IL-10 and TNF-α promoter haplotypes are associated with childhood Crohn’s disease location

AIM： To determine the distribution and frequencies of the genotypes and haplotypes of the genes encoding for the glucocorticoid receptor （GR）, the tumor necrosis factor （TNF）-α and the interleukin （IL）-10 in childhood Crohn＇s disease （CD） and to assess the impact of the corresponding DNA variants on clinical and disease phenotypes.METHODS： Ten variants in GR, TNF-a and IL-10 were genotyped in 113 childhood CD cases and 95 healthy subjects, both of French-Canadian origin.RESULTS： For the GR polymorphisms （R23K and N363S） and IL-10 variants in the 5＇flanking region （-1082 G 〉 A, -819 T 〉 C and -592 A 〉 C）, no difference was observed in allele and genotype frequencies between CD patients and controls. At the haplotype level, we found three IL-10 haplotypes previously described in Caucasians （GCC, ACC and ATA） and three novel haplotypes only present in IBD patients. When we analyzed the haplotype distribution with the anatomical location of the disease, the GCC haplotype was associated with the colonic and the ACC haplotype with the terminal ileum location, respectively. The genotyping of five polymorphisms in the promoter region of the TNF-α gene （-1031 T 〉 C, -863 A 〉 C, -857 T 〉 C, -308 A 〉 G and -238 A 〉 G） revealed a significant overrepresentation of homozygous -1031 CC among CD patients （OR = 9.9） and an association with the colonic location. For TNF-α, eleven haplotypes were inferred, including two frequent ones, TCCGG and CACGG, which were significantly observed more frequently in controls and cases, respectively.CONCLUSION： This is one of the first studies investigating the association between haplotype structure and disease location in a CD pediatric cohort. Our results will help to increase our understanding of the genetic determinants of childhood CD.

Flooding an extremely alkaline(pH 10.6) saline soil of the former Lake Texcoco to reduce salinity will affect the soil carbon(C)and nitrogen(N) dynamics.A laboratory incubation experiment was done to investigate how decreasing soil salt content affected dynamics of C and N in an extremely alkaline saline *** soil with electrical conductivity(EC) of 59.2 dS m^(-1) was packed in columns,and then flooded with tap water,drained freely and conditioned aerobically at 50%water holding capacity for a *** process of flooding-drainage-conditioning was repeated eight *** original soil and the soil that had undergone one,two,four and eight flooding-drainage-conditioning cycles were amended with 1000 mg glucose-^(14)C kg^(-1) soil and 200 mg NH_4^+-N kg^(-1)soil,and then incubated for 28 *** CO_2 emissions,soil microbial biomass,and soil ammonium(NE_4^+),nitrite(NO_2^-) and nitrate(NO_3^-) were monitored in the aerobic incubation of 28 *** soil EC decreased from 59.2 to 1.0 dS m^(_1) after eight floodings,and soil pH decreased from 10.6 to *** the added ^(14)C-labelled glucose,only 8%was mineralized in the original soil,while 24%in the soil flooded eight times during the 28-d *** priming effect was on average 278 mg C kg^(-1) soil after the 28-d *** microbial biomass C(mean 66 mg C kg^(-1) soil) did not change with flooding times in the unamended soil,and increased 1.4 times in the glucose-NH_4^+-amended *** immobilization and NO_2^- concentration in the aerobically incubated soil decreased with increasing flooding times,while NO_3^- concentration *** was found that flooding the Texcoco soil decreased the EC sharply,increased mineralization of glucose,stimulated nitrification,and reduced immobilization of inorganic N,but did not affect soil microbial biomass C.

委内瑞拉是一个位于南美洲北海岸的国家,经济资源短缺,并且与世界上一些具有高度影响力的国家的政治关系不稳定,这使得国防部采取极端的保护措施,以避免对该国交通系统最重要的组成部分之一——地铁网络的攻击。本研究的主要目的是寻找提高地铁网络攻击容忍度的可行方法。我们采用复杂网络方法评估加拉加斯地铁网络的鲁棒性,模拟随机和有计划的攻击,并定量测量结构变化以反映攻击的破坏程度。基于仿真结果,我们发现关键节点保护策略可以提高地铁系统的鲁棒性,并且节点12、13、24、31、46和47是关键节点。增加现有节点之间的连通性,以创建一个更具交错性的网络是很有必要的,一旦受到任何类型的干扰（有意的或无意的）,该网络能够维持运行。此外,加强安全系统、协议、设备和人员也很重要,尤其是在那些关键站点。加强与地铁网络相连的地面交通可能是在网络中创建更多环路的另一个策略,这能够提高连通性和鲁棒性。

Non-alcoholic fatty liver disease(NAFLD) is increasing considerably due to the current lifestyle,which means that it is becoming one of the main indications for liver *** the other hand,there is a strong association between NAFLD and cardiovascular *** has been evidenced in many studies revealing a higher presence of carotid plaques or carotid intima-media thickness,leading to cardiovascular events and,ultimately,*** to the liver transplant guidelines,screening for heart disease in transplant candidates should be performed by electrocardiogram and transthoracic echocardiography while a stress echocardiogram should be reserved for those with more than two cardiovascular risk factors or greater than 50 years ***,there are no specific recommendations in NAFLD patients requiring a liver transplantation,despite its well-known cardiovascular risk *** studies have shown that these patients probably require a more exhaustive assessment and a global approach including other specialists such as cardiologists or ***,the incidence of cardiovascular disease is also increased in NAFLD patients in the post-transplantation period in comparison with other etiologies,because of the pre-existent risk factors together with the immunosuppressive ***,an early intervention on the lifestyle and the individualized selection of the immunosuppressive regimen could lead to a modification of the cardiovascular risk factors in NAFLD patients requiring a liver transplantation.

AIM:To evaluates the effectiveness and safety of the first generation,NS3/4A protease inhibitors(PIs) in clinical practice against chronic C virus,especially in patients with advanced fibrosis. METHODS:Prospective study and non-experimental analysis of a multicentre cohort of 38 Spanish hospitals that includes patients with chronic hepatitis C genotype 1,treatment-na?ve(TN) or treatment-experienced(TE),who underwent triple therapy with the first generation NS3/4A protease inhibitors,boceprevir(BOC) and telaprevir(TVR),in combination with pegylated interferon and ribavirin. The patients were treatment in routine practice settings. Data on the study population and on adverse clinical and virologic effects were compiled during the treatment period and during follow ***:One thousand and fifty seven patients were included,405(38%) were treated with BOC and 652(62%) with TVR. Of this total,30%(n = 319) were TN and the remaining were TE:28%(n = 298) relapsers,12%(n = 123) partial responders(PR),25%(n = 260) null-responders(NR) and for 5%(n = 57) with prior response unknown. The rate of sustained virologic response(SVR) by intention-to-treatment(ITT) was greater in those treated with TVR(65%) than in those treated with BOC(52%)(P < 0.0001),whereas by modified intention-to-treatment(m ITT) no were found significant differences. By degree of fibrosis,56% of patients were F4 and the highest SVR rates were recorded in the non-F4 patients,both TN and TE. In the analysis by groups,the TN patients treated with TVR by ITT showed a higher SVR(P = 0.005). However,by m ITT there were no significant differences between BOC and TVR. In the multivariate analysis by m ITT,the significant SVR factors were relapsers,IL28 B CC and non-F4; the type of treatment(BOC or TVR) was not significant. The lowest SVR values were presented by the F4-NR patients,treated with BOC(46%) or with TVR(45%). 28% of the patients interrupted the treatment,mainly by non-viral response(51%):this outcome was
