Genetic dissection and validation of a major QTL for grain weight on chromosome 3B in bread wheat(Triticum aestivum L.)

Grain weight is one of the key components of wheat(Triticum aestivum L.)*** manipulation of grain weight is an efficient approach for improving yield potential in breeding programs.A recombinant inbred line(RIL)population derived from a cross between W7268 and Chuanyu 12(CY12)was employed to detect quantitative trait loci(QTLs)for thousand-grain weight(TGW),grain length(GL),grain width(GW),and the ratio of grain length to width(GLW)in six *** major QTLs,***-2D,***-2D,***-3B,***-4B.1,***-2D.1,***-2D.1 and ***-3B.1,were consistently identified in at least four environments and the best linear unbiased estimation(BLUE)datasets,and they explained 2.61 to 34.85%of the phenotypic *** interactions were detected between the two major TGW QTLs and three major GW *** addition,***-3B.1 and ***-3B were co-located,and the improved TGW at this locus was contributed by *** other loci,***-3B.1/***-3B had no effect on grain number per spike(GNS).They were further validated in advanced lines using Kompetitive Allele Specific PCR(KASP)markers,and a comparison analysis indicated that ***-3B.1/***-3B is likely a novel *** haplotypes were identified in the region of this QTL and their distribution frequencies varied between the landraces and *** to gene annotation,spatial expression patterns,ortholog analysis and sequence variation,the candidate gene of ***-3B.1/***-3B was ***,the major QTLs and KASP markers reported here provide valuable information for elucidating the genetic architecture of grain weight and for molecular marker-assisted breeding in grain yield improvement.

2021年8月在贝加尔针茅草甸草原刈割、自由放牧与围封(对照)样地进行土壤线虫采集,探究草地不同利用方式对土壤线虫群落结构特征及代谢足迹的影响.本研究共捕获土壤线虫15632条,隶属于55属,群落优势属为鹿角唇属(Cervidellus),垫咽属(Tylencholaimus)和丝尾垫刃属(Filenchus),分别占总捕获量的7.05%,7.14%和6.18%.较围封生境,线虫总多度在自由放牧生境显著降低.从功能类群来看,3种生境下植食性线虫(33.84%~37.45%)与c-p 2类群(38.78%~47.58%)相对多度占比最大,且均在刈割生境与自由放牧生境显著增加.从多样性与生态指数来看,相较于围封,自由放牧生境成熟度指数(MI)与富集指数(EI)显著下降,基础指数(BI)显著增加,表明在自由放牧生境线虫群落多样性及稳定性降低,而食物网抗干扰能力增强;刈割生境结构指数(SI)显著提高,刈割生境食物网连通性提高,食物链加长.且在不同草地利用方式下线虫通路比值(NCR)>0.5瓦斯乐思卡指数(WI)>1,说明草甸草原土壤矿化途径的主要承担者是食微线虫,有机质分解以细菌通道为主.从代谢足迹来看,刈割和自由放牧生境均降低了线虫复合代谢足迹(TNf)和功能代谢足迹,食物网处于结构化状态,围封生境显著增加了线虫群落的整体活性和碳流动,食物网处于较为稳定的代谢平衡状态.不同草地利用方式下地下生物量(BGB),植物个体数(P-N)和速效钾(AK)是影响线虫群落发展的主要环境因子.综上所述,土壤线虫群落结构在不同草地利用方式下差异显著,不同生境中环境因子的变化是驱动土壤线虫群落变化的主要因子.

Calculating the Relativistic Wave Functions of 11S0 and 13S1 Positronium

Based cn the relativistic Bethe-Salpeter (BS) equation, the positronium wavefunctions in Euclidean momentum space are obtained. Meanwhile the energy levels of positronium ground states 11S0 and 13S1 are fitted to be 6.7934 eV and 6.7929 eV respectively, which qualitatively agree with the previous theoretical values. It is shown that the BS theory is valid and reliable to treat positronium.

