Mitochondrial DNA T7719G in tRNA-Lys gene affects litter size in Small-tailed han sheep

Background： In farm animals, mitochondrial DNA（mtDNA） effect on economic performance remains hot-topic for breeding and genetic selection. here, 53 maternal lineages of Small-tailed han sheep were used to investigate the association of mitochondrial DNA variations and the lambing litter ***： Sequence sweeping of the mitochondrial coding regions discovered 31 non-synonymous mutations, and the association study revealed that T7719G in mt DNA t RNA-Lys gene was associated with litter size（P 〈 0.05）,manifesting 0.29 lambs per litter between the G and T carriers. Furthermore, using the mixed linear model, we assayed the potential association of the ovine litter size and haplogroups and multiple-level mtDNA haplotypes,including general haplotypes, assembled haplotypes of electron transport chain contained sequences（h-ETC）,mitochondrial respiratory complex contained sequences（h-MRC） and mitochondrial genes（h-gene, including polypeptide-coding genes, rRNA genes and tRNA genes）. The strategy for assembled mitochondrial haplotypes was proposed for the first time in mtDNA association analyses on economic traits, although none of the significant relations could be concluded（P 〉 0.05）. In addition, the nuclear major gene BMPR1B was significantly correlated with litter size in the flock（P 〈 0.05）, however, did not interact with mtDNA T7719G mutation（P 〉 0.05）.Conclusions： our results highlight mutations of ovine mitochondrial coding genes, suggesting T7719G in tRNA-Lys gene be a potentially useful marker for selection of sheep litter size.

Temporal and elevation trends in rainfall erosivity on a 149 km2watershed in a semi-arid region of the American Southwest

Changes in sleep parameters following biomimetic oral appliance therapy: A case report

BACKGRoUND Mandibular advancement devices(MADs)are used to treat mild to moderate obstructive sleep apnea(oSA),but there is a risk that the underlying condition can worsen in the ***,this case report is based on biomimetic oral appliance therapy as an alternative to MADs,which was found to be beneficial in the treatment of a case with severe *** SUMMARY An overnight sleep study was undertaken in a 50-year-old male with excessive daytime sleepiness that lead to a diagnosis of severe oSA as the apnea-hypopnea index(AhI)was found to be 32.8/*** the patient was unable to comply with continuous positive airway pressure therapy and declined surgical intervention,treatment with a MAD was *** 10 years later,another sleep study was performed with no MAD in the mouth,which revealed an AhI of 67.9/*** view of the deterioration in sleep quality,the patient sought alternative treatment and elected on biomimetic oral appliance therapy,using a mandibular repositioning nighttime appliance(mRNA appliance®,Vivos Therapeutics,Inc.,United States).After 10 mo,another sleep study was performed with no device in the patient’s mouth,which revealed an AhI of 11.8/h,a mean oxygen saturation of 94%and a mean oxygen desaturation index of 5.3%while *** analysis of the pre-and post-treatment study models of the upper jaw showed localized size increases of 15%-17%in the premolar regions and 15%-23%in the molar *** In adults with severe oSA that are unable to accept continuous positive airway pressure or surgical treatment,biomimetic oral appliance therapy may be preferable over MADs since biomimetic oral appliance therapy may be able to prevent worsening of sleep parameters by remodeling the nasomaxillary ***-term follow up studies are required to verify these novel findings.

