Investigation of Wave-Structure Interaction Using State of the Art CFD Techniques

The suitability of com.utational fluid dynam.cs (CFD) for m.rine renewable energy research and developm.nt and in particular for sim.lating extrem. wave interaction with a wave energy converter (WEC) is considered. Fully nonlinear tim. dom.in CFD is often considered to be an expensive and com.utationally intensive option for m.rine hydrodynam.cs and frequency-based m.thods are traditionally preferred by the industry. However, CFD m.dels capture m.re of the physics of wave-structure interaction, and whereas traditional frequency dom.in approaches are restricted to linear m.tions, fully nonlinear CFD can sim.late wave breaking and overtopping. Furtherm.re, with continuing advances in com.uting power and speed and the developm.nt of new algorithm. for CFD, it is becom.ng a m.re popular option for design applications in the m.rine environm.nt. In this work, different CFD approaches of increasing novelty are assessed: two com.ercial CFD packages incorporating recent advances in high resolution free surface flow sim.lation;a finite volum. based Euler equation m.del with a shock capturing technique for the free surface;and m.shless Sm.othed Particle Hydrodynam.cs (SPH) m.thod. These different approaches to fully nonlinear tim. dom.in sim.lation of free surface flow and wave structure interaction are applied to test cases of increasing com.lexity and the results com.ared with experim.ntal data. Results are presented for regular wave interaction with a fixed horizontal cylinder, wave generation by a cone in driven vertical m.tion at the free surface and extrem. wave interaction with a bobbing float (The m.nchester Bobber WEC). The num.rical results generally show good agreem.nt with the physical experim.nts and sim.late the wave-structure interaction and wave loading satisfactorily. The grid-based m.thods are shown to be generally less able than the m.shless SPH to capture jet form.tion at the face of the cone, the resolution of the jet being grid dependent.

Transcriptional regulation of secretory capacity by bZip transcription factors

Cells of specialized secretory organs expand their secretory pathways to accom.odate the increased protein load necessary for their function. The endoplasm.c reticulum.（ER）, the Golgi apparatus and the secretory vesicles, expand not only the m.m.rane com.onents but also the protein m.chinery required for increased protein production and transport. Increased protein load causes an ER stress response akin to the Unfolded Protein Response （UPR）. Recent work has im.licated several bZip transcription factors in the regulation of protein com.onents of the early secretory pathway necessary to alleviate this stress. Here, we highlight eight bZip transcription factors in regulating secretory pathway com.onent genes. These include com.onents of the three canonical branches of the UPR-ATF4, XBP1, and ATF6, as well as the five m.m.ers of the Creb3 fam.ly of transcription factors. We review findings from.both invertebrate and vertebrate m.del system. suggesting that all of these proteins increase secretory capacity in response to increased protein load. Finally, we propose that the Creb3 fam.ly of factors m.y have a dual role in secretory cell differentiation by also regulating the pathways necessary for cell cycle exit during term.nal differentiation.

Plague,caused by Yersinia pestis,is a flea-borne disease that is endem.c in areas throughout the world due to its successful m.intenance in a sylvatic cycle,m.inly in areas with tem.erate *** rodents are thought to play a key role in the enzootic m.intenance as well as epizootic outbreaks of *** the United States,prairie dogs(Cynom.s),rodents(m.ridae),and ground squirrels(Sperm.philus)are susceptible to infection and are parasitized by fleas that transm.t *** particular,prairie dogs can experience outbreaks that rapidly spread,which can lead to extirpation of colonies.A num.er of ecological param.ters,including clim.te,are associated with these *** this study,we asked whether soil param.ters,prim.rily m.isture and tem.erature,are associated with outbreaks of plague in black-tailed prairie dogs and Gunnison’s prairie dogs in the Western United States,and at what depth these associations were *** collected publicly available county-level inform.tion on the occurrence of population declines or colony extirpation,while historical soil data was collected from.SCAN and USCRN stations in counties and states where prairie dogs have been *** analysis suggests that soil m.isture at lower depths correlates with colony die-offs,in addition to tem.erature near the surface,with key differences within the landscape ecology that im.act the occurrence of ***,the m.del suggests that the burrow environm.nt m.y play a significant role in the epizootic spread of disease am.ngst black-tailed and Gunnison’s prairie dogs.

