对顺铂和紫杉醇耐药的卵巢上皮癌患者采用Adp53(INGN201;AdVEXIN)治疗的Ⅰ期研究

Purpose. Adenoviral p53 (Adp53) is a replication-deficient adenovirus contain ing human p53 cdNA. This phase I study was designed as a toxicity study of multi ple dosing of Adp53 administered by intraperitoneal (IP) delivery to patients wi th ovarian cancer. Experimental design. Eligibility criteria included patients w ith platinum-and paclitaxel-resistant metastatic epithelial ovarian cancer; a Zubrod performance status of 0, 1, or 2; and adequate bone marrow, liver, and re nal function. Patients underwent laparoscopy, washings, biopsies, and placement of an IP catheter within 10 days of Adp53 administration. Adp53 was given daily for 5 days every 3 weeks at one of the following four dose levels: 3 ×1010, 3 ×1011, 1 ×1012, or 3 ×1012 viral particles (vp). Results. Seventeen patients were enrolled in the trial. Fifteen (88%) patients are evaluable for toxicity. The mean age of the study group was 51 years (range 32-67). All but one patient received two or more chemotherapy regimens before study entry. No doselimiting toxicities (dLT) were observed. Grade 3 toxicities included fatigue (six patient s), fever (two patients), chills (one patient), abdominal pain (three patients), nausea (two patients), and sinus congestion (one patient). One patient had Grad e 3 edema and head.c.e. There were no hematologic toxicities. Eleven patients (6 5%) are evaluable for response. Two of 17 patients (12%) had a mixed response. Four patients (24%) had stable disease for up to four courses. Five patients ( 29%) had progressive disease after one to two courses. conclusions. Multiple do sing of IP Adp53 was well tolerated in this group of heavily pretreated patients ; however, the dosing schedule and the amount cannot be concluded from this stud y. With a negative randomized trial of ovarian cancer in front-line treatment t hat included an adenovirus p53 plus chemotherapy, we feel that further refinemen t of gene therapy is required before additional trials are undertaken. The levered-dual response(LEdAR)coulomb-damped system attains near resonant vibration isolation by differential preloads/offsets in linear *** takes the advantages of both the preloads/offsets in linear springs and the guiderail friction for realizing different levels of vibration *** isolation capacities are investigated on the strategies with both the horizontal and vertical guiderails,with the horizontal rail only,and without *** compressive preloads generally result in the consumption of most of the initial excitation energy so as to overcome the potential *** isolation onsets at the frequency ratio of 1∓0.095 on the left-hand side(LHS)and the right-hand side(RHS)of the lever are relative to the load plate *** observed near resonant isolation thus makes the LEdAR system a candidate for the isolation of the mechanical systems about resonance while opening a path for simultaneous harvesterisolation functions and passive functions at extreme frequencies.

dear Editor,This letter is concerned with prescribed-time Nash equilibrium(PTNE)seeking problem in a pursuit-evasion game(PEG)involving agents with second-order *** order to achieve the prior-given and user-defined.c.nvergence time for the PEG,a PTNE seeking algorithm has been developed to facilitate collaboration among multiple pursuers for capturing the evader without the need for any global ***,it is theoretically proved that the prescribedtime convergence of the designed algorithm for achieving Nash equilibrium of ***,the effectiveness of the PTNE method was validated by numerical simulation results.A PEG consists of two groups of agents:evaders and *** pursuers aim to capture the evaders through cooperative efforts,while the evaders strive to evade *** is a classic noncooperative *** has attracted plenty of attention due to its wide application scenarios,such as smart grids[1],formation control[2],[3],and spacecraft rendezvous[4].It is noteworthy that most previous research on seeking the Nash equilibrium of the game,where no agent has an incentive to change its actions,has focused on asymptotic and exponential convergence[5]-[7].

