Me.hanical force. play critical role. in the.de.e.opme.t and re.ode.ing proce.se. of bone. As an alte.native.ce.l source.for bone.e.gine.ring, adipose.de.ive. ste. ce.ls （ASCs） should be.fully inve.tigate. for the.r re.ponse. to me.hanical stre.s. Similarly, the.oste.ge.ic pote.tial, stimulate. by me.hanical stre.s, should be.compare. with bone.marrow stromal ce.ls （BMSCs）, which have.be.n clinically use. for bone.tissue.e.gine.ring. In this study, ASCs and BMSCs we.e.oste.ge.ic-induce. for 48 hours, and the. subje.te. to uniaxial me.hanical stre.ching for 2 or 6 hours. Ce.l orie.tation, oste.ge.ic re.ulatory ge.e., oste.ge.ic ge.e. and ALP activitie. we.e.me.sure. and compare. be.we.n ASCs and BMSCs. ASCs could align in a pe.pe.dicular way to the.dire.tion of stre.ching stre.s, while.BMSCs did not pre.e.t a spe.ific alignme.t. Both 2 and 6 hours me.hanical stre.ching could e.hance.the.mRNA e.pre.sion of Osx and Runx2 in BMSCs and ASCs, while.OCN mRNA only incre.se. in ASCs afte. 6 hours me.hanical loading. Me.hanical stre.ching e.hance. the.BMP-2 mRNA e.pre.sion in ASCs, while.only afte. 6 hours of me.hanical loading significantly incre.se. the.BMP-2 ge.e.e.pre.sion in BMSCs. Significant diffe.e.ce. only e.ist be.we.n ASCs and BMSCs loade. at 2 hours of me.hanical stre.ching. It is conclude. that ASCs are.more.rapid re.ponde.s to me.hanical stre.s, and have.gre.te. pote.tial than BMSCs in oste.ge.e.is whe. stimulate. by me.hanical stre.ching, indicating the.r use.ulne.s for bone.study in a rat mode..

Me.hodology Human tongue.carcinoma Tca8113 ce.ls we.e.culture. with DAPT. Ce.l growth was de.e.mine. using Indigotic Re.uction me.hod. The.ce.l cycle.and apoptosis we.e.analyze. by flow cytome.ry. Re.l-time.PCR and Immuno-Fluore.ce.ce.（IF） we.e.e.ploye. to de.e.mine.the.intrace.lular e.pre.sion le.e.s. Re.ults DAPT inhibite. the.growth of human tongue.carcinoma Tca8113 ce.ls by inducing G0-G1 ce.l cycle.arre.t and apoptosis, The.mRNA le.e.s of Hairy/e.hance. of Split-1 （He.-1）, a targe. of Notch activation, we.e.re.uce. by DAPT in a dose.de.e.de.t manne.. Coincide.t with this obse.vation, DAPT induce. a dose.de.e.de.t promotion of constitutive.Caspase.3 in Tca8113 ce.ls. Conclusion DAPT may have.a the.ape.tic value.for human tongue.carcinoma. More.ve., the.e.fe.ts of DAPT in tumor inhibition may arise.partly via the.modulation of Notch- 1 and Caspase.3.

Me.hodology DPSCs we.e.harve.te. from the.de.tal pulp tissue.of transge.ic nude.mice. and the. transfe.re. to oste.ge.ic, adipoge.ic, and chondroge.ic me.ia. The.morphological characte.ization of induce. ce.ls was obse.ve. by microscopy and histological staining. The.e.pre.sion of marke. ge.e. was me.sure. by RT-PCR. Re.ults The.e.doge.ous GFP and multiline.ge.pote.tial of transge.ic DPSCs had no influe.ce.on e.ch othe.. More.ve., the.re.ults of fluore.ce.ce.microscopic imaging sugge.t that the.e.was no significant de.line.of GFP e.pre.sion during DPSCs diffe.e.tiation. Conclusion As the.population of GFP labe.e. In the.mode. organism Drosophila me.anogaste., we.have.atte.pte. 1) to inhibit the.sole.Drosophila homologue.of foxo through the.dire.te. e.pre.sion of a stable.inducible.RNAi transge.e.and 2) to indire.tly incre.se.foxo transcription activity through the.inhibition of the.kinase.minibrain (mnb), a foxo transcriptional inhibitor. To e.aluate.the.life.ime.conse.ue.ce. upon the.flie., longe.ity assays and locomotion ove. time.assays we.e.conducte..
