Dysregulation of G9a,a histone-lysine N-methyltransferase,has been observed in Alzheimer’s disease and has been correlated with increased levels of chronic inflammation and oxidative ***,microRNAs are involved in many biological processes and diseases playing a key role in pathogenesis,especially in multifactorial diseases such as Alzheimer’s ***,our aim has been to provide partial insights into the interconnection between G9a,microRNAs,oxidative stress,and *** better understand the biology of G9a,we compared the global microRNA expression between senescence-accelerated mouse-prone 8(SAMP8)control mice and SAMP8 treated with G9a inhibitor *** found a downregulation of miR-128 after a G9a inhibition treatment,which interestingly binds to the 3′untranslated region(3′-UTR)of peroxisome-proliferator activator receptor γ(PPARG)***,Pparg gene expression levels were higher in the SAMP8 group treated with G9a inhibitor than in the SAMP8 control *** also observed modulation of oxidative stress responses might be mainly driven Pparg after G9a *** confirm these antioxidant effects,we treated primary neuron cell cultures with hydrogen peroxide as an oxidative *** this setting,treatment with G9a inhibitor increases both cell survival and antioxidant ***,up-regulation of PPARγby G9a inhibitor could also increase the expression of genes involved in DNA damage responses and *** addition,we also described that the PPARγ/AMPk axis partially explains the regulation of autophagy markers ***,PPARγ/GADD45αpotentially contributes to enhancing synaptic plasticity and neurogenesis after G9a ***,we propose that pharmacological inhibition of G9a leads to a neuroprotective effect that could be due,at least in part,by the modulation of PPARγ-dependent pathways by miR-128.

Explaining the influence of dopant concentration and excitation power density on the luminescence and brightness of β-NaYF4:Yb^3+,Er^3+ nanoparticles:Measurements and simulations

We assessed the influence of Yb^3+ and Er34 dopant concentration on the relative spectral distribution, quantum yield (Фuc), and decay kinetics of the upconversion luminescence (UCL) and particle brightness (BUC) for similarly sized (33 nm) oleate-capped p-NaYF4:Yb3t,Er^3+ upconversion (UC) nanoparticles (UCNPs) in toluene at broadly varied excitation power densities (F). This included an Yb^3+ series where the Yb^3+ concentration was varied between 11%—21% for a constant Er^3+ concentration of 3%, and an Er^3+ series, where the Er^3+ concentration was varied between 1%-4% for a constant Yb^3+ concentration of 14%. The results were fitted with a coupled rate equation model utilizing the UCL data and decay kinetics of the green and red Er34 emission and the Yb34 luminescence at 980 nm. An increasing Yb34 concentration favors a pronounced triphotonic population of 4F9/2 at high P by an enhanced back energy transfer (BET) from the 4G11/2 level. Simultaneously, the Yb^3+-controlled UCNPs absorption cross section overcompensates for the reduction in Фuc with increasing Yb^3+ concentration at high P, resulting in an increase in Sue. Additionally, our results show that an increase in Yb34 and a decrease in Er^3+ concentration enhance the color tuning range by P. These findings will pave the road to a deeper understanding of the energy transfer processes and their contribution to efficient UCL, as well as still debated trends in green-to-red intensity ratios of UCNPs at different P.

Trade-offs between ecosystem service provision and the predisposition to disturbances: a Nfi-based scenario analysis

Background: Scenario analyses that evaluate management effects on the long-term provision and sustainability of forest ecosystem services and biodiversity(ESB) also need to account for disturbances. The objectives of this study were to reveal potential trade-offs and synergies between ESB provision and disturbance predisposition at the scale of a whole ***: The empirical scenario model MASSIMO was used to simulate forest development and management from years 2016 to 2106 on 5086 sample plots of the Swiss National Forest Inventory(Nfi). We included a businessas-usual(BAU) scenario and four scenarios of increased timber harvesting. Model output was evaluated with indicators for 1) ESB provision including a) timber production, b) old-growth forest characteristics as biodiversity proxies and c) protection against rockfall and avalanches and 2) for a) storm and b) bark beetle ***: The predisposition indicators corresponded well(AUC: 0.71–0.86) to storm and insect(mostly bark beetle)damage observations in logistic regression models. Increased timber production was generally accompanied with decreased predisposition(storm: >-11%, beetle: >-37%, depending on region and scenario), except for a scenario that promoted conifers where beetle predisposition increased(e.g. + 61% in the Southern Alps). Decreased disturbance predisposition and decreases in old-growth forest indicators in scenarios of increased timber production revealed a trade-off situation. In contrast, growing stock increased under BAU management along with a reduction in conifer proportions, resulting in a reduction of beetle predisposition that in turn was accompanied by increasing old-growth forest indicators. The International Union of Forest Research Organizations （IUFRO） and Debates About Forest-Water Relations During the Late 19th Century

