Role of simple biomarkers in predicting fibrosis progression in HCV infection

AIM: To examine the accuracy of the aspartate aminotransferase (AST)/platelet Ratio Index (ApRI) and FIB-4, in predicting longitudinal changes in liver histology in hepatitis C virus (HCV) ***: patients that underwent repeat liver biopsies at least 1 year apart from 1999 to 2007 were identified. Liver fibrosis was staged on needle core biopsies evaluated by a single expert liver pathologist. Only laboratory values within 3 mo of the liver biopsies were ***: Thirty-six patients met the inclusion criteria with 50% stage 1 on initial biopsy, 25% stage 2, and 22% stage 3. Nineteen of 36 (53%) had progression of fibrosis on repeat biopsies, while 16 (44%) showed no change in stage, and one (3%) showed improvement. patients that showed progression of fibrosis had significantly higher alanine aminotransferase and aspartate aminotransferase levels than the group that did not show progression. A significant correlation was seen between change in stage of fibrosis and change in ApRI (r2 = 0.39, p = 0.00001) and a change in FIB-4 (r2 = 0.31, p = 0.00004). A change in ApRI (ΔApRI) of 0.18 had 80% positive predictive value (ppV) and 67% nega- tive predictive value (NpV) for progression of fibrosis. A change in FIB-4 (ΔFIB-4) of 0.39 had 75% ppV and 75% NpV for predicting progression of fibrosis. CONCLUSION: ΔApRI and ΔFIB-4 parallel changes in fibrosis progression, and could be useful tools for clinicians in following patients with active chronic HCV infection.

AIM:Cytokine release by macrophages critically determines the type of immune response to an ***,we studied hepatitis C virus(HCV)-specific induction of interleukins-1β,-10,-12(IL-1β,IL-10,IL-12),and tumor necrosis factor-α(TNF-α)in monocytes. METHODS:Intracallular cytokine expression was studied by flow cytometry in 23 patients with chronic hepatitis C,14 anti-HCV seropositives without viremia and 11 controls after stimulation of peripheral blood mononuclear calls with recombinant core,NS3,NS4,NSSa and NSSb proteins. RESULTS:patients with HCV viremia revealed greater spontaneous expression of IL-1β,TNF-α,and IL-10. Furthermore,greater than twofold higher IL-10 expression was induced by the HCV antigens in chronic hepatitis C than in the other two groups(ppreferentially. CONCLUSION:In chronic hepatitis C antigen-specific cytokine induction in monocytes is apparently shifted towards predominant IL-10 induction-not counterbalanced by antiviral type 1 *** may contribute to persistent viral replication.

Role of Mdm2 and Mdmx in DNA repair

Mdm2 and Mdmx are critical regulators of the p53 tumour suppressor and are overexpressed in many human malignancies. However, in recent years, their impact on genome instability was shown to be at least, in part, independent of p53. Both Mdm2 and Mdmx inhibit DNA break repair through their association with the Mrell/RadSO/Nbs1 DNA repair complex. Recent evidence indicates that harnessing Mdm2 and/or Mdmx-mediated inhibition of DNA break repair in cancer cells could provide a therapeutic opportunity, particularly for those malignancies that have lost functional p53.

Optimizing photorespiration for improved crop productivity

In C3 plants, photorespiration is an energyexpensive process, including the oxygenation of ribulose-1,5-bisphosphate(RuBp) by ribulose 1,5-bisphosphate carboxylase/oxygenase(Rubisco) and the ensuing multiorganellar photorespiratory pathway required to recycle the toxic byproducts and recapture a portion of the fixed carbon. photorespiration significantly impacts crop productivity through reducing yields in C3 crops by as much as 50%under severe conditions. Thus, reducing the flux through, or weive R improving the efficiency of photorespiration has the potential of large improvements in C3 crop ***, we review an array of approaches intended to engineer photorespiration in a range of plant systems with the goal of increasing crop productivity. Approaches include optimizing flux through the native photorespiratory pathway, installing non-native alternative photorespiratory pathways, and lowering or even eliminating Rubiscocatalyzed oxygenation of RuBp to reduce substrate entrance into the photorespiratory cycle. Some proposed designs have been successful at the proof of concept level.A plant systems-engineering approach, based on new opportunities available from synthetic biology to implement in silico designs, holds promise for further progress toward delivering more productive crops to farmer’s fields.

