mammals, birds, duck, and fish. Sediments play a significant role in determining the overall environmental quality for the living organism. Therefore, in this work, chemical characterization of pond water and sediment in the largest coal burning basin of India i.e. Korba basin is described. Elements i.e. C, O, S, F, Cl, Na, Mg, Al, Si, P, K, Ca, Ti, Fe, As, Cr, Cu, Zn, Cd, Pb, and Hg were quantified. Toxic metals i.e. As, Cd, Hg, and Pb were highly enriched in the sediments, ranging from 36 - 154, 0.14 - 1.19, 0.12 - 0.82 and 26 - 127 mg/kg with mean value (p = 0.05) of 95 ± 12, 0.62 ± 0.11, 0.35 ± 0.08 and 75 ± 13 mg/kg, respectively. The concentration variations, pollution indices and sources of elements in water and sediment are discussed.

The aim of this paper is to highlight the issue of resolving conflicts within multicultural teams in industrial enterprises. The authors build upon the concept of multiculturalism which seeks for possible ways to enable different cultures to coexist and the means of communication between them. The authors base the study on the assumption that extensively developed intercultural relationships lead to mutual understanding between people and consequently to less interpersonal conflicts. In the introduction part, the authors explain the importance of increased attention and interest in the area of multiculturalism. Industrial enterprises nowadays are increasingly aware of this issue as they become more open to different cultures and they are confronted with intensive international migration and previously isolated societies become more pluralistic. As a result of these processes, individuals are more frequently in contact with members of different cultures. ＂Think globally, act locally＂ has become a slogan for our society. This way of thinking and acting was transferred to almost all areas of life （economic, political, and educational） and it is applied in the context of schools, societies, and enterprises as well. The ability to understand and accept cultural diversity is becoming essential in the ordinary working environment. It helps to create connections between emotional perception and rational acceptance. In practice, the offer of training seminars about intercultural differences is widespread. Enterprises supporting the knowledge improvement of their own employees have understood that a friendly working environment leads to increased motivation, consequently reflected in their performance. The training focused on skills development in this area can help to reduce interpersonal misunderstandings, bring new ways of conflict resolution and harmony in the workplace and thereby also increase productivity. Compliant mechanisms transfer an input force or displacement to another point through elastic body deformation. The field of compliant mechanisms is expected to continue to grow as materials with superior properties are developed. The paper takes into consideration some typical spring guiding systems, being able to realize translating planar displacement of the link. Some types of compound spring guiding systems, such as S-shaped guiding spring system, U-shaped guiding spring system and four spring guiding system with additional rigid-body link have been considered and analyzed as compliant structures. The mobility as well as guiding accuracy of the developed compliant mechanisms, that is, their capability to realize translating planar displacement of the link, have been researched, and the optimal dimensions of the mechanism have been determined in order to provide minimal guiding inaccuracy.

Mechanical Characteristics of Chemically Degraded Surface Layers of Wood

The aim of this research was to find out whether the effect of chemical corrosion changes mechanical characteristics of surface layers of wooden construction elements. Degradation of the surface layers of wood was caused by chemical reactions of the basic substances of wood mass with compounds contained in antifire coatings. Fire retardants containing corrosive substances were often and repeatedly used in the Czech Republic on many wooden building constructions. This process of chemical corrosion is in practise called as ＂surface defibering of wood＂. This contribution presents standard and special experimental methods used for measuring the selected mechanical characteristics （compression strength, tension strength, bending strength, hardness and impact resistance） in the damaged surface layer of wooden construction elements. The material for experimental measuring was a construction element removed from a historical roof （ca 150 years old）. Mechanical characteristics of the surface layer of the defibered element were compared with the values measured in the deeper subsurface layer of non-damaged wood. The results of the experiments proved loss of cohesive strength and decrease of mechanical characteristics of wood only in a thin surface layer.

The paper aims to defme the lawfulness （model）, by which one can reliably estimate the distribution of motor units＇ force from the moment when leg extensor muscles start to generate the force until the moment when the maximal level is achieved. The study included 110 participants. To assess the contractile characteristics of leg extensors, standardized equipment and standardized isometric test in sitting position with the angle of the knee joint of 125° were used. The participants were instructed to exert their maximal force as quickly as possible. Using descriptive statistics, cluster analysis and fitting the exponential model of the distribution of force of leg extensor motor units in the whole range of force generation is defined. The model has the following form： y = 0.2051e3.3855x, where y is the motor units force expressed in daN （decanewtons）, x is the time expressed in s （seconds）. It provides an understanding of the control of multivariate motor unit recruitment and distribution of their force during sports movements as well as training programming for the adoption of forms for conlrolling force distribution of motor units, the development of their maximum force and their involvement speed.
