Self-assembly nanomicelle-microneedle patches with enhanced tumor penetration for superior chemo-photothermal therapy

Nanomedicine with high specificity has been a promising tool for cancer diagnosis and ***,the successful application of nanoparticle-based superficial cancer therapy is severely hindered by restricted deep tumor tissue accumulation and ***,a self-assembly nanomicelle dissolving microneedle(DMN)patch according to the“nano in micro”strategy was conducted to co-deliver a first-line chemotherapeutic agent paclitaxel(PTX),and a photosensitizer IR780(PTX/IR780-NMs@DMNs)for chemo-photothermal synergetic melanoma *** direct insertion into the tumor site,DMNs created a regular and multipoint three-dimensional drug depot to maximize the tumor *** by the DMN dissolution,the composition of the needle matrixes self-assembled into nanomicelles,which could efficiently penetrate deep tumor *** laser irradiation,the nanomicelles could not only ablate tumor cells directly by photothermal conversion but also trigger PTX release to induce tumor cell *** vivo results showed that compared with intravenous injection,IR780 delivered by PTX/IR780-NMs@DMNs was almost completely accumulated at the tumor *** antitumor results revealed that the PTX/IR780-NMs@DMNs could effectively eliminate tumors with an 88%curable rate without any damage to normal *** work provides a versatile and generalizable framework for designing self-assembly DMN-mediated combination therapy to fight against superficial cancer.

Malignant tumor has become an urgent threat to global public *** of the heterogeneity of tumor,single therapy presents great limitations while synergistic therapy is arousing much attention,which shows desperate need of intelligent carrier for co-delivery.A core-shell dual metaleorganic frameworks(MOFs)system was delicately designed in this study,which not only possessed the unique properties of both materials,but also provided two individual specific functional zones for co-drug *** indocyanine green(ICG)and chemotherapeutic agent doxorubicin(DOX)were stepwisely encapsulated into the nanopores of MIL-88 core and ZIF-8 shell to construct a synergistic photothermal/photodynamic/chemotherapy *** for efficient drug delivery,the MIL-88 could be functioned as a nanomotor to convert the excessive hydrogen peroxide at tumor microenvironment into adequate oxygen for photodynamic *** DOX release from MIL-88-ICG@ZIF-8-DOX nanoparticles was triggered at tumor acidic microenvironment and further accelerated by near-infrared(NIR)light *** in vivo antitumor study showed superior synergistic antitumor effect by concentrating the nanoparticles into dissolving microneedles as compared to intravenous and intratumoral injection of nanoparticles,with a significantly higher inhibition *** is anticipated that the multi-model synergistic system based on dual-MOFs was promising for further biomedical application.

Ferroptosis is a nonapoptotic form of cell death and differs considerably from the well-known forms of cell death in terms of cell morphology,genetics,and *** three primary pathways for cell ferroptosis are system Xc^(-)/glutathione peroxidase 4(GPX4),lipid metabolism,and ferric *** the discovery of ferroptosis,mounting evidence has revealed its critical regulatory role in several diseases,especially as a novel potential target for cancer therapy,thereby attracting increasing attention in the fields of tumor biology and anti-tumor ***,broad prospects exist for identifying ferroptosis as a potential therapeutic *** this review,we aimed to systematically summarize the activation and defense mechanisms of ferroptosis,highlight the therapeutic targets,and discuss the design of nanomedicines for ferroptosis *** addition,we opted to present the advantages and disadvantages of current ferroptosis research and provide an optimistic vision of future directions in related ***,we aim to provide new ideas for further ferroptosis research and inspire new strategies for disease diagnosis and treatment.

Mesoporous silica nanoparticles(MSNs) are attracting increasing interest for potential biomedical applications. With tailored mesoporous structure, huge surface area and pore volume,selective surface functionality, as well as morphology control, MSNs exhibit high loading capacity for therapeutic agents and controlled release properties if modified with stimuli-responsive groups, polymers or proteins. In this review article, the applications of MSNs in pharmaceutics to improve drug bioavailability, reduce drug toxicity, and deliver with cellular targetability are summarized. Particularly,the exciting progress in the development of MSNs-based effective delivery systems for poorly soluble drugs, anticancer agents, and therapeutic genes are highlighted.

The effect of organic ligand modification on protein corona formation of nanoscale metal organic frameworks

Nanoscale metal organic frameworks(NMOFs)have been widely reported in biomedical field for their unique porous structure and tunable ***,when administrated in vivo,the protein corona will be formed on the surface of NMOFs,significantly affecting their biodistribution,pharmacokinetics and drug *** studies paid attention to the protein corona formation process and its influencing factors of *** a well-established strategy for altering structure features of NMOFs,the organic ligand modification may have effect on the protein corona formation process,which is to be *** this study,the zirconium(Zr)-based UIO66 was chosen as model NMOFs,the organic ligand of which was modified with amino group(-NH_(2))or carboxyl group(-COOH)to synthesize UIO66-NH_(2)and UIO66-2COOH,*** serum albumin(BSA)was chosen as model protein to investigate the protein corona formation process of *** current results showed that the-COOH modification remarkably enhanced the BSA adsorption on NMOFs while-NH_(2)slightly decreased the protein binding *** differences may be ascribed to the two different dominate protein corona formation modes,i.e.,surface coating mode and porous embedded *** protein corona formation did not affect the crystal phase of NMOFs but increased the content ofα-helix of ***,upon protein corona formation,the cellular uptake of NMOFs was significantly *** believe our study will provide a new research paradigm to the design and applications of NMOFs.

Microneedles(MNs)can be used for the topical treatment of skin disorders as they directly deliver therapeutics to the site of skin lesions,resulting in increased therapeutic efficacy while having minimum side *** are used to deliver different kinds of therapeutics(e.g.,small molecules,macromolecules,nanomedicines,living cells,bacteria,and exosomes)for treating various skin disorders,including superficial tumors,wounds,skin infections,inflammatory skin diseases,and abnormal skin *** therapeutic efficacy of MNs can be improved by integrating the advantages of multiple therapeutics to perform combination *** careful designing,MNs can be further modified with biomimetic structures for the responsive drug release from internal and external stimuli and to enhance the transdermal delivery efficiency for robust therapeutic *** studies have proposed the use of drug-free MNs as a promising mechanotherapeutic strategy to promote wound healing,scar removal,and hair regeneration via a mechanical communication *** MNs have several advantages,the practical application of MNs suffers from problems related to industrial manufacture and clinical evaluation,making it difficult for clinical *** this study,we summarized the various applications,emerging challenges,and developmental prospects of MNs in skin disorders to provide information on ways to advance clinical translation.

Pulmonary drug delivery has attracted increasing attention in biomedicine,and porous particles can effectively enhance the aerosolization performance and bioavailability of ***,the existing methods for preparing porous particles using porogens have several drawbacks,such as the inhomogeneous and uncontrollable pores,drug leakage,and high risk of *** this study,a series of cyclodextrin-based metal-organic framework(CD-MOF)particles containing homogenous nanopores were delicately engineered without *** with commercial inhalation carrier,CDMOF showed excellent aerosolization performance because of the homogenous nanoporous *** great biocompatibility of CD-MOF in pulmonary delivery was also confirmed by a series of experiments,including cytotoxicity assay,hemolysis ratio test,lung function evaluation,in vivo lung injury markers measurement,and histological *** results of ex vivo fluorescence imaging showed the high deposition rate of CD-MOF in ***,all results demonstrated that CD-MOF was a promising carrier for pulmonary drug *** study may throw light on the nanoporous particles for effective pulmonary administration.