Examining the International Union of Forest Research Organizations （IUFRO） and its research on forest-water relations, the article investigates the relation between scientific internationalism and national expertise. It juxtaposes existing arguments regarding the paradigmatic character of 19th century arguments about deforestations. In particular, in 1999, christian Pfister and Daniel Br^indli argued that, in Switzerland during the second half of the 19th century, forestry administrators forged an argument that deforestations in mountainous regions lead to floods in the valleys. As this argument helped to install new Swiss forest legislation in 1876, forbidding deforestations in the mountains, Pfister and Brandli called it a ＂deforestation paradigm＂. In contrast, sources of the IUFRO provide a different picture. At IUFRO meetings, i.e., at the international level of debate, no such paradigm existed. Instead, IUFRO participants discussed various conclusions that could be drawn fi＇om research on deforestations. Regarding the outcome of IUFRO projects, participants reported partly in opposing ways about the results of the research projects in forestry journals in their respective countries. Exploring these reports, the article provides an explanation for these different national representations of international research projects.

AIM: To study the expression of HBV enhancer Ⅱ by transcription factor COUP-TF1. METHODS: In order to study the regulation of HBV variants in the vicinity of the NRRE we cloned luciferase constructs containing the HBV enhancer Ⅱ from variants and from HBV genotypes A and D and cotransfected them together with expression vectors for COUP-TF1 into HepG2 cells. RESULTS: Our fi ndings show that enhancer Ⅱ of HBV genotype A is also repressed by COUP-TF1. In contrast, two different enhancer Ⅱ constructs of HBV genotype D were activated by COUP-TF1. The activation was independent of the NRRE because a natural variant with a deletion of nt 1763-1770 was still activated by COUP- TF1. CONCLUSION: Regulation of transcription of the HBV genome seems to differ among HBV genomes derived from different genotypes. These differences in transcriptional control among HBV genotypes may be the molecular basis for differences in the clinical course among HBV genotypes.

Bcl-2 degradation is an additional pro-apoptotic effect of polo-like kinase inhibition in cholangiocarcinoma cells

To examine the influence on apoptotic mechanisms following inhibition of polo-like kinases as therapeutically approach for cholangiocellular cancer *** most cholangiocarcinomas are chemotherapy-resistant due to mechanisms preventing tumor cell death, we investigated the effect of Cisplatin on cholangiocellular carcinoma (CCA) cell lines kMCH-1 and Mz-Ch-1. Polo-like kinases (PLk) are important regulators of the cell cycle and their inhibition is discussed as a potential therapy while PLk inhibition can regulate apoptotic mediators. Here, cells were treated with PLk inhibitor BI6727 (Volasertib), Cisplatin, and in combination of both compounds. Cell viability was assessed by MTT; apoptosis was measured by DAPI staining and caspase-3/-7 assay. Western blot and qRT-PCR were used to measure expression levels of apoptosis-related molecules Bax and *** cell viability in the CCA cell lines kMCH-1 and Mz-Ch-1 was reduced in all treatment conditions compared to vehicle-treated cells. Co-treatment with BI6727 and cisplatin could even enhance the cytotoxic effect of cisplatin single treatment. Thus, co-treatment of cisplatin with BI6727 could slightly enhance the cytotoxic effect of the cisplatin in both cell lines whereas there was evidence of increased apoptosis induction solely in Mz-Ch-1 as compared to kMCH-1. Moreover, PLk inhibition decreases protein levels of Bcl-2; an effect that can be reversed by the proteasomal degradation inhibitor MG-132. In contrast, protein levels of Bax were not found to be altered by PLk inhibition. These findings indicate that cytotoxic effects of Cisplatin in Mz-Ch-1 cells can be enhanced by cotreatment with *** conclusion, BI6727 treatment can sensitize CCA cells to cisplatin-induced apoptosis with proteasomal Bcl-2 degradation as an additional pro-apoptotic effect.

