Interrelationship of soil moisture and temperature to sylvatic plague cycle among prairie dogs in the Western United States

Plague,caused by Yersinia pestis,is a flea-borne disease that is endemic in areas throughout the world due to its successful maintenance in a sylvatic cycle,mainly in areas with temperate *** rodents are thought to play a key role in the enzootic maintenance as well as epizootic outbreaks of *** the United States,prairie dogs(Cynomys),rodents(muridae),and ground squirrels(Spermophilus)are susceptible to infection and are parasitized by fleas that transmit *** particular,prairie dogs can experience outbreaks that rapidly spread,which can lead to extirpation of colonies.A number of ecological parameters,including climate,are associated with these *** this study,we asked whether soil parameters,primarily moisture and temperature,are associated with outbreaks of plague in black-tailed prairie dogs and Gunnison’s prairie dogs in the Western United States,and at what depth these associations were *** collected publicly available county-level information on the occurrence of population declines or colony extirpation,while historical soil data was collected from SCAN and USCRN stations in counties and states where prairie dogs have been *** analysis suggests that soil moisture at lower depths correlates with colony die-offs,in addition to temperature near the surface,with key differences within the landscape ecology that impact the occurrence of ***,the model suggests that the burrow environment may play a significant role in the epizootic spread of disease amongst black-tailed and Gunnison’s prairie dogs.

Characterising populations living close to intensive farming and composting facilities in England

Pneumocystis jiroveci pneumonia and pneumomediastinum in an anti-TNFαnaive patient with ulcerative colitis

We report the case of a 21-year-old man who was noted to have pneumomediastinum during an admission for an acute flare of ulcerative colitis. At that time, he was on maintenance treatment with azathioprine at a dose of 2.25 mg/kg per day, and had not received supplementary steroids for 9 mo. He had never received anti-tumor necrosis factor （TNF）α therapy. Shortly after apparently effective treatment with intravenous steroids and an increased dose of azathioprine, he developed worsening colitic and new respiratory symptoms, and was diagnosed with Pneumocystis jiroveci （carinii） pneumonia （PCP）. Pneumomediastinum is rare in immunocompetent hosts, but is a recognized complication of PCP in human immunodeficiency virus （HIV） patients, although our patient＇s HIV test was negative. Treatment of PCP with co-trimoxazole resulted in resolution of both respiratory and gastrointestinal symptoms, without the need to increase the steroid dose. There is increasing vigilance for opportunistic infections in patients with inflammatory bowel disease following the advent of anti-TNFα therapy. This case emphasizes the importance of considering the possibility of such infections in all patients with inflammatory bowel disease, irrespective of the immunosuppressants they receive, and highlights the potential of steroid-responsive opportunistic infections to mimic worsening colitic symptoms in patients with ulcerative colitis.

Transcriptional regulation of secretory capacity by bZip transcription factors

Cells of specialized secretory organs expand their secretory pathways to accommodate the increased protein load necessary for their function. The endoplasmic reticulum （ER）, the Golgi apparatus and the secretory vesicles, expand not only the membrane components but also the protein machinery required for increased protein production and transport. Increased protein load causes an ER stress response akin to the Unfolded Protein Response （UPR）. Recent work has implicated several bZip transcription factors in the regulation of protein components of the early secretory pathway necessary to alleviate this stress. Here, we highlight eight bZip transcription factors in regulating secretory pathway component genes. These include components of the three canonical branches of the UPR-ATF4, XBP1, and ATF6, as well as the five members of the Creb3 family of transcription factors. We review findings from both invertebrate and vertebrate model systems suggesting that all of these proteins increase secretory capacity in response to increased protein load. Finally, we propose that the Creb3 family of factors may have a dual role in secretory cell differentiation by also regulating the pathways necessary for cell cycle exit during terminal differentiation.

Cigarette smoke-induced cell cycle arrest in spermatocytes [GC-2spd(ts)] is mediated through crosstalk between Ahr–Nrf2 pathway and mAPK signaling

Our earlier studies have demonstrated that the cigarette smoke in the form of cigarette smoke condensate(CSC)causes growth arrest of a mouse spermatocyte cell line[GC-2spd(ts)]through activation of the AHR–NRF2 *** present study demonstrates the CSC-activated p38 and ERK mAPK signaling in GC-2spd(ts)via arylhydrocarbon receptor(AHR).Pharmacological inhibition by using AHR-antagonist,or p38 mAPK and ERK(mEK1)inhibitors significantly abrogates CSC-induced growth arrest by AHR and mAPK ***-PCR,western blot,and immunofluorescence of Ahr-target of Nrf2,and stress-inducible growth suppressive Atf3 and E2f4 following treatments indicate a crosstalk among these *** of Atf3 by Nrf2 and Ahr through RNA interference suggests the existence of a cross-regulatory loop between the *** induction of E2f4 via Atf3 and its regulation by pharmacological inhibitors reveal a possible regulatory mechanism of growth inhibitory *** silencing of Ahr,Nrf2,Atf3,and E2f4 genes and downregulation of cyclins by CSC corroborate the growth inhibitory effect of cigarette ***,the data obtained suggest that the CSC-mediated mAPKs and AHR–NRF2 crosstalks lay the molecular basis for the growth arrest and cell death of spermatocytes.

Associations of blood biomarkers with glomerular filtration rate in patients with TIA and stroke: population-based study

Background and purpose Non-traditional risk factors such as chronic inflammation,oxidative stress and thrombogenic factors are believed to contribute to the excess stroke risk in chronic kidney disease(CKD)by triggering vascular injury and endothelial *** aimed to determine how well a panel of biomarkers representative of these factors would correlate with estimated glomerular filtration rate(eGFR)in patients with recent transient ischaemic attack(TIA)or *** also investigated whether eGFR would confound previously reported associations between biomarkers and *** We studied a panel of 16 blood biomarkers related to inflammation,thrombosis,atherogenesis and cardiac or neuronal cell damage in TIA or ischaemic stroke in a population-based study(Oxford Vascular Study).Biomarker levels were log-transformed and correlated with eGFR,adjusted for *** proportional hazard models were used for survival *** Among 1297 patients with TIA or stroke,52.7%(n=684)of patients had CKD(eGFRmL/min/1.73 m^(2)).There was a moderate correlation between log-eGFR and the log-transformed soluble tumour necrosis factor receptor-1(R^(2)=0.21),attenuating with adjustment for age(R^(2)=0.12).There were moderate-to strong correlations with markers of cardiac injury,N-terminal pro-brain natriuretic peptide and heart-type fatty acid binding protein(hFABP,R2=0.14 and 0.34,respectively).The strongest correlation after adjustment for age was between hFABP and eGFR(R^(2)=0.20).Adjusting for eGFR did not impact any biomarker associations with *** Correlations between biomarkers related to inflammation and thrombosis with renal dysfunction in the setting of cerebrovascular events were generally modest after adjustment for age,suggesting that putative risk factors such as chronic inflammation or coagulopathy are unlikely to be important stroke mechanisms in patients with CKD.
