Simultaneous determination of clopidogrel,2-oxo-clopidogrel,and the thiol metabolite of clopidogrel in human plasma by LC-MS/MS

Clopidogrel is a pro-drug which needs two-step metabolism to produce the active thiol metabolite.This study aimed to explore an efficient method to simultaneously determine the plasma clopidogrel,2-oxo-clopidogrel(2-Oxo-CLP),and the clopidogrel active metabolite(CAM).A high-throughput liquid chromatography tandem mass spectrometry(LC-MS/MS)was therefore developed.The analytes were extracted from plasma by using methyl tert-butyl ether(MTBE).Chromatographic separation was performed on a C18 column under an isocratic elution,accompanied with acetonitrile and deionized water containing 0.1%formic acid.After optimizing the condition of LC-MS/MS,a stable linearity was observed in the standard curves over the concentration ranges of 0.05 to 50.0 ng/mL for clopidogrel,0.5 to 50.0 ng/mL for 2-Oxo-CLP,and 0.5 to 100 ng/mL for clopidogrel active metabolite derivative(CAMD).The retention time was 4.78 minutes,3.79 minutes,3.59 minutes,and 4.82 minutes for clopidogrel,2-Oxo-CLP,CAMD,and internal standard,respectively.Both the relative standard deviation and the relative error were within the requirement of operating criteria.No significant degradation of clopidogrel,2-Oxo-CLP,and CAMD occurred under different storage conditions.This method was successfully validated in 3 patients with coronary artery disease.The results showed that the current LC-MS/MS method was efficient for simultaneously detecting clopidogrel,2-Oxo-CLP,and CAM with fine linearity,accuracy,precision,and stability.

Improving sentiment analysis accuracy with emoji embedding

Due to the diversity and variability of Chinese syntax and semantics,accurately identifying and distinguishing individual emotions from online texts is challenging.To overcome this limitation,we incorporate a new source of individual sentiment,emojis,which contain thousands of graphic symbols and are increasingly being used for expressing emotion in online conversations.We examined popular sentiment analysis algorithms,including rule-based and classification algorithms,to evaluate the impact of supplementing emojis as additional features to improve the algorithm performance.Emojis were also translated into corresponding sentiment words when con-structing features for comparison with those directly generated from emoji label words.In addition,considering different functions of emojis in texts,we classified all posts in the dataset by their emoji usage and examined the changes in algorithm performance.We found that emojis are effective as expanding features for improving the accuracy of sentiment analysis algorithms,and the algorithm performance can be further increased by taking different emoji usages into consideration.In this study,we developed an improved emoji-embedding model based on Bi-LSTM(namely,CEmo-LSTM),which achieves the highest accuracy(around 0.95)when analyzing online Chinese texts.We applied the CEmo-LSTM algorithm to a large dataset collected from Weibo from December 1,2019 to March 20,2020 to understand the sentiment evolution of online users during the COVID-19 pandemic.We found that the pandemic remarkably impacted individual sentiments and caused more passive emotions(e.g.,horror and sadness).Our novel emoji-embedding algorithm creatively combined emojis as well as emoji usage with the sentiment analysis model and can handle emotion mining tasks more effectively and efficiently.

Dear Editor,The coronavirus disease 2019(COVID-19) broke out in early December 2019 in Wuhan, China~([1]), which put tremendous pressure on the medical system. A nationwide lockdown and strict quarantine measures proved effective in reducing the spread of the pandemic. However, it might have affected the management of time-dependent diseases such as STelevation myocardial infarction(STEMI)~([2]).

2020 was an unprecedented year,with rapid and drastic changes in human mobility due to the COVID-19 pandemic.To understand the variation in commuting patterns among the Chinese population across stable and unstable periods,we used nationwide mobility data from 318 million mobile phone users in China to examine the extreme fluctuations of population movements in 2020,ranging from the Lunar New Year travel season(chunyun), to the exceptional calm of COVID-19 lockdown,and then to the recovery period.We observed that cross-city movements,which increased substantially in chunyun and then dropped sharply during the lockdown,are primarily dependent on travel distance and the socio-economic development of cities.Following the Lunar New Year holiday,national mobility remained low until mid-February,and COVID-19 interventions delayed more than 72.89 million people returning to large cities.Mobility network analysis revealed clusters of highly connected cities,conforming to the social-economic division of urban agglomerations in China.While the mass migration back to large cities was delayed,smaller cities connected more densely to form new clusters.During the recovery period after travel restrictions were lifted,the netflows of over 55% city pairs reversed in direction compared to before the lockdown.These findings offer the most comprehensive picture of Chinese mobility at fine resolution across various scenarios in China and are of critical importance for decision making regarding future public-health-emergency response,transportation planning and regional economic development,among others.

