A meta-analysis was performed to address the efficacy and safety of paclitaxel-eluting stent(PES) in ST-segment elevation myocardial infarction(STEMI) patients.A systematic literature search was conducted to identify all randomized clinical trials in mortality,recurrent myocardial infarction(rMI),repeat revascularization(RR),and stent thrombosis(ST).A total of 4190 STEMI patients were enrolled in six randomized trials comparing PES with bare-metal stent(BMS).The pooled repeat revascularization rate was 5.7% in PES group,significantly lower than 10.0% in BMS group with an odds ratio(OR) of 0.56,95% confidence interval(CI) [0.44,0.72](P<0.00001).No significant difference was found between PES and BMS groups in mortality at one year after the indexing procedure(3.9% vs.5.1%,OR 0.88,95% CI [0.63,1.21],P=0.42).Similarly,rMI rate did not differ significantly between the two groups(3.4% vs.4.1%,OR 0.80,95% CI [0.56,1.13],P=0.21).PES was also associated with the comparable pooled rate of definite stent thrombosis with BMS(2.3% vs.2.4%,OR 0.81,95% CI [0.52,1.26],P=0.35).The results show that PES improved clinical outcomes in STEMI patients with a decreased need for repeat revascularization and no concerns for safety.

The present study was designed to determine the mechanism underlying the treatment of nephrotic syndrome using astragaloside by observing the effects of astragaloside on the expression of nephrin and podocin proteins and genes in kidneys of rats with adriamycin nephropathy. The rats were injected with adriamycin and, after successful model establishment, randomly divided into a model group, a Methylprednisolone(MP) group, and an astragaloside group. The 24-h complete urine samples were collected. Biochemical indicators were monitored, and kidney tissues were collected for pathological analysis using light microscopy and electron microscopy. The m RNA expression of nephrin and podocin was measured in the kidney tissues using the real-time q PCR, and the protein expression levels of nephrin and podocin were detected using Western blot analysis. At the end of 12 weeks of drug intervention, the urinary protein level was lower in the MP and astragaloside groups than that in the model group(P = 0.008 and P = 0.01, respectively). Serum albumin was higher in the MP and astragaloside groups than in the model group(P < 0.001 and P = 0.012, respectively). Podocytes in the MP group were nearly normal, and fusion of podocytes in the astragaloside group was significantly less than that in the control group. The nephrin and podocin m RNA and protein expression levels in the intervention groups were higher(P < 0.05) than that in the model group. Due to the increased expression of podocyte-related nephrin and podocin proteins, astragaloside maintained slit diaphragm integrity and decreased the level of proteinuria in rats with adriamycin nephropathy.

Genetic and Physiological Analysis of a Novel Type of Interspecific Hybrid Weakness in Rice

Hybrid weakness is an important reproductive barrier that hinders genetic exchange between different species at the post-zygotic stage. However, our understanding of the molecular mechanisms underlying hybrid weakness is limited. In this study, we report discovery of a novel interspecific hybrid weakness in a rice chromosome segment substitution line （CSSL） library derived from a cross between the indica variety Teqing （Oryza sativa） and common wild rice （O. rufipogon）. The dominant Hybrid weakness il （Hwil） gene from wild rice is genetically incompatible with Teqing and induced a set of weakness symptoms, including growth suppression, yield decrease, impaired nutrient absorption, and the retardation of crown root initiation. Phytohormone treatment showed that salicylic acid （SA） could restore the height of plants expressing hybrid weakness, while other phytohormones appear to have little effect. Fine mapping indicated that Hwil is located in a tandem leucine-rich repeat receptor-like kinase （LRR-RLK） gene cluster. Within the 13.2-kb candidate region on the short arm of chromosome 11, there are two annotated LRR-RLK genes, LOC_Osllg07230 and LOC_Osllg07240. The Teqing allele of LOCOsllg07230 and the wild rice allele of LOC_Osllg07240 encode predicted functional proteins. Based on the genetic inheritance of hybrid weakness, LOC_Osllg07240 is implicated as the candidate gene for Hwil. Functional analysis of Hwil will expand our knowledge of the regulation of hybrid weakness in rice.

A scheme for generating cluster states via Raman interaction is proposed. In the scheme, we firstly prepare cluster states of multi-cavities with information encoded in the coherent states and then generate cluster states of multiatoms, which encode the information in the ground states of A-type atoms. The advantages of our scheme are that the atomic spontaneous radiation can be efficiently reduced since the cavity frequency is largely detuned from the atomic transition frequency and the Hadamard gate operation of the coherent states is replaced by measuring the coherent states.

