Objective:Bushen Tiansui formula(BSTSF),a traditional Chinese medicine prescription,has been widely used to treat Alzheimer’s disease(AD).However,the mechanisms underlying its effects remain largely *** this study,a rat AD model was used to study the effects of BSTSF on cognitive performance and expression of transfer RNA-derived small RNAs(tsRNAs)in the hippocampus,to determine whether treatment of AD with BSTSF could regulate the expression of tsRNAs,a novel small non-coding ***:To generate a validated AD model,oligomeric amyloid-β_(1-42)(Aβ_(1-42))was injected intracerebroventricularly into *** Morris water maze(MWM)test was used to evaluate rat cognitive performance,and tsRNA-sequencing was conducted to examine tsRNA expression in the rat *** targets were validated by quantitative real-time polymerase chain reaction(qRT-PCR).Bioinformatic analyses were conducted to investigate the biological function of candidate ***:The learning and memory deficits of Aβ_(1-42)-induced AD rats,assessed by MWM tests,were clearly ameliorated by BSTSF treatment.A total of 387 tsRNAs were detected in the rat *** them,13 were significantly dysregulated in AD rats compared with sham control rats,while 57 were markedly altered by BSTSF treatment,relative to untreated AD rats(fold change>2 and P<0.05).Moreover,six BSTSF treatment-related tsRNAs were identified and validated by *** analyses indicated that the six treatment-related tsRNAs had potential therapeutic roles,via multiple signaling pathways and Gene Ontology biological functions,including cyclic adenosine monophosphate and retrograde endocannabinoid ***:This study identified a previously uncharacterized mechanism underlying the effects of BSTSF in alleviating the learning and memory deficits in Aβ_(1-42)-induced AD rats,demonstrating that tsRNAs are potential therapeutic targets of BSTSF in the treatment of AD.

The SARG04 quantum key distribution protocol can offer greater robustness against photon number splitting attacks than the BB84 protocol that is implemented with weak *** this paper,we propose a tight key analysis for the SARG04 protocol,by considering the one-decoy method and investigating its performance under the influence of a detector *** results demonstrate that an increase in block size leads to a slight increase in both the secure key rate and the maximum transmission ***,the detector afterpulse plays a crucial role in practical applications and has a more pronounced effect on the SARG04 protocol compared to the BB84 protocol.

Effect of Central Multiphase Microstructure of Thick Plates on Work Hardening and Crack Propagation

The effect of multiphase microstructure in the center segregation zones(CSZ)of high-strength steel thick plates on work hardening characteristics and crack initiation/propagation were *** was correlated with deformation damage behavior by using Taylor factor(TF)gradients,kernel average misorientation(KAM),and geometric necessary dislocation(GND)*** results show that the segregation leads to a mixed structure of ferrite(soft phase)and martensite/bainite(hard phase)in the center of the thick *** to the 1/4 thickness(1/4 T)region,grain refinement occurred in the CSZ,with KAM values increasing from 0.24 to 0.49 and a decrease in the proportion of high-angle grain boundaries(>15°)from 77.4 to 51.8%.In the process of deformation,due to the influence of grain refinement and adjacent martensite/bainite,ferrite grains were divided into structural units with different TF *** higher KAM value and GND density at the interface between soft/hard phases resulted in severe work *** addition,the presence of micron-sized inclusions in the CSZ caused local strain concentration and non-continuous deformation of the matrix,which induced crack *** fracture surface showed a mixed mode of brittle cleavage fracture and ductile fracture.

The role of innate immunity in diabetic nephropathy and their therapeutic consequences

Diabetic nephropathy (DN) is an enduring condition that leads to inflammation and affects a substantial number of individuals with diabetes worldwide. A gradual reduction in glomerular filtration and emergence of proteins in the urine are typical aspects of DN, ultimately resulting in renal failure. Mounting evidence suggests that immunological and inflammatory factors are crucial for the development of DN. Therefore, the activation of innate immunity by resident renal and immune cells is critical for initiating and perpetuating inflammation. Toll-like receptors (TLRs) are an important group of receptors that identify patterns and activate immune responses and inflammation. Meanwhile, inflammatory responses in the liver, pancreatic islets, and kidneys involve inflammasomes and chemokines that generate pro-inflammatory cytokines. Moreover, the activation of the complement cascade can be triggered by glycated proteins. This review highlights recent findings elucidating how the innate immune system contributes to tissue fibrosis and organ dysfunction, ultimately leading to renal failure. This review also discusses innovative approaches that can be utilized to modulate the innate immune responses in DN for therapeutic purposes.

