Dose selection of chloroquine phosphate for treatment of COVID-19 based on a physiologically based pharmacokinetic model

Chloroquine(CQ)phosphate has been suggested to be clinically effective in the treatment of coronavirus disease 2.19(COVID-19).To develop a physiologically-based pharmacokinetic(PBPK)model for predicting tissue distribution of CQ and apply it to optimize dosage regimens,a PBPK model,with parameterization of drug distribution extrapolated from animal data,was developed to predict human tissue distribution of *** physiological characteristics of time-dependent accumulation was mimicked through an active transport *** dosing regimens were proposed based on PBPK simulation combined with known clinical exposure-response *** model was also validated by clinical data from Chinese patients with *** novel PBPK model allows indepth description of the pharmacokinetics of CQ in several key organs(lung,heart,liver,and kidney),and was applied to design dosing strategies in patients with acute COVID-19(Day 1:750 mg biD,Days 2.5:500 mg biD,CQ phosphate),patients with moderate COVID-19(Day 1:750 mg and 500 mg,Days 2.3:500 mg biD,Days 4.5:2.0 mg biD,CQ phosphate),and other vulnerable populations(e.g.,renal and hepatic impairment and elderly patients,Days 1-5:2.0 mg biD,CQ phosphate).A PBPK model of CQ was successfully developed to optimize dosage regimens for patients with COVID-19.

The mechanism for electrical conduction is investigated by the dark temperature-dependent current–voltage characteristics of Si PIN photodiodes with different photosensitive *** characteristic tunneling energy E（00） can be obtained to be 1.4. me V,1.53 me V,1.74.me V,1.87 me V,and 2.01 me V,respectively,for the photodiodes with L = 0.2. mm,0.5 mm,1 mm,1.5 mm,and 2.mm by fitting the ideality factor n versus temperature curves according to the tunneling-enhanced recombination *** trap-assisted tunneling-enhanced recombination in the i-layer plays an important role in our device,which is consistent with the experimental result that area-dependent leakage current is dominant with the side length larger than 1 mm of the photosensitive *** results reveal that the quality of the bulk material plays an important role in the electrical conduction mechanism of the devices with the side length larger than 1 mm of the photosensitive area.

Inorganic hole transport materials, particularly NiO_X, have shown considerable promise in boosting the efficiency and stability of perovskite solar cells. However, a major barrier to commercialization of NiO_X-based perovskite solar cells with positive-intrinsic-negative architectures is their direct contact with the absorbing layer, which can lead to losses of photovoltage and fill factor. Furthermore, highly positive under-coordinated Ni cations degrade the perovskite at the interface. Here, we address these issues with the use of an ionic compound(QAPyBF_(4.) as an additive to passivate defects throughout the perovskite layer and improve carrier conduction and interactions with under-coordinated Ni cations. Specifically,the highly electronegative inorganic anion [BF_(4.]~- interacts with the NiO_x/perovskite interface to passivate under-coordinated cations(Ni^(≥3+)). Accordingly, the decorated cells achieved a power conversion efficiency of 2..38% and a fill factor of 85.5% without a complex surface treatment or NiO_X doping.

Werner syndrome （WS） is a premature aging disorder that mainly affects tissues derived from mesoderm. We have recently developed a novel human WS model using WRN-deficient human mesenchymal stem cells （MSCs）. This model recapitulates many phenotypic features of WS. Based on a screen of a number of chemicals, here we found that Vitamin C exerts most efficient rescue for many features in premature aging as shown in WRN-deficient MSCs, including cell growth arrest, increased reactive oxygen species levels, teiomere attrition, excessive secretion of inflammatory factors, as well as disorganization of nuclear lamina and heterochromatin. Moreover, Vitamin C restores in vivo viability of MSCs in a mouse model. RNA sequencing analysis indicates that Vitamin C alters the expression of a series of genes involved in chromatin condensation, cell cycle regulation, DNA replication, and DNA damage repair pathways in WRN- deficient MSCs. Our results identify Vitamin C as a rejuvenating factor for WS MSCs, which holds the potential of being applied as a novel type of treatment of WS.