Response evaluation of chemotherapy in metastatic colorectal cancer by contrast enhanced ultrasound

AIM： To evaluate whether contrast enhanced ultra- sound （CEUS） might also be used for response predic- tion and early response evaluation in patients receiving bevacizumab based chemotherapy for metastasized colorectal ***： Thirty consecutive patients with non prima- ry resectable liver metastases from colorectal cancer underwent CEUS before treatment （CEUS date 1） and before the second （CEUS date 2） and fourth （CEU5 date 3） cycle of bevacizumab based chemotherapy. Three parameters [PEAk, Time to peak （l-I-P） and RISE RATE]were correlated with radiological ***： For neoadjuvant purpose a reduction of tu- mour mass was required to assume clinical response. Based on these response criteria there was a significant （P 〈 0.001） correlation in TTP between metastases of responders （9.08 s） and non-responders （14.76 s） ar- chived on CEUS date 1. By calculating a standardized quotient （metastases divided by normal liver tissue） we were able to define a cut off, predicting response with a sensitivity of 92.3 % and a specificity of 100 %. To reflect a palliative intention only those patients with progressive disease were classified as non-responders. In this stetting -fi-P was also significantly （P 〈 0.01） dif- ferent between responders and non-responders. In con- trast, Peak and Rise rate did not show any significant difference between responder and non-responder. CONCLUSION： CEUS might serve as a surrogate mark- er to predict treatment response in patients with me- tastasized colorectal cancer who receive antiangiogenic therapy.

Assessing Lysine Requirement of Growing Chicken by Direct Comparison between Supplementation Technique and “Goettingen Approach”

Validated procedures play an important role to obtain accurate information about individual amino acid requirement data. The aim of the present study was to assess lysine (Lys) requirement of growing chicken both by classical supplementation technique and principles of diet dilution technique as applied with “Goettingen approach”. During the starter period (1 - 21 d), a growth study with male meat type chicken (Ross 308) was conducted making use of five graded dietary Lys-levels (3 repetition boxes with 3 birds/box). L-Lys×HCl was gradually added to a diet based on wheat, soybean protein concentrate, wheat gluten and fishmeal to yield 80%, 87.5%, 95%, 102.5% and 110% of the expected requirement level (13 g Lys/kg as fed). Diets were iso-energetic (12.8 MJME/kg) and iso-nitrogenous (21.65% crude protein). Birds were fed on free choice level also to assess the feed intake (fi) effects as important factor on traditional response criteria. Analyzed body composition at start and end of the growth study yielded N deposition (ND) data for further data assessment using exponential approximations depending on dietary Lys content or observed Lys intake. The results indicated significant differences (p ke) and aimed level of daily ND, the needed in-feed content of Lys varied between 1.24% and 1.46%. Application of the exponential modelling by “Goettingen approach” overcame these misleading conclusions by modelling the relationship between required Lys intake and observed response data (BWG, ND) taking also into account the expected real feed intake to formulate the needed in-feed concentration.

Approximately 950 individual sequences of nonribosomally biosynthesised peptides are produced by the genus Trichoderma/Hypocreathat belong to a perpetually growing class of mostly linear antibiotic oligopeptides,which are rich in the non-proteinogenic α-aminoisobutyric acid(Aib).Thus,they are comprehensively named ***,peptaibiotics represent ca.80% of the total inventory of secondarymetabolites currently known from Trichoderma/*** unique membrane-modifying bioactivity results from amphipathicity and helicity,thus making them ideal candidates in assisting both colonisation and defence of the natural habitats by their fungal *** this,reports on the in vivo-detection of peptaibiotics have scarcely been published in the *** order to evaluate the significance of peptaibiotic production for a broader range of potential producers,we screened nine specimens belonging to seven hitherto uninvestigated fungicolous or saprotrophic Trichoderma/Hypocrea species by liquid chromatography coupled to electrospray high resolution mass *** of peptaibiotics found were independently confirmed by analysing the peptaibiome of pure agar cultures obtained by single-ascospore isolation from the *** the nine species examined,five were screened positive for peptaibiotics.A total of 78 peptaibiotics were sequenced,56(=72%)of which are ***,dihydroxyphenylalaninol and O-prenylated tyrosinol,two C-terminal residues,which have not been reported for peptaibiotics before,were found as well as new and recurrent sequences carrying the recently described tyrosinol residue at their *** majority of peptaibiotics sequenced are 18-or 19-residue *** homologies with‘classical representatives’of subfamily 1(SF1)-peptaibiotics argue for the formation of transmembrane ion channels,which are prone to facilitate the producer capture and defence of its substratum.