Semen microbiota in normal and leukocytospermic males

Large numbers of microbes can be present in seminal fluid,and there are differences in the semen microbiota between normal and abnormal semen *** evaluate the semen microbiota in patients with leukocytospermia,87 seminal fluid samples,including 33 samples with a normal seminal leukocyte count and 54 samples with leukocytospermia,were obtained for a cross-sectional *** samples with a normal seminal leukocyte count had normal sperm parameters(Control group),and 13 samples with a normal seminal leukocyte count were from asthenozoospermia patients(Ast group).However,32 samples with leukocytospermia were from asthenozoospermia patients(LA group),and only 22 samples with leukocytospermia had normal sperm parameters(Leu group).The 16S ribosomal RNA(rRNA)gene sequencing method was used to sequence the microbiota in the seminal fluid,and multiple bioinformatics methods were utilized to analyze the ***,the results showed that the worse sperm parameters were observed in the leukocytospermia-related *** microbiota analysis found that there was increased alpha diversity in the leukocytospermia-related ***,Proteobacteria,Actinobacteria,and Bacteroidetes were the primary phyla in the seminal *** microbiota profiles,namely,Lactobacillus-enriched and Streptococcus-enriched groups,were identified in this *** majority of the samples in the groups with a normal seminal leukocyte count could be categorized as Lactobacillus-enriched,whereas the majority of the leukocytospermia samples could be categorized as *** study indicated that males with leukocytospermia have worse sperm parameters and a different semen microbiota composition compared to males with a normal seminal leukocyte count.

Xuefu Zhuyu decoction improves neurological dysfunction by increasing synapsin expression after traumatic brain injury

Xuefu Zhuyu decoction has been used for treating traumatic brain injury and improving post-traumatic dysfunction, but its mechanism of action needs further investigation. This study established rat models of traumatic brain injury by controlled cortical impact. Rat models were intragastrically administered 9 and 18 g/kg Xuefu Zhuyu decoction once a day for 14 or 21 days. Changes in neurological function were assessed by modified neurological severity scores and the Morris water maze. Immunohistochemistry, western blot assay, and re- verse-transcription polymerase chain reaction were used to analyze synapsin protein and mRNA expression at the injury site of rats. Our results showed that Xuefu Zhuyu decoction visibly improved neurological function of rats with traumatic brain injury. These changes were accompanied by increased expression of synaptophysin, synapsin I, and postsynaptic density protein-95 protein and mRNA in a dose-de- pendent manner. These findings indicate that Xuefu Zhuyu decoction increases synapsin expression and improves neurological deficits alder traumatic brain injury.

Automated regression test method for scientific computing libraries:Illustration with SPHinXsys

Scientific computing libraries,whether in-house or open-source,have witnessed enormous progress in both engineering and scientific ***,it is important to ensure that modifications to the source code,prompted by bug fixing or new feature development,do not compromise the accuracy and functionality that have been already validated and *** paper introduces a method for establishing and implementing an automatic regression test environment,using the open-source multi-physics library SPHinXsys as an illustrative ***,a reference database for each benchmark test is generated from observed data across multiple *** comprehensive database encapsulates the maximum variation range of metrics for different strategies,including the time-averaged,ensemble-averaged,and dynamic time warping *** accounts for uncertainties arising from parallel computing,particle relaxation,physical instabilities,and ***,new results obtained after source code modifications undergo testing based on a curve-similarity comparison against the reference *** the source code is updated,the regression test is automatically executed for all test cases,providing a comprehensive assessment of the validity of the current *** regression test environment has been successfully implemented in all dynamic test cases within SPHinXsys,including fluid dynamics,solid mechanics,fluid-structure interaction,thermal and mass diffusion,reaction-diffusion,and their multi-physics couplings,and demonstrates robust capabilities in testing different *** is noted that while the current test environment is built and implemented for a particular scientific computing library,its underlying principles are generic and can be easily adapted for use with other libraries,achieving equal effectiveness.

Fracture mechanism of inclusions in thick plate center under tensile loading

Segregation of solute atoms in the center of thick plates of the tempered steel can cause an inhomogeneous structural transformation and generate micron-sized inclusions,which leads to lamellar tearing of thick plate and decreases the plasticity and *** formation and fragmentation mechanisms of micron-sized inclusions,like MnS and(Nb,Ti)C,in the center of thick plates were investigated by using thermodynamic calculations,finite element simulations,and electron backscatter diffraction characterization *** results show that micron-sized inclusions nucleate and grow in the liquid phase,and under tensile loading,they exhibit three fragmentation *** local stress during the fragmentation of inclusions is lower than the critical fracture stress of adjacent grains,and phase boundaries can effectively impede crack propagation into the *** existence of a low proportion of high-angle grain boundaries(58.1%)and high Kernel average misorientation value(0.534°)in the segregation band promotes inclusions fragmentation and crack *** difference in crack initiation and propagation direction caused by the morphology of inclusions and physical properties,as well as different matrix arrest abilities,is the main reasons for the diversity of inclusion fragmentation.
