Computational Investigation of the Substituent Effect on the Intramolecular Proton Transfer Reaction of 3-Hydroxytropolone

The effects of substituent type and position on the proton transfer reaction of 3-hydroxytropolone(3-OHTRN) have been investigated theoretically by using density functional theory at the level of B3LYP/ 6-31+G** method. The influence of solvent on the proton transfer reactions of substituted 3-OHTRN has been examined using the self-consistent isodensity polarized continuum model(SCI-PCM) in water. As a result, while the proton transfer reaction is kinetically the easiest by substitution on position 3 of-NH2 group in the gas phase, it is kinetically the easiest by substitution on position 5 of the same group in water. In addition, these reactions are either kinetically or thermodynamically easier in the gas phase than that in water, except the reaction of structure with-NH2 group at position 6.

RNA targets of TDP-43: Which one is more important in neurodegeneration?

Nuclear loss and cytoplasmic aggregation of TDP-43 is the most common shared pathological feature of fronto-temporal dementia(FTD)and amyotrophic lateral scle-rosis(ALS),a motor neuron disease also known as Lou Gehrig’s disease[1].TDP-43 pathology is found in 97%of ALS cases,approximately 45%of FTD cases,and in some other neurodegenerative diseases such as Alzheimer’s disease[2].However,the direct mechanisms by which TDP-43 dysregulation contributes to neurodegenera-tion remain largely ***-43 is an RNA-binding protein with multiple roles in RNA metabolism,includ-ing transcription,splicing,transport,and the localization and stability of its target mRNAs,as well as in microRNA biogenesis[3].TDP-43 has thousands of RNA targets,but it is largely unknown which targets,if any,directly contribute to *** is it known whether any of them can be either manipulated as therapeutic targets to influence disease progression or,if not,could serve as biomarkers of TDP-43 pathology.

Identification of Lactic Acid Bacteria (LAB) from pre-fermented liquids of selected cereals and legumes and characterization of their exopolysaccharides (EPS)

Recently, isolation of Lactic Acid Bacteria (LAB) strains with important technological functions from plant-based products has gained special interests related with the increment of consumer preferences to the plant-based fermented products. In this context, various cereals and legumes that were underwent to germination or not were selected for the isolation of exopolysaccharide (EPS) producer LAB strains in this study. Following genotypic discrimination, 17 distinct strains belonging to Weissella confusa, Weissella cibaria, Lactococcus lactis subsp. hordniae, Leuconostoc lactis, Lactiplantibacillus plantarum and Leuconostoc mesenteroides were identified from pre-ferment liquids. Two strains Weissella confusa W-16 and Leuconostoc lactis GW-6 were the highest EPS producers, and their EPSs were further characterized. The 1H and 13C NMR results revealed that both strains were dextran producers containing (1 → 3)-linked α-d-glucose units as branches with 9.2% and 6.9% levels for Weissella confusa W-16 and Leuconostoc lactis GW-6, respectively. FTIR analysis also supported the dextran structure and demonstrated the functional groups within the dextrans. A high level of thermal stability was observed in both dextrans detected by TGA and DSC analysis. Both dextrans were in amorphous characteristics determined by XRD analysis. These findings demonstrate that pre-fermented liquids of different cereals and legumes can be source of technologically important LAB strains with EPS production characteristics.

Evaluation of nintedanib as a new postoperative antiscarring agent in experimental extraocular muscle surgery

AIM:To investigate the efficacy of nintedanib on reducing postoperative inflammation,fibrosis and adhesion formation following extraocular muscle surgery in rabbits in comparison with triamcinolone acetonide(TA).METHODS:Reinsertion of superior rectus muscle in right eyes of 30 New Zealand white rabbits were *** were randomized to receive one of the following treatments:0.9%normal saline,one of 1-,5-,and 10μmol doses of nintedanib subconjunctivally immediately after surgery and on postoperative day 1,2,3,5,and 7,and TA immediately after *** a control group,unoperated left eyes(n=6)were *** the 28 th day,six eyes from each group were enucleated and histopathologically and immunohistochemically analyzed to assess the postoperative inflammatory changes,fibrosis and *** growth factor beta,matrix metalloproteinase-2 and alpha smooth muscle actin expressions were ***:Conjunctival and scleral inflammation in TA and nintedanib groups were significantly reduced compared to saline(sham)*** vascularity and rectus muscle fibrosis were significantly reduced in 10μmol nintedanib *** groups were the most effective groups in reduction of perimuscular *** three nintedanib groups nor TA group differed statistically from sham group with regard to *** expressions of transforming growth factor beta,alpha smooth muscle actin and matrix metalloproteinase-2 were reduced in nintedanib groups compared to saline ***:Nintedanib appears to attenuate postoperative inflammation and fibrosis after extraocular muscle *** may be a safer and stronger alternative agent in extraocular muscle surgery when compared to *** investigation is needed to prove antiadhesive effect of nintedanib.