Use of a saline-coupled bipolar sealer open liver resection for hepatic malignancy: Medical resource use and costs

AIM To evaluate outcomes associated with use of a saline coupled bipolar sealer during open partial liver *** This retrospective analysis utilized the United States premier? insurance claims database(2010-2014). patients were selected with codes for liver malignancy and partial hepatectomy or lobectomy. Cases were defined by use the saline-coupled bipolar sealer; controls had no use. A propensity Score algorithm was used to match one case to five controls. A deviationbased cost modeling(DBCM) approach provided an estimate of *** One hundred and forty-four cases and 720 controls were available for analysis. patients in the case cohort received fewer transfusions vs controls(18.1% vs 29.4%, p = 0.007). In DBCM, more patients in the case cohort experienced "on-course" hospitalizations(53.5% vs 41.9%, p = 0.009). The cost calculation showed an average savings in total hospitalization costs of $1027 for cases vs controls. In multivariate analysis, cases had lower odds of receiving a transfusion(OR = 0.44, 95%CI: 0.27-0.71, p = 0.0008).CONCLUSION Use of a saline-coupled bipolar sealer was associated with a greater proportion of patients with an "on course" hospitalization.

To give soils and soil degradation,which are among the most crucial threats to ecosystem stability,social and political visibility,small and large scale modelling and mapping of soil erosion is *** most widely used approaches during an 80year history of erosion modelling are Universal Soil Loss Equation (USLE)-type based algorithms which have been applied in 109 *** soil erosion by water (excluding gully erosion and land sliding),we start this review with a statistical evaluation of nearly 2,000 publications).We discuss model developments which use USLE-type equations as basis or side modules,but we also address recent development of the single USLE parameters (R,K,LS,C,p).Importance,aim and limitations of model validation as well as a comparison of USLE-type models with other erosion assessment tools are *** comparisons demonstrate that the application of process-based physical models (e.g.,WEpp or pESERA) does not necessarily result in lower uncertainties compared to more simple structured empirical models such as USLE-type *** identified four key areas for future research:(i) overcoming the principally different nature of modelled (gross) versus measured (net) erosion rates,in coupling on-site erosion risk to runoff patterns,and depositional regime,(ii) using the recent increase in spatial resolution of remote sensing data to develop process based models for large scale applications,(iii) strengthen and extend measurement and monitoring programs to build up validation data sets,and (iv) rigorous uncertainty assessment and the application of objective evaluation criteria to soil erosion modelling.

Effects of mountain pine beetle-killed forests on source water contributions to streamflow in headwater streams of the Colorado Rocky Mountains

Natural or human-influenced disturbances are important to the health and diversity of forests, which in turn, are important to the water quantity and quality exported from a catchment. However, human-induced disturbances （prescribed fire and harvesting） have been decreasing, and natural disturbances （fires and insects） have been increasing in frequency and severity. One such natural disturbance is the mountain pine beetle （MpB）, （Denclroctonus ponderosae） an endemic species. A recent epidemic resulted in the mortality of millions of hectares of lodgepole pine （pinus contorta） forests in Colorado, USA. Beetle-induced tree mortality brings about changes to the hydrologic cycle, including decreased transpiration and interception with the loss of canopy cover. This study examined the effect of the mountain pine beetle kill on source water contributions to streamflow in snowmelt- dominated headwater catchments using stable isotopes （2H and ~80） as tracers. Study catchments with varying level of beetle-killed forest area （6% to 97%） were sampled for groundwater, surface water, and precipitation. Streams were sampled to assess whether beetle-killed forests have altered source water contributions to streamflow. Ground- water contributions increased with increasing beetle-killed forest area （p = 0.008）. Both rain and snow contributions were negatively correlated with beetle-killed forest area （p = 0.035 and p = 0.011, respectively）. As the beetle-killed forest area increases, so does fractional groundwater contribution to streamflow.

Mesenchymal stem cells(MSCs)derived from human embryonic stem cells(hESCs)have significant potential for cell-mediated bone *** recent study revealed that inhibiting the epigenetic regulator EZH2 plays a key role in promoting the mesodermal differentiation of *** this study,an epigenome-wide analysis of hESCs and MSCs revealed that growth differentiation factor 6(GDF6),which is involved in bone formation,was the most upregulated gene associated with MSCs compared to ***,we identified GDF6 as a repressive target of EZH2 and found that ectopic GDF6 selectively promoted hESC differentiation towards the mesodermal lineage and enriched the MSC *** results provide molecular insights governing the mesenchymal commitment of hESCs and identify an inducing factor that offers strong promise for the future of regenerative medicine.