Objective. To determine the safety and efficacy of a novel illudin S derivative, irofulven(MGI- 114), in patients with recurrent ovarian cancer who had received extensive prior chemotherapy. Methods. The trial was an open label phase II study. Patients initially enrolled in this study were treated every 14 days with a dose of 24 mg/m2. Unexpected retinal toxicity associated with this dose and schedule lead to modification of the dosing to 0.55 mg/kg on the same schedule with a maximum individual dose of 50 mg. dose red.c.ions were permitted based on both hematologic and non- hematologic toxicities. Results. Seventy- four women were accrued and stratified into two cohorts including 58 women with platinum- resistant disease and 16 with platinum- sensitive disease. Non- hematologic toxicities included nausea, vomiting, and fatigue. Thirty- one women had between one and six visual symptoms, most were Grade 1 and 2 in nature. The majority of visual toxicities resolved either during treatment or post- treatment with irofulven. There was one partial response in each cohort with 19 (33% ) and 8 (50% ) of women having stable disease in the platinum- resistant and platinum- sensitive cohorts, respectively. conclusions. Irofulven at 24 mg/m2 on every 14- day schedule is associated with significant retinal toxicity in this patient population. dosing at 0.55 mg/kg has persistent retinal toxicity, yet demonstrated only limited anti- tumor activity in a population of women who had received extensive prior chemotherapy.

Objectives. To evaluate the efficacy and tolerability of ima-tinib mesylate (Gleevec; Novartis Pharmaceuticals, Basel, Switzerland) in patients with recurrent ovarian and primary peritoneal cancer. Methods. This was an open- label, single- institution phase II trial. Patients were eligible if they had measurable platinum/taxane- resistant disease, received 2- 4 prior treatment regimens, and over- expressed at least one imatinib target (c- Kit, PdGFR- β , or c- Abl) by immunohistochemistry. Imatinib was administered orally at 600 mg daily for 6 weeks (one course)- and was repeated in the absence of measurable progre-ssion. Results. Sixteen enrolled patients were evaluable for toxicity and 12 for response. The median number of prior treatments was 4. A total of 29 courses were initiated. No complete or partial responses were d.c.mented during a median follow- up of 6.6 months. However, 4 (33% ) of the 12 evaluable patients had stable disease lasting 3.8, 6.4, 7.5, and 8+ months. Expression of PdGFR- β and.c. Abl was seen in 15 (94% ) and.c. Kit in 8 (50% ) patients’ tumors. There was no relationship between best response (stable disease) and target expression. Adverse events were uncommon,with fatigue and nausea/vomiting being reported in 34% and 31% of cycles, respectively. Two patients underwent dose red.c.ion for rash and edema (n = 1) and grade 3 neutropenia (n = 1). No grade 4 toxicity was observed. conclusion. Imatinib mesylate was well tolerated but did not prod.c. clinical responses in patients with previously treated metastatic ovarian and primary peritoneal carcinoma.

How can Practitioners Support citizen Volunteers in disaster Risk Red.c.ion? Insight from “Good and Ready” in Aotearoa New Zealand

Global and national policy frameworks emphasize the importance of people's participation and volunteers' role in disaster risk red.c.ion. While research has extensively focused on volunteers in disaster response and recovery, less attention has been paid on how organizations involved in disaster risk management can support volunteers in leading and.c.ordinating communitybased disaster risk red.c.ion. In 2019, the New Zealand Red.c.oss piloted the Good and Ready initiative in Auckland,Aotearoa New Zealand, with the objective to empower local people in resilience building with a focus on volunteers and.c.mmunity participation. This research examined the positive and negative outcomes of Good and Ready and investigated volunteers' experiences in the disaster resilience initiative. It involved the codesign of a questionnaire-based survey using participatory methods with Good and Ready volunteers, the dissemination of the survey to gather volunteers' viewpoints, and a focus group discussion with participatory activities with Red.c.oss volunteers. The findings highlight that a key challenge lies in finding a balance between a program that provides flexibility to address contextual issues and fosters communities' ownership, versus a prescriptive and standardized approach that leaves little room for creativity and self-initiative. It pinpoints that supporting volunteers with technical training is critical but that soft skills training such as coordinating, communicating,or facilitating activities at the local level are needed. It concludes that the sustainability of Good and Ready requires understanding and meeting volunteers' motivations and expectations and that enhancing partnerships with local emergency management agencies would strengthen the program.