Extracellular matrix remodelling in dental pulp tissue of carious human teeth through the prism of single-cell RNA sequencing

Carious lesions are bacteria-caused destructions of the mineralised dental tissues,marked by the simultaneous activation of immune responses and regenerative events within the soft dental pulp *** major molecular players in tooth decay have been uncovered during the past years,a detailed map of the molecular and cellular landscape of the diseased pulp is still *** this study we used single-cell RNA sequencing analysis,supplemented with immunostaining,to generate a comprehensive single-cell atlas of the pulp of carious human *** data demonstrated modifications in the various cell clusters within the pulp of carious teeth,such as immune cells,mesenchymal stem cells(MSC)and fibroblasts,when compared to the pulp of healthy human *** immune response in the carious pulp tissue is accompanied by specific changes in the fibroblast and MSC *** changes include the upregulation of genes encoding extracellular matrix(ECM)components,including COL1A1 and Fibronectin(FN1),and the enrichment of the fibroblast cluster with *** incremental changes in the ECM composition of carious pulp tissues were further confirmed by immunostaining *** of the Fibronectin fibres under mechanical strain conditions showed a significant tension reduction in carious pulp tissues,compared to the healthy *** present data demonstrate molecular,cellular and biomechanical alterations in the pulp of human carious teeth,indicative of extensive ECM remodelling,reminiscent of fibrosis observed in other *** comprehensive atlas of carious human teeth can facilitate future studies of dental pathologies and enable comparative analyses across diseased organs.

Attitudes of the Host Population towards Syrian Refugees: A New Theoretical Perspective

A Study on the Evaluation of Heating and Cooling Energy Efficiency in Residential Buildings for Climatic Zones of Turkey

In Turkey, most of the common type projects of mass production residential buildings are being developed and constructed by TOK1 （Housing Development Administration of Turkey）. These buildings, in which energy efficient approach has been disregarded for years, cause to gradually increase on heating and cooling energy consumptions. In regards to national economics, it is essential to evaluate energy efficiency and to develop suggestions to decrease energy consumptions in residential buildings. To achieve appropriate solutions, cost evaluation also becomes necessary. Therefore, this paper aims to introduce a study which serves the purpose of producing a choice of energy efficient solutions in order to reduce energy consumptions and energy cost. In this study, different heating and cooling energy efficient scenarios have been developed for a selected residential building, constructed by TOKI, for climatic zones of Turkey. For each scenario, energy simulations have been executed by means of the simulation program--DesignBuilder, the user-friendly visual interface of EnergyPlus, and cost analysis has been carried out by using the net present value and discounted payback period method. As a result, energy and cost effective solutions have been presented and discussed for different climatic zones.

2020年新型冠状病毒疫情席卷了全球,此次新冠疫情的高传染性给全球各国的公共卫生都带来了前所未有的挑战。其中,医疗卫生资源配置工作作为防患化解疫情蔓延与救治病患的重要保障,成为国家层面疫情防控的重点内容。聚焦于疫情防控中卫生资源的配置状况与治理效率,各国实务部门与学术界均产生了诸多讨论。从这些实践做法与文献研究的结果可以看出,医疗卫生资源供给与配置的实际效果,将决定疫情防控的治理效能,而医疗卫生资源的实际供给过程与国家治理结构、社会运行机制与地区卫生事业管理能力均存在密切关系,但现有文献较少讨论疫情防控中借鉴他国疫情防控卫生资源配置管理的可行性问题。因此,文章以吉尔吉斯斯坦为案例切入点,通过论证3个中国典型省份的疫情防控资源配置案例,从中总结具体经验,并从吉尔吉斯斯坦的实际出发,提出可供提升吉尔吉斯斯坦疫情防控医疗卫生资源配置效率的实践举措与学理思考。本文主要针对疫情防控下的吉尔吉斯斯坦公共卫生资源配置现状进行调查分析。首先,在了解吉尔吉斯斯坦卫生资源管理现状的基础上,对其医疗卫生资源的配置进行统计分析,并梳理吉尔吉斯斯坦在疫情防控中的抗疫管理举措。其次,通过分析发现,吉尔吉斯斯坦存在医疗人力资源匮乏、医疗设施资源短缺、卫生资源配置效率低下、卫生信息资源不足等问题,以此作为吉尔吉斯斯坦学习其先进治理经验的重要途径与有效切入点。通过借鉴中国先进的疫情防控经验与治理范式,对吉尔吉斯斯坦卫生资源治理提出优化对策,即创新治理机制,优化医疗人力资源供给效率、加强协同合作,促进卫生设施资源供给充足、强化应急管理,提升医疗卫生资源配置效率、构建数字平台,加强卫生信息资源供给力度,以此推进吉尔吉斯斯坦疫情防控工作中卫生资源配置效率显著提升,促进吉尔吉斯斯坦公共卫生事业不断发展,从而为保障国家经济、科技、政治稳定发展持续赋能。