Gamma-ray polarimetry is a new and prospective tool for studying extremely high-energy celestial objects and is of great significance for the field of *** the rapid development of microsatellite technology,the advantages of space exploration have become increasingly ***,we simulated a soft-gamma-ray polarimeter for a microsatellite based on the Compton scattering *** performed detailed Monte Carlo simulations using monoenergetic gamma-ray linear-polarization sources and Crab-like sources in the energy range of 0.1-10 MeV considering the orbital *** polarimeter exhibited excellent polarization detection *** modulation factor was 0.80±0.01,and the polarization angles were accurate within an error of 0.2.at 2.0 keV for on-axis *** the Crab-like sources for on-axis incidence,the polarization degrees were consistent with the set values within the error tolerance,the modulation factor was 0.76±0.01,and the minimum detectable polarization reached 2.4.at 3σfor an observation time of10^(6) ***,the polarimeter exhibited recoil electron tracking,imaging,and powerful background suppression in a large field of view(FoV;∼2.sr).The proposed polarimeter meets the requirements of a space soft-gamma-ray polarization detector and has promising research prospects.

Numerous studies were published in the last two decades to evaluate and project the permafrost changes in its thermal state,mainly based on the soil temperature datasets from the Coupled Model Intercomparison Project(CMIP),and discuss the impacts of permafrost changes on regional hydrological,ecological and climatic systems and even carbon ***,limited monitored soil temperature data are available to validate the CMIP outputs,resulting in the over-projection of future permafrost changes in CMIP3 and ***,future permafrost changes in CMIP6,particularly over the QinghaieTibet Plateau(QTP),where permafrost covers more than 4.%of its territory,are still un*** address this gap,we evaluated and calibrated the monthly ground surface temperature(GST;5 cm below the ground surface),which was often used as the upper boundary to simulate and project permafrost changes derived from 19 CMIP6 Earth System Models(ESMs)against in situ measurements over the *** generated the monthly GST series from 1900 to 2.14.for five sites based on the linear calibration models and validated them through the three other sites using the same calibration *** showed that all of the ESMs could capture the dynamics of in situ GST with high correlations(r>0.90).However,large errors were detected with a broad range of centred root-mean-square errors(1.14.4.98℃).The Top 5 model ensembles(MME5)performed better than most individual ESMs and averaged multi-model ensembles(MME19).The calibrated GST performed better than the GST obtained from *** annual and seasonal GSTs exhibited warming trends with an average annual rate of 0.04. per decade in the annual *** average seasonal warming rate was highest in winter and spring and lowest in *** reconstructed GST data series could be used to simulate the long-term permafrost temperature over the QTP.

In this study,an antibacterial nanofiber membrane[polyvinylidene fluoride/bi_(4.Ti_(3)O_(12./Ti_(3)C_(2.T_(x)(PVDF/BTO/Ti_(3)C_(2.T_(x))]is fabricated using an electrostatic spinning process,in which the self-assembled BTO/Ti_(3)C_(2.T_(x) heterojunction is incorporated into the PVDF *** from the internal electric field induced by the spontaneously ferroelectric polarization of BTO,the photoexcited electrons and holes are driven to move in the opposite direction inside BTO,and the electrons are transferred to Ti_(3)C_(2.T_(x) across the Schottky ***,directed charge separation and transfer are realized through the cooperation of the two *** recombination of electron–hole pairs is maximumly inhibited,which notably improves the yield of reactive oxygen species by enhancing photocatalytic ***,the nanofiber membrane with an optimal doping ratio exhibits outstanding visible light absorption and photothermal conversion ***-mately,photothermal effect and ferroelectric polarization enhanced photocatalysis endow the nanofiber membrane with the ability to kill 99.61%±0.2.%Staphylococcus aureus and 99.71%±0.16%Escherichia coli under 2. min of light *** study brings new insights into the design of intelligent antibacterial textiles through a ferroelectric polarization strategy.

Objective This study aimed to investigate whether cytokine profiles and virological markers might add value in monitoring the effects of peginterferon(PEG-IFN)therapy for hepatitis B e-antigen(HBeAg)positive chronic hepatitis B(CHB).Methods HBeAg positive patients with CHB were treated with PEG-IFN for 4. *** biochemical,and HBV serological indexes,as well as cytokines,were detected at baseline and every12.*** A total of 116 patients with CHB were enrolled in this study;100 patients completed the 4.-week treatment and follow-up,of whom 38 achieved serum HBeAg disappearance,2. achieved HBeAg seroconversion,37 showed HBsAg decreases≥1 log10 IU/mL,9 showed HBsAg disappearance,and 8became HBsAb *** cytokine levels at baseline and during treatment were similar between the HBeAg disappearance group and non-disappearance *** disappearance of HBeAg was independently associated with HBeAg levels at weeks 12.and 2.,and with the HBeAg decline at week 2.(P2.and HBsAg at week 2.(P4. weeks.