The aim of this study was to isolate the full-length cDNA sequences of an unusually highly conserved olfactory receptor （orthologue to the Drosophila melanogaster DOR83b） from the antennae of Spodoptera exigua and S. litura by using reverse transcription-polymerase chain reaction （RT-PCR） and rapid amplification of cDNA ends （RACE） methods. Bioinformatics methods were used to further analyze the cDNA sequences and putative amino acid sequences. The two full-length cDNA sequences from olfactory receptor （OR） of male S. exigua and S. litura were named as SexiOR2 and SlitOR2, respectively. SexiOR2 and SlitOR2 consisted of nucleotide sequence of 1 906 and 2 483 bp, respectively, and both with deduced amino acid sequences of 473 residues. The sequence analysis indicated that the deduced amino acid sequences of the cDNA shared the high homologies with OR83b orthologue chemoreceptor sequences from previously reported moths, implying that the cDNA sequences were of OR83b orthologue chemoreceptor genes.

为探索低分子量有机酸对亚热带地区典型土壤磷吸附的影响,采用批处理法研究了柠檬酸和草酸对旱地土、水稻土磷素吸附特征的作用,重点探讨了不同浓度有机酸和柠檬酸钠、有机酸加入顺序等对土壤磷吸附的影响。结果表明:柠檬酸和草酸显著降低了土壤磷吸附动力学的颗粒内扩散模型拟合度,减弱了磷素在土壤颗粒内部扩散过程。Langmuir和Freundlich方程对有机酸存在下土壤磷吸附等温数据均有较好的拟合效果(R2=0.863~0.996)。有机酸降低了土壤磷素最大理论吸附量。土壤磷吸附量与有机酸浓度呈指数函数关系,0.1~5 mmol/L有机酸对土壤磷吸附的影响最大。有机酸浓度小于1 mmol/L时,柠檬酸对土壤磷素吸附的抑制程度大于草酸;而当浓度大于5 mmol/L时,草酸的抑制程度更大。在相同浓度时,0.1~5 mmol/L的柠檬酸钠比柠檬酸对土壤磷吸附能力抑制程度更大,先加入有机酸进一步降低了土壤对磷素吸附量,说明阴离子竞争吸附是低浓度柠檬酸降低土壤磷吸附能力的主要原因,而柠檬酸浓度为5~50 mmol/L时,有机酸与土壤铁铝等金属离子的络合作用占主导。

Responses of plant diversity and primary productivity to nutrient addition in a Stipa baicalensis grassland, China

Nutrient addition can affect the structure and diversity of grassland plant communities, thus alter the grassland productivity. Studies on grassland plant community composition, structure and diversity in response to nutrient addition have an important theoretical and practical significance for the scientific management of grassland, protection of plant diversity and the recovery of degraded grassland. A randomized block design experiment was conducted with six blocks of eight treatments each： control（no nutrient addition） and K, P, N, PK, NK, NP, and NPK addition. We evaluated plant composition, height, coverage, density, and aboveground biomass to estimate primary productivity and plant diversity. Results showed that all treatments increased primary productivity significantly（P〈0.05） with the exception of the K and the NPK treatments had the greatest effect, increasing aboveground biomass 2.46 times compared with the control（P〈0.05）. One-way ANOVA and factorial analysis were used for the species richness, Shannon-Wiener index, Pielou index and aboveground biomass, and the relationships between the diversity indices and aboveground biomass were determined through linear regression. We found that fertilization altered the community structure; N（but not P or K） addition increased the proportion of perennial rhizome grasses and significantly reduced that of perennial forbs（P〈0.05）, thus it presented a trend of decrease in species richness, Shannon-Wiener and Pielou indexex, respectively. Only the main effects of N had significant impacts on both the diversity indices and the aboveground biomass（P〈0.05）, and the interactions between N-P, N-K, P-K and N-P-K could be neglected. With fertilization, plant diversity（correlation coefficient, –0.61）, species richness（–0.49）, and species evenness（–0.51） were all negatively linearly correlated with primary productivity. 糖尿病足是糖尿病最常见的并发症之一,该症状会使足部神经、血管病变,进而导致患者足底压力异常。为了预防糖尿病足,根据糖尿病足的主要特点,选用延展性很好的柔性传感器阵列,基于STC8H8K64U单片机设计了一个传感器密度高、传输速度快、精度高的足底压力监测系统。使用系统采集了600组足底压力数据,选用200 kPa作为压力异常阈值,计算了足底各个区域的异常点占比,并通过t检验得出了正常人左右脚压力异常点占比的范围。最后将压力数据以压力云图的形式在系统软件上显示出来,并显示各个区域的压力异常点占比及其是否处于正常区间内,为糖尿病足的监测预防提供了可靠性高的测试平台。
