Mitochondrial dNA T7719G in tRNA-Lys gene affects litter size in Small-tailed Han sheep

Background： In farm animals, mitochondrial dNA（mtdNA） effect on economic performance remains hot-topic for breeding and genetic selection. Here, 53 maternal lineages of Small-tailed Han sheep were used to investigate the association of mitochondrial dNA variations and the lambing litter ***： Sequence sweeping of the mitochondrial coding regions discovered 31 non-synonymous mutations, and the association study revealed that T7719G in mt dNA t RNA-Lys gene was associated with litter size（P 〈 0.05）,manifesting 0.29 lambs per litter between the G and T carriers. Furthermore, using the mixed linear model, we assayed the potential association of the ovine litter size and haplogroups and multiple-level mtdNA haplotypes,including general haplotypes, assembled haplotypes of electron transport chain contained sequences（H-ETC）,mitochondrial respiratory complex contained sequences（H-MRC） and mitochondrial genes（H-gene, including polypeptide-coding genes, rRNA genes and tRNA genes）. The strategy for assembled mitochondrial haplotypes was proposed for the first time in mtdNA association analyses on economic traits, although none of the significant relations could be concluded（P 〉 0.05）. In addition, the nuclear major gene BMPR1B was significantly correlated with litter size in the flock（P 〈 0.05）, however, did not interact with mtdNA T7719G mutation（P 〉 0.05）.Conclusions： Our results highlight mutations of ovine mitochondrial coding genes, suggesting T7719G in tRNA-Lys gene be a potentially useful marker for selection of sheep litter size.

The presence of Total coliform, Eschericha coli and enterococci were enumerated in the cattle farming areas of the Oconee Watershed using colilertTM and enterolertTM IdEXX plates, respectively. Microbial Source Tracking (MST) using Bacteroidales molecular markers for ruminant (RuBac) and human (HuBac) specific bacterial groups were used to determine the source of the fecal pollution in the watershed. In the cattle farming regions of the watershed higher levels of fecal bacteria were detected compared to the levels of fecal bacteria at the forested and residential sites. MST indicated that the cattle farming regions (except dC2) of the lake was impacted by fecal pollution from a ruminant source such as cattle. In addition, qPCR for the tuf gene of E. coli and the Stx2 gene that is commonly found in enterohemorragic E. coli O157:H7 were used to evaluate the presence of these bacteria in the study area. E. coli O157:H7 (Stx2 gene) was detected only in the beef cattle regions of the watershed. The presences of E. coli and Stx2 gene in the Oconee Watershed represent a potential public health risk because Lake Oconee and its tributaries are used for recreational activities as well as crop irrigation.

Photosynthetic Water Use Efficiency of Heritage and Modern Potatoes under Limited and Unlimited Water Environments

Photosynthetic capacity for heritage (Taewa) and modern potato cultivars were compared at different water and nitrogenregimes in the glasshouse and field. The glasshouse was 2*2*4 factorial design with two irrigation: 100% ET and 60% ET;two applied N: 50 kg N ha-1 and 200 kg N ha-1, two Taewa (Moe Moe, Tutaekuri) and two modern potatoes (Moonlight, Agria). The 2009/2010 field experiment was a split-plot, with irrigation and rain-fed regimes as the main treatments: four potatoes above were sub-treatments. The 2010/2011 field experiment was a split-split-plot, with three water regimes as the main treatments: three cultivars (Moe Moe, Tutaekuri, and Agria) were subplots;two N rates were sub-sub-treatments. Gaseous exchange was measured by CIRAS-2 at different days from emergence. Leaf water potential was measured using pressure chamber method. Taewa achieved high photosynthetic WUE in glasshouse and 2010/2011 experiment by maintaining high An, low gs and low Ci compared to modern cultivars (p The An, gs and T increased with irrigation and N increase while decreasing Ci (p d VPd resulting in low An and photosynthetic WUE in Moonlight in the glasshouse. The leaf water potential for Taewa was very tolerant while modern potatoes were weakened by water stress. The study indicated that Taewa can be scheduled at partial irrigation without more detrimental effects on photosynthetic capacity while modern potatoes need full irrigation to avoid detrimental effects on photosynthetic capacity.

dear Sir.With interest we have read the article of Ozbek et al. [ 1 ] regarding a possible association of the 5-HTTLPR gene polymorphism and lifelong premature ejaculation （PE） in Turkish men. However, by performing a reanalysis of their data in Table 1, we found major insufficiencies that seriously diminish the validity of their conclusion that there is a higher prevalence of SS genotype in men with lifelong PE. Apart from the fact that in Table 1, the total number of patients and controls should be 69 instead of 70, our reanalysis of their patient data showed that the observed number of genotypes in their patients is not in Hardy Weinberg equilibrium, as the authors have erroneously suggested.