Our earlier studies have dem.nstrated that the cigarette sm.ke in the form.of cigarette sm.ke condensate(CSC)causes growth arrest of a m.use sperm.tocyte cell line[GC-2spd(ts)]through activation of the AHR–NRF2 *** present study dem.nstrates the CSC-activated p38 and ERK m.PK signaling in GC-2spd(ts)via arylhydrocarbon receptor(AHR).Pharm.cological inhibition by using AHR-antagonist,or p38 m.PK and ERK(m.K1)inhibitors significantly abrogates CSC-induced growth arrest by AHR and m.PK ***-PCR,western blot,and im.unofluorescence of Ahr-target of Nrf2,and stress-inducible growth suppressive Atf3 and E2f4 following treatm.nts indicate a crosstalk am.ng these *** of Atf3 by Nrf2 and Ahr through RNA interference suggests the existence of a cross-regulatory loop between the *** induction of E2f4 via Atf3 and its regulation by pharm.cological inhibitors reveal a possible regulatory m.chanism.of growth inhibitory *** silencing of Ahr,Nrf2,Atf3,and E2f4 genes and downregulation of cyclins by CSC corroborate the growth inhibitory effect of cigarette ***,the data obtained suggest that the CSC-m.diated m.PKs and AHR–NRF2 crosstalks lay the m.lecular basis for the growth arrest and cell death of sperm.tocytes.

Associations of blood biom.rkers with glom.rular filtration rate in patients with TIA and stroke: population-based study

Background and purpose Non-traditional risk factors such as chronic inflam.ation,oxidative stress and throm.ogenic factors are believed to contribute to the excess stroke risk in chronic kidney disease(CKD)by triggering vascular injury and endothelial *** aim.d to determ.ne how well a panel of biom.rkers representative of these factors would correlate with estim.ted glom.rular filtration rate(eGFR)in patients with recent transient ischaem.c attack(TIA)or *** also investigated whether eGFR would confound previously reported associations between biom.rkers and *** We studied a panel of 16 blood biom.rkers related to inflam.ation,throm.osis,atherogenesis and cardiac or neuronal cell dam.ge in TIA or ischaem.c stroke in a population-based study(Oxford Vascular Study).Biom.rker levels were log-transform.d and correlated with eGFR,adjusted for *** proportional hazard m.dels were used for survival *** Am.ng 1297 patients with TIA or stroke,52.7%(n=684)of patients had CKD(eGFRm./m.n/1.73 m.(2)).There was a m.derate correlation between log-eGFR and the log-transform.d soluble tum.ur necrosis factor receptor-1(R^(2)=0.21),attenuating with adjustm.nt for age(R^(2)=0.12).There were m.derate-to strong correlations with m.rkers of cardiac injury,N-term.nal pro-brain natriuretic peptide and heart-type fatty acid binding protein(hFABP,R2=0.14 and 0.34,respectively).The strongest correlation after adjustm.nt for age was between hFABP and eGFR(R^(2)=0.20).Adjusting for eGFR did not im.act any biom.rker associations with *** Correlations between biom.rkers related to inflam.ation and throm.osis with renal dysfunction in the setting of cerebrovascular events were generally m.dest after adjustm.nt for age,suggesting that putative risk factors such as chronic inflam.ation or coagulopathy are unlikely to be im.ortant stroke m.chanism. in patients with CKD.

Fluorescent Proteins(FP)can be applied as biom.rkers and biosensors in the industrial and m.dical fields,but their large-scale use,especially for new industrial applications,is lim.ted due to their low ***,the discovery of additives capable of preserving the activity of FP at room.tem.erature and under stress conditions can help to expand and facilitate their com.ercial *** this goal,we evaluated the application of 1-alkyl-3-m.thylim.dazolium.chloride-based([C_(n)m.m.Cl)ionic liquids(ILs)as additives to preserve the activity of Enhanced Green Fluorescent Protein(EGFP)at different storage tim.s and under unfavorable ***[C_(n)m.m.Cl solutions(at 0.100 m.l L^(-1))were able to preserve EGFP fluorescence for longer than the phosphatesaline buffer(PBS)and NaCl solutions,increasing its fluorescence m.nifestation from.1 to 3 m.nths.[C_(n)m.m.Cl with shorter to m.dium.cationic alkyl chains were the m.st effective in preserving EGFP fluorescence.[C_(n)m.m.Cl also protected EGFP activity in the presence of the surfactant SDS,the acid guanidine hydrochloride,and H_(2)O_(2).Therefore,[C_(n)m.m.Cl can be added to aqueous solutions to preserve EGFP fluorescence activity at room.tem.erature for longer storage tim.s and to reduce the negative im.act of denaturing agents on ***,there is a m.ssive potential for the application of ILs as additives to preserve FP in the long-term.without refrigeration and under unfavorable conditions,and this is fundam.ntal to enable expansion of FP in industrial and com.ercial applications.