Fluidization and bubbling behavior of potash particles in a deep fluidized bed

Most existing models for predicting bubble size and bubble frequency have been developed for freely bubbling fluidized *** prediction of bubbling behavior in deep fluidized beds,however,has been a challenge due to the higher degree of bubble coalescence and break up,high probability of the slugging regime,partial fluidization,and chaotic behavior in the bubbling *** this work,the bubbling and fluidization behavior of potash particles was investigated in a deep fluidized bed employing a twin-plane electrical capacitance tomography(ECT)*** volume fraction,average bubble velocity,average bubble diameter,and bubble frequency in both bubbling and slugging regimes were measured at two different bed height ratios(h/D=3.5 and h/D=3.78).This work is the first to illustrate a sequential view of bubbles at different superficial gas velocities in a fluidized *** results show that both the bubble diameter and rising velocity increased with increasing the superficial gas velocity for the two bed heights,with larger values observed in the deeper bed compared to the shallower *** values for bubble diameter,bubble rise velocity and bubble frequency from different models are compared with the experimental data obtained from the ECT system in this *** agreement has been achieved between the values predicted by the previous models and the experimental data for the bubble diameter and bubble rise velocity with an average absolute deviation of 16%and 15%for the bed height of 49 cm and 13%and 8%for the bed height of 53 cm,respectively.

Design and evaluation of aceclofenac ocular inserts with special reference to cataracts and conjunctivitis

The eyes present unique opportunities and challenges when it comes to the delivery of *** absorption by this route is bungling,there are a few side effects with conventional dosage *** inserts were prepared with prolonged release of drug and minimum swelling within cul-de-sac using *** work focused on treatment of conjunctivitis and cataracts by formulating ocular inserts of different polymeric combination of aceclofenac using hydroxypropyl methyl cellulose(hPMC,3% to 5%),chitosan(3% to 5%),poly vinyle alcohol(PVA,3% to 5%),methyl cellulose(MC,3% to 5%) as drug reservoir and ethyl cellulose(EC) polymer as rate controlling membrane by solvent casting technique with the objective of increasing contact time,achieving controlled release and greater therapeutic *** prepared ocular insert were then evaluated for physical appearances tensile strength,elongation at break(%),weight variation,uniformity of thickness,moisture absorption(%),ph,folding endurance,Fourier Transform Infrared spectroscopy,differential scanning *** characterization and in vitro transcorneal permeation studies reveals that,the prepared ocular insert formulations F2 and F8 containing hPMC and PVA had released their drug content,98.54% and 96.24%,respectively,over an extended period of 24 *** these formulations were selected as best optimized *** can be concluded that hydroxy propyl methyl cellulose is a good film forming hydrophilic polymer which shows potential agent for ocular drug delivery *** of polyethylene glycol enhances the permeability of aceclofenac ocular insert and has perfect zero order release,proving a promising controlled release delivery system.

Lipid-polymer hybrid nanoparticles: Development & statistical optimization of norfloxacin for topical drug delivery system

Poly lactic acid is a biodegradable, biocompatible, and non-toxic polymer, widely used in many pharmaceuticalpreparations such as controlled release formulations, parenteral preparations, surgicaltreatment applications, and tissue engineering. In this study, we prepared lipid-polymer hybrid nanoparticlesfor topical and site targeting delivery of Norfloxacin by emulsification solvent evaporationmethod (ESE). The design of experiment (DoE) was done by using software to optimize the result, andthen a surface plot was generated to compare with the practical results. The surface morphology, particlesize, zeta potential and composition of the lipid-polymer hybrid nanoparticles were characterized bySEM, TEM, AFM, and FTIR. The thermal behavior of the lipid-polymer hybrid nanoparticles was characterizedby DSC and TGA. The prepared lipid-polymer hybrid nanoparticles of Norfloxacin exhibited anaverage particle size from 178.6 ± 3.7 nm to 220.8 ± 2.3 nm, and showed very narrow distribution withpolydispersity index ranging from 0.206 ± 0.36 to 0.383 ± 0.66. The surface charge on the lipid-polymerhybrid nanoparticles were confirmed by zeta potential, showed the value from t23.4 ± 1.5 mVto t41.5 ± 3.4 mV. An Antimicrobial study was done against Staphylococcus aureus and Pseudomonasaeruginosa, and the lipid-polymer hybrid nanoparticles showed potential activity against these ***-polymer hybrid nanoparticles of Norfloxacin showed the %cumulative drug release of 89.72% in24 h. A stability study of the optimized formulation showed the suitable condition for the storage oflipid-polymer hybrid nanoparticles was at 4 ± 2 C/60 ± 5% Rh. These results illustrated high potential oflipid-polymer hybrid nanoparticles Norfloxacin for usage as a topical antibiotic drug carriers.