为了减少永磁同步电机调速中的动态误差,提出了一种积分性能最优的滑模控制方法。该方法以滑模控制中的动态误差为性能指标,在此基础上建立最优切换函数,并采用最优控制理论对滑模控制器进行设计。用该方法设计的滑模控制系统,通过滑模面斜率的连续变化,能够加速系统状态变量到达滑模面的过程,极大地提高对参数摄动和外部干扰的鲁棒性。仿真结果表明,该时变滑模面控制方法使系统具有无超调、快速、稳定等优点,提高了系统的鲁棒性。

本研究旨在明确塞内卡病毒A(Senecavirus A,SVA)FJLY株的基因组特性及其与其他毒株的同源性关系,并了解SVA-FJLY株的致病性。以分离自福建省某养殖场中的一株SVA-FJLY株为研究对象,参照其原型毒株SVV-001(GenBank No.DQ641257.1)全基因组序列设计5对引物,通过RT-PCR扩增、测序、拼接,获得SVA-FJLY株全基因组序列并进行序列分析;并通过滴鼻攻毒3~4周龄仔猪。结果表明,SVA-FJLY株基因组全长7275 bp(不包括PolyA),SVA-FJLY株基因组存在一个多聚蛋白、VP1蛋白和VP2蛋白。SVA-FJLY株与参考毒株之间的基因组一级结构有较高的相似性,与HeB01-2017株(MF967574.1)相似性最高,达98.5%;而与SVA原型毒株SVV-001株(DQ641257.1)的相似性较低,为93.9%。SVA-FJLY株与国内几个毒株属于同一个分支,与国内的HeB01-2017株亲缘关系最近,同时与国外Colombia-2016株(KX857728.1)的亲缘关系最近。通过动物试验发现试验组猪攻毒10 d后,试验组2/3发病,对照组3/3正常,未出现仔猪死亡。通过全基因组序列测定获得了SVA-FJLY株全基因组序列并进行序列分析,明确SVA-FJLY株的基因组特性及其与其他毒株的相似性关系,了解了SVA-FJLY株的致病性,为SVA的反向遗传学和疫苗的研制提供参考依据,同时也丰富了SVA的基因组信息数据库。

目的研究四方木皮化学成分及抗炎镇痛的作用。方法利用UPLC-MS/MS技术定性分析四方木皮次生代谢成分,并借助TCMSP数据库平台分析其活性成分对应靶点和疾病。采用小鼠耳肿胀法、足趾肿胀法研究四方木皮的抗炎作用;热板法、扭体实验研究四方木皮的镇痛作用。结果四方木皮共鉴定出7类541种次生代谢物,其中,TCMSP平台鉴定出120种中药化学成分,52种成分与抗炎镇痛显著相关,其中数量较多的是黄酮类与酚酸类代谢物;四方木皮低、中、高剂量组均可明显抑制小鼠耳肿胀和足趾肿胀以及减少小鼠的扭体次数(P 0.05)。结论四方木皮乙醇提取物具有一定抗炎镇痛作用,黄酮和酚类物质在四方木皮的中药活性及抗炎镇痛方面中起重要作用,为进一步阐明四方木皮的抗炎、镇痛作用机制提供了基础。

目的:探讨氯吡格雷低反应(clopidogrel low response,CLR)与正常反应的冠心病(coronary artery disease,CAD)患者血小板miRNA表达谱的差异。方法:连续入选78例接受氯吡格雷治疗(负荷剂量300 mg,维持剂量75 mg/d)至少5 d的冠心病患者,通过光学血小板聚集仪检测所有患者二磷酸腺苷诱导的血小板聚集率(platelet aggregation induced by adenosine diphosphate,PLADP),PLADP大于上四分位数的19例患者为CLR组,小于下四分位数的19例患者为对照组。提取所有入选患者纯化血小板中的总RNA,将上述两组患者的总RNA分别混为2个总RNA池,通过RNA变性电泳质检后,采用高通量测序筛选两组患者血小板miRNA的差异表达谱。通过靶基因预测软件TargetScan、miRanda、PITA和miRWalk对差异miRNA的功能进行预测。结果:两组患者临床基线资料无统计学意义。CLR组PLADP显著高于对照组(P 2的拷贝数前20位miRNA依次是hsa-miR-300、hsa-miR-151b、hsa-miR-1299等。通过至少2个靶基因预测软件印证8个miRNA(hsa-miR-188-5p、hsa-miR-6874-3p、hsa-miR-218-5p、hsa-miR-3150b-3p、hsa-miR-1288-3p、hsa-miR-1299、hsa-miR-6862-5p、hsa-miR-4421)对血小板聚集关键蛋白有调控作用。结论:CLR患者中存在血小板miRNA的显著下调,本研究筛选出8个对血小板聚集关键蛋白可能有调控作用的miRNA,可能成为个体化抗血小板治疗提供新的干预手段。

目的:探究冠心病患者服用氯吡格雷后血小板抑制作用达到稳态的时间,为氯吡格雷药效学研究提供借鉴。方法:入选接受经皮冠状动脉介入治疗且服用氯吡格雷3 d后的冠心病患者,采用光比浊法检测二磷酸腺苷诱导的血小板聚集率(PL;)。分别比较检测时已服用氯吡格雷不同天数患者住院期间和服用氯吡格雷30 d后的PL;水平。结果:入选240例患者,住院期间及出院后检测PL;距离首次服用氯吡格雷的时间分别为6(5,8)d和39(35,47)d,患者住院期间PL;水平显著高于服用氯吡格雷≥30 d的PL;水平[36(25,46)%vs 29(20,44)%,P0.05)。入选患者中服用氯吡格雷负荷剂量(150 mg或300 mg)者84例,住院期间与服用氯吡格雷≥30 d的PL;水平差异无统计学意义[(35.99±15.38)%vs(32.61±16.30)%,P>0.05];检测时已服用氯吡格雷不同天数患者住院期间与服用氯吡格雷30 d的PL;水平差异均无统计学意义(P>0.05)。结论:冠心病患者服用维持量氯吡格雷≥6 d或负荷加维持量氯吡格雷≥3 d时残余血小板聚集功能与服用氯吡格雷30 d后相当;在以上时间窗检测的PL;水平可准确体现氯吡格雷的长期抗血小板疗效。
