Liver angiogenesis as a risk factor for hepatocellularcarcinoma development in hepatitis C virus cirrhotic patients

AIM： To evaluate the predictive value of hepatocyte proliferation and hepatic angiogenesis for the occurrence of Hepatocellular carcinoma （HCC） in hepatitis C virus （HCV） cirrhotic patients.MeTHODS： One hundred-five patients （69 males, 36 females; age range, 51-90 year; median 66 year） with biopsy proven HCV cirrhosis were prospectively monitored for HCC occurrence for a median time of 64 too. Angiogenesis was assessed by using microvessel density （MVD）, hepatocyte turnover by MIB1 and PCNA indexes at inclusion in liver biopsies.ReSULTS： Forty six patients （43.8%） developed HCC after a median time of 55 （6-120） mo while 59 （56.2%） did not. Patients were divided into two groups according to the median value of each index. The difference between patients with low （median MVD = 3; range 0-20） and high （median MVD = 7; range 1-24） MVD was statist（caUy s（gn（ficant （χ^2= 22.06; P 〈 0.0001） wh（ch was not the case for MIB1 or PCNA （MIB-I： χ^2 = 1.41; P = 0.2351; PCNA： χ^2 = 1.27; P = 0.2589）. The median MVD was higher in patients who developed HCC than in those who did not. HOe-free interval was significantly longer in patients with the MVD ≤ 4 （P = 0.0006）. No relationship was found between MIB1 or PCNA and MVD （MIB-1 r^2 = 0.00007116, P = 0.9281; PCNA： P =0.001950; P = 0.6692）. MVD only was able to predict the occurrence of HCC in these patients. Among other known risk factors for HCC, only male sex was statistically associated with an increased risk. CONCLUSION： Liver angiogenesis has a role for in HCV- related liver carcinogenesis and for defining patients at higher risk.

Reverse cholesterol transport (RCT) is a complex process which transfers cholesterol from peripheral cells to the liver for subsequent elimination from the body via feces. Thyroid hormones (THs) affect growth, develop- ment, and metabolism in almost all tissues. THs exert their actions by binding to thyroid hormone receptors (TRs). There are two major subtypes of TRs, TRα and TRβ, and several isoforms (e.g. TRα1, TRα2, TRβ1, and TRβ2). Activation of TRα1 affects heart rate, whereas activation of TRβ1 has positive effects on lipid and lipoprotein metabolism. Consequently, particular interest has been focused on the development of thyromimetic compounds targeting TRβ1, not only because of their ability to lower plasma cholesterol but also due their ability to stimulate RCT, at least in pre-clinical models. In this review we focus on THs, TRs, and on the effects of TRβ1-modulating thyromimetics on RCT in various animal models and in humans.

Iron toxicity resistance strategies in tropical grasses:The role of apoplastic radicular barriers

The revegetation of mined areas poses a great challenge to the iron ore mining *** initial recovery process in degraded areas might rely on the use of Fe-resistant *** grasses, such as Paspalum densum and echinochloa crus-galli, show different resistance strategies to iron toxicity; however, these mechanisms are poorly *** Fe-resistance mechanisms and direct iron toxicity as a function of root apex removal were investigated. To achieve this purpose, both grass species were grown for up to 480 hr in a nutrient solution containing 0.019 or 7 mmol/L Fe-eDTA after the root apices had been removed or maintained. Cultivation in the presence of excess iron-induced leaf bronzing and the formation of iron plaque on the root surfaces of both grass species, but was more significant on those plants whose root apex had been removed. Iron accumulation was higher in the roots, but reached phytotoxic levels in the aerial parts as well. It did not hinder the biosynthesis of chloroplastidic pigments. No significant changes in gas exchange and chlorophyll a fluorescence occurred in either grass when their roots were kept intact; the contrary was true for plants with excised root apices. In both studied grasses, the root apoplastic barriers had an important function in the restriction of iron translocation from the root to the aerial plant parts, especially in e. crus-galli. Root apex removal negatively influenced the iron toxicity resistance mechanisms(tolerance in P. densum and avoidance in e. crus-galli).