Polio erad.c.tion surveillance in Sri Lanka,2019-2023

Objective:To evaluate the polio laboratory surveillance carried out from January,2019 to May,2023 by the Polio Regional Reference Laboratory,Sri ***:This retrospective study analyzed all stool samples received under the acute flaccid paralysis(AFP)and immunodeficient vaccine-derived poliovirus(VdPV)surveillance at Polio Regional Reference Laboratory,Sri Lanka from January,2019 to May,*** results of the testing methodologies were extracted from the laboratory data system,i.e.,poliovirus virus isolation,intra-typic differentiation/VdPV real time reverse transcriptase polymerase chain reaction(ITd/VdPV rRTPcR)and sequencing,along with the data on timing of reporting results,stool adequacy and *** was analyzed using descriptive ***:A total of 2141 stool samples from 1644 cases were received for AFP surveillance from Sri Lanka(93.61%),Maldives(1.52%),and immunodeficient VdPV(4.86%)*** polioviruses(19/1644,1.15%)and non-polio enteroviruses(73/1644,4.44%)were isolated,while Sabin-like 3 virus was detected in majority(12/19,63.15%)among the poliovirus *** polioviruses or circulating VdPVs were not detected among the *** all years of the study,the non-polio AFP detection rate was>1/100000 in children aged less than 15 years,whereas stool adequacy rate was>80%.All results were reported within 14 days of receipt,ensuring timely reporting as per global ***:The Polio Regional Reference Laboratory,Sri Lanka plays a vital role in maintaining the polio-free status in the country through its robust laboratory surveillance,while adhering to the surveillance ***-detection of wild polioviruses and.c.rculating VdPV during the study period reinforces the polio-free status in the country.

This work describes the use of TiO_(2)nanotubes-based electrodes(TNT)modified with cu_(2)O nanostructures and gold nanoparticles for the photoelectrored.c.ion of cO_(2)to prod.c. value-added.c.mpounds.A thin layer of polydopamine was used as both an adherent agent and an electron transfer mediator,due to itsπ-conjugated electron *** highest prod.c.ion yield was achieved using a TNT@PdA/Nc/Au40%electrode,with Faradaic efficiencies of 47.4%(110.5μM cm^(-2))and 27.8%(50.4μM cm^(-2))for methanol and methane,*** performance of the photoelectrodes was shown to be cu_(2)O facet-dependent,with cubic structures leading to greater conversion of cO_(2)to methanol(43%)and methane(27%),compared to the octahedral morphology,while a higher percentage of metallic gold on the nanostructured.c._(2)O surface was mainly important for *** functional theory(dFT)calculations supported these findings,attributing the superior photoelectrocatalytic performance of the TNT@PdA/Nc/Au40%electrode for cH4generation to the formation of an OcH3intermediate bonded to Au *** using isotope-labeling and analysis by gas chromatograph-mass(Gc-MS)demonstrated that13cO_(2)was the source for photoelectrocatalytic generation of13cH3OH and13cH313cH2OH.

亚里士多德确实有一种关于恶的理论。善在定义上优先于恶,这是由善所需要的本体论边界决定的。如果善实际上就是卓越的存在,那么恶必定是这种存在的缺乏。在实体范畴中,亚里士多德的善是一种活的存在,在对自身的沉思中,第一性的神获得了善并且是幸福的。由此类推,如果存在一种实体范畴上的恶,那么它必定是一种能对自身加以沉思的活的存在。然而在亚里士多德勾勒的事物体系中,这种观念是无法实现的。亚里士多德的善的理论排除了把一种恶(即实体性的巨大的恶)等同于善的可能性。这种理论认真对待伦理上的恶,把它当作心理学、政治学问题,而不是当作本体论问题。无条件的恶说的不是理性的神灵,而是心理变态之人或反社会之人,说的是一位恶人,尽管这个人处在一种特定的社会,即良好的社会之中。