Gastrointestinal Stromal Tumor Mimicking ovarian Tumor

Background: Gastrointestinal stromal tumors (GIST) are rare mesenchymal tumors that may mimic ovarian tumor on presurgical testing. They are most often located in the stomach and proximal small intestine. The gross appearance of GISTs is highly variable. These tumors are usually asymptomatic, often discovered accidentally during endoscopic or radiologic examinations. Case Report: We reported a case of 55-year-old post menopausal woman who presented to us as ovarian mass which turned out to be malignant epithelioid gastrointestinal stromal tumor intraoperatively and by immunohistochemistry. Conclusion: In the patients with a pelvic mass, especially if unusual symptoms and laboratory data being not compatible with gynecological disease, the possibility of diseases other than a gynecologic disease has to be considered. Considering that only few patients with gastrointestinal stromal tumors have been reported in the obstetrical and gynecological literature, the awareness of such an entity by the obstetricians-gynecologists is necessary in order to facilitate coordinated approach with the general surgeons and oncologists for the optimal care of the patients.

The constitutively-expressed cyclooxygenase 1（CoX-1） and the inducible CoX-2 are both involved in the conversion of arachidonic acid（AA） to prostaglandins（PGs）.however,the functional roles of CoX-1 at the cellular level remain *** hypothesized that by comparing differential gene expression and eicosanoid metabolism in lung fibroblasts from wild-type（WT） mice and CoX-2^（-/-） or CoX-1^（-/-） mice may help address the functional roles of CoX-1 in inflammation and other cellular *** to WT,the number of specifically-induced transcripts were altered descendingly as follows：CoX-2^（-/-）〉 CoX-1^（-/-）〉 WT ＋ IL-1β.CoX-1^（-/-） or CoX-2^（-/-） cells shared about 50%of the induced transcripts with WT cells treated with IL-1β,*** interactive ＂anti-inflammatory,proinflammatory,and redox-activated＂ signature in the protein-protein interactome map was observed in CoX-2^（-/-） *** augmented CoX-1mRNA（in CoX-2^（-/-） cells） was associated with the upregulation of mRNAs for glutathione S-transferase（GST）,superoxide dismutase（SoD）,NAD（P）h dehydrogenase quinone 1（NQo1）,aryl hydrocarbon receptor（AhR）,peroxiredoxin,phospholipase,prostacyclin synthase,and prostaglandin E synthase,resulting in a significant increase in the levels of PGE2,PGD2,leukotriene B4（LTB4）,PGF（1α）,thromboxane B2（TXB2）,and PGF（2α）.The CoX-1 plays a dominant role in shifting AA toward the LTB4 pathway and anti-inflammatory *** to WT,the upregulated CoX-1 mRNA in CoX-2^（-/-） cells generated an ＂eicosanoid storm＂.The genomic characteristics of CoX-2^（-/-） is similar to that of proinflammatory cells as observed in IL-1β induced WT ***-1^（-/-） and CoX-2^（-/-） cells exhibited compensation of various eicosanoids at the genomic and metabolic levels.

Nibrin expression in oral squamous cell carcinoma:association with clinicopathological parameters

Aim:The present study sought to discover the role of Nibrin protein in 100 patients with oral squamous cell carcinoma(oSCC)and its potential relationship with clinicopathological ***:Nibrin expression was evaluated immunohistochemically using the modified h-score ***:The present study included 20%of patients with stage I disease,22%of patients with stage II disease,18%of patients with stage III disease,and 40%of patients with stage IV *** showed a significant positive correlation with moderately/poorly differentiated tumor tissues(P=0.028),while significant inverse correlation of Nibrin expression was observed with tumor size(P=0.018)and tumor stage(P=0.039).Further,using univariate survival analysis it was observed that strong Nibrin expression was significantly associated with disease relapse in early stage oSCC patients(P=0.049).Conclusion:Thus,the present study revealed that Nibrin could be used as a prognostic marker in patients with early stage oSCC.