Objective:To explore the potential of metanephric mesenchymal cells(MMCs)for osteogenesis and naringin’s ability to enhance this process and its molecular ***:Porcine MMCs at 70 days of gestation were used as tool cells,cultured in osteogenic induction medium,identified by immunocytochemistry *** potential of porcine MMCs and naringin’s ability to enhance this process was tested by detecting changes in cell viability,alkaline phosphatase(ALP)activity,the expression of runt-related transcription factor 2(Runx2),osteopontin(OPN)and osteocalcin(OCN),and the formation of mineralized nodules,and the application of the p38 signaling pathway inhibitor SB203580 vitiated the osteogenesis-promoting effect of ***:Immunocytochemical staining showed that the cells were Vimentin and Six2(+),E-cadherin and CK-18(−).Naringin can activate the p38 signaling pathway to enhance the osteogenesis of porcine MMCs by increasing cell viability,ALP activity,the expressions of Runx2,OPN and OCN,and the formation of mineralized nodules(P<0.05).The application of p38 signaling pathway inhibitor SB203580 vitiated the osteogenesis-promoting effect of naringin,manifested by decreased ALP activity,the expressions of Runx2,OPN and OCN,and the formation of mineralized nodules(P<0.05).Conclusion:Naringin,the active ingredient of Chinese herbal medicine Rhizoma Drynariae for nourishing Shen(Kidney)and strengthening bone,enhances the osteogenic differentiation of renal MMCs through the p38 signaling pathway.

Background Tissue inhibitor of matrix metaUoproteinase-1 （TIMP-1） is related to the aging of many organs, but few data are available on the change of TIMP-1 in liver aging. The purpose of this study was to investigate the expression and role of TIMP-1, matrix metalloproteinase-2 （MMP-2） and MMP-9 in the process of natural aging in the livers of normal and transgenic mice, and to detect the effects of TIMP-1 on oxidative level and anti-oxidative ability of the livers of transgenic young mice. Methods Normal and transgenic mice were divided into 3 groups according to their age： 3-month-old group （n=5）, 12-month-old group （n=5） and 24-month-old group （n=5）. Histopathological changes of the liver were observed after HE and Masson staining. The messenger RNA （mRNA） levels of TIMP-1, MMP-2 and MMP-9 were determined by semi-quantitative reverse transcriptional polymerase chain reaction; protein expression was measured by Western blot in the livers of normal and transgenic mice of various ages. Changes in levels of superoxide dismutase （SOD）, monoamine oxidase （MAO）, malondialdehyde （MDA） as well as oxidative and anti-oxidative ability were measured. Results Histologically, more fatty degeneration and collagen deposition were found in the aging livers of transgenic mice than in those of the normal mice as their age of months increased. The mRNA and protein expressions of TIMP-1 were significantly high in the oldest animals. The histopathological changes, mRNA and protein expressions of TIMP-1 increased significantly in the liver of transgenic mice as compared with normal mice. The expression of MMP-2 and MMP-9 showed a minor change in the process of aging. Liver change and collagen deposition were not observed in young mice, but the activity of SOD decreased （P〈0.05）, and the activity of MAO （P〈0.01） and the content of MDA increased in the liver of transgenic mice （P〈0.01）. Objective There is a paucity of published works that systematically evaluate gene anomalies or clinical features of patients with renal cysts and diabetes syndrome （RCAD）/maturity onset diabetes of the young type 5 （MODY5）. The purpose of this review was to systematically assess the detection rate, genetic and phenotypic implications of heterozygous autosomal dominant TCF2 anomalies. Data sources MEDLINE database was searched to select articles recorded in English from 1997 to 2008. The focus was monoallelic germline TCF2 gene mutations/deletions. Biallelic inactivation, polymorphisms, DNA modification （hypomethylation and hypermethylation）, loci associated with cancer risk, and somatic TCF2 anomalies were all excluded. Study selection After searching the literature, 50 articles were selected. Results The detection rate of TCF2anomalies was 9.7% and varied considerably among MODY （1.4%）, renal structure anomalies （RSA）（21.4%） and RSA with MODY （41.2%） subgroups. Mutations were strikingly located within the DNA binding domain and varied among exons of the DNA binding domain： exons 2 and 4 were the hottest spots, while mutations were sporadically distributed in exon 3. The consistent phenotypes were RSA （89.6%） and diabetes mellitus （DM）（45.0%）. However, the concurrence of RSA and DM was relatively low （27.5%）, which hinders the optimal performance of genetic testing and obtainment of timely diagnosis. Other organ involvements were complementary and necessary for the early identification of patients with TCF2 anomalies. Analysis of phenotypes of TCF2 point mutations showed significant differences in the detection rates of RSA, impaired renal function （IRF） and DM according to mutation type but not mutation location. Conclusion These valuable features of TCF2 anomalies that previously did not receive sufficient attention should not be neglected.