Updated overview on interplay between physical exercise,neurotrophins,and cognitive function in humans

The many important benefits of physical exercise also encompass maintenance or improvement of cognitive functions. Among the variousmechanisms underlying the association between physical exercise and brain health, recent evidence attests that neurotrophin receptor signalingmay have an important role, because the activation of this pathway leads to growth and differentiation of new neurons and synapses, supportsaxonal and dendritic growth, fosters synaptic plasticity, and preserves survival of existing neurons. In this review of published evidence, we highlightthat a positive relationship exists between physical exercise and circulating brain-derived neurotrophic factor levels and that the postexercisevariation of this molecule is associated with improvement of neurocognitive functioning. Less clear evidence has instead been published forother neurotrophins, such as nerve growth factor, neurotrophin-3, and neurotrophin-4. Overall, promotion of adequate volumes and intensities ofphysical exercise (i.e., approximately 3 months of moderate-intensity aerobic exercise, with 2—3 sessions/week lasting not less than 30 min)may hence be regarded as an inexpensive and safe strategy for boosting brain-derived neurotrophic factor release, thus preserving or restoringcognitive functions.

MINI-eX:Integrative inference of single-cell gene regulatory networks in plants

Multicellular organisms,such as plants,are characterized by highly specialized and tightly regulated cell populations,establishing specific morphological structures and executing distinct *** regulatory networks(GRNs)describe condition-specific interactions of transcription factors(TFs)regulating the expression of target genes,underpinning these specific *** efficient and validated methods to identify cell-type-specific GRNs from single-cell data in plants are lacking,limiting our understanding of the organization of specific cell types in both model species and crops,we developed MINI-eX(Motif-Informed Network Inference based on single-cell eXpression data),an integrative approach to infer cell-type-specific networks in ***-eX uses single-cell transcriptomic data to define expression-based networks and integrates TF motif information to filter the inferred regulons,resulting in networks with increased ***,regulons are assigned to different cell types,leveraging cell-specific expression,and candidate regulators are prioritized using network centrality measures,functional annotations,and expression *** embedded prioritization strategy offers a unique and efficient means to unravel signaling cascades in specific cell types controlling a biological process of *** demonstrate the stability of MINI-eX toward input data sets with low number of cells and its robustness toward missing data,and show that it infers state-of-the-art networks with a better performance compared with other related single-cell network ***-eX successfully identifies key regulators controlling root development in Arabidopsis and rice,leaf development in Arabidopsis,and ear development in maize,enhancing our understanding of cell-type-specific regulation and unraveling the roles of different regulators controlling the development of specific cell types in plants.

Impacts of Norwegian large carnivore management strategy on national grazing sector

Increasing populations of large carnivores are leading to tension and conflicts with livestock production,a situation that potentially might *** Norway the objective of the large carnivore policy is two-folded:to ensure viable carnivore populations and to secure a sustainable grazing *** main instrument is zonation,with carnivore management zones(CMZs)prioritized for reproduction of the large carnivore species separated from other areas prioritized for grazing *** objective of this paper is to describe current knowledge about the impact of the zoning management strategy on the grazing *** is done by documenting status and changes in sheep production,losses of livestock to predating carnivores,and the use of grazing areas inside and outside the *** protection for lynx,wolverine,bear and wolf cover 55%of the Norwegian mainland.30%of the sheep and 50%of the Sami reindeer grazing areas are found inside the ***(semi-domestic reindeer excluded)is using 59%of the available natural pasture areas outside the CMZs,but only 26%inside the *** lowest use of available grazing areas was found inside zones for wolves(12%)and brown bears(6%).Livestock in these zones are confined to fenced enclosures,mostly on the farm itself,or moved to pastures outside the management zone for summer *** losses increased in the affected regions during the period when carnivores were ***,losses declined when CMZs were established and mitigation efforts were implemented in these *** bulk of sheep and reindeer killed by carnivores are now found in boundary areas within 50 km off the CMZs,where sheep are still grazing on open mountain and forest ***,instruments to protect livestock in areas close to the CMZs are also *** number of sheep declined inside the CMZs from 1999 to 2014,but increased outside the *** reduction in the absolute number of sheep in the CMZs is balanced by
