Differential expression of cell cycle regulators in HCV-infection and related hepatocellular carcinoma

AIM:To investigate cell cycle proteins in chronic hepatitis C virus infection in order to analyze their role in the process of hepatocyte transformation and to characterize their prognostic properties. MeTHODS:Subjects of the current study included 50 cases of chronic hepatitis C(CHC) without cirrhosis,30 cases of CHC with liver cirrhosis(LC) ,and 30 cases of hepatitis C-related hepatocellular carcinoma(HCC) admitted to the Department of Hepato-Gastroenterology,Theodor Bilharz Research Institute(TBRI) ,Giza,*** wedge liver biopsies,taken during laparoscopic cholecystectomy,were also included as normal *** investigations including urine and stool analysis,liver function tests and prothrombin concentration;serologic markers for viral hepatitis and ultrasonography were done for all cases of the study together with immunohistochemical analysis using primary antibodies against Cyclin D1,Cyclin e,p21,p27 and Rb/p105 proteins. ReSULTS:Normal wedge liver biopsies didn't express Cyclin e or Rb/p105 immunostaining but show positive staining for Cyclin D1,p21 and *** D1 expressed nuclear staining that was sequentially increased from CHC to LC(Pes;meanwhile,Cyclin e revealed nuclear positivity only in the case of HCCs patients that was directly correlated to Rb/p105 *** expression of p21 and p27 was significantly increased in CHC and LC cases compared to normal controls and HCCs with no significant difference between well-and poorlydifferentiated tumors.p21 showed only a nuclear pattern of staining,while,p27 presented with either cytoplasmic and/or nuclear reactivity in all studied *** analysis revealed a direct relation between Cyclin D1 and p21 in CHC cases(Petween Cyclin D1 and Cyclin e in HCCs(Pever,an inverserelationship was detected between Cyclin D1 and p21 or p27(Petween p21 and Rb/p105(Pegulation of Cyclin D1 in CHC p

Molecular and serological prevalence of Toxoplasma gondii in pregnant women and sheep in egypt

Objective: To investigate molecular and serological prevalence of Toxoplasma gondii(***) in pregnant women and sheep in ***: Blood samples collected from healthy 364 pregnant women and 170 sheep were investigated for *** antibodies and parasitemia using highly specific and sensitive surface antigen(TgSAG2) based enzyme linked immunosorbent assay(elISA)and real time-polymerase chain reaction(RT-PCR).Results: Overall prevalence of *** was 51.76%, 17.65% in sheep, 33.79%, 11.81%in pregnant women, using elISA and RT-PCR *** differences in *** prevalence were observed on the basis of contact with cats or soil in pregnant women using either RT-PCR or *** pregnant women, a significant increase was detected in aged and those eating under-cooked mutton using simultaneous elISA/***: Consumption of under-cooked infected mutton is an important source of human infection and the combination of the two assays provide accurate and precise data during infection.

Objective:To investigate the changes in total phenols,flavonoids,tannins,vitamin e, β-carotene and antioxidant activity during soaking of three white sorghum ***: The changes in total phenols,total ilavonoids,tannins,phenolic acids compounds,flavonoid components,vitamin e,P-carotene and antioxidant activity during soaking of sorghum grains were ***:Total phenols,total flavonoids,tannins,vitamin e,P-carotene and antioxidant activity in raw sorghum were ranged from 109.21 to 116.70,45.91 to 54.69,1.39 to 21.79 mg/100 g,1.74 to 5.25,0.54 to 1.19 mg/kg and 21.72%to 27.69%and 25.29%to 31.97%,respectively. The above measured compounds were significantly decreased after soaking.p-Hydroxybenzoic acid,vanillic acid,syringic acid and cinnamic acid represent the major phenolic acids in Dorado *** ferulic acid,p-coumaric acid,gallic acid and caffeic acid represent the major phenolic acids in *** the other hand,protocatechuic acid represents the major phenolic acids in *** flavonoids components,Dorado was the highest variety in kampferol and naringenin while Shandaweel-6 was the highest variety in luteolin, apigenin,hypersoid,quercelin and ***,Giza-15 was the highest variety in catechin. Phenolic acids,flavonoid compounds and antioxidant activities were decreased after soaking. Conclusions:Sorghum varieties have moderate quantities from total phenols,total flavonoids, tannins,phenolic acids compounds,flavonoid components,vitamin e,P-carotene and antioxidant activity which decreased after soaking.

Role of Immunostaining in Detecting extra-Pattern and Subtle Lymphomatous Infiltration in Bone Marrow Biopsies of NHL Patients

Introduction: Immunohistochemistry (IHC) enables the examination of a greater number of trephine biopsy levels and is helpful in determining additional scattered malignant cells. The aim of this study is to detect extra-pattern and subtle lymphomatous infiltration in bone marrow biopsies using CD20 and CD3 immunostaining. Patients and Methods: This study was conducted on 100 newly diagnosed Non Hodgkin Lymphoma (NHL) patients. Their bone marrow trephine biopsies were assessed on routine histology [Hematoxylin and eosin (H & e)], and were further subjected to IHC using CD20 and CD3. Results: Pattern of involvement by H & e was highlighted by IHC. It showed additional interstitial pattern in 9 cases, parasinusoidal streaks in one case and highlighted a patchy pattern in another case with interstitial involvement on H & e. IHC also detected subtle infiltrations on additional 5.5% cases compared with histology alone. It helped in differentiating reactive (12 cases) and malignant lymphoid infiltration (33 cases). Conclusion: CD20 and CD3 immunostaining performed routinely on bone marrow trephine biopsies has the ability to reveal extra-pattern of infiltration and improve detection of subtle lymphoid involvement. A combined procedure identifying several distinctive features, in particular histotopography and IHC, provides a promising way of discriminating reactive from neoplastic lymphoid infiltrates in bone marrow trephine biopsies.

Harnessing of Chemically Modified Rice Straw Plant Waste as Unique Adsorbent for Reducing Organic and Inorganic Pollutants

Conversion of rice straw (RS) as one of agricultural plant wastes (about 45% of the volume of rice production) to valuable industrial product was achieved, by grafting different amounts of dimethylaminoethyl methacrylate (DMAeM) on it using potassium permanganate/nitric acid redox system. This was done to obtain six levels of poly (DMAeM)—rice straw graft copolymers (PDMAeMRS) having different graft yields (expressed as N%) with increasing order and designated as (PDMAeMRS 1 to PDMAeMRS 6). The latter copolymers were dispersed in aqueous solution of heavy metal ions Cu (II) ions and filtered to form rice straw co-polymer—metal ions complex. Different factors affecting the heavy metal ions removal such as pH, extent of grafting, treatment time and rice straw dose were studied in detail. It was found from the obtained results that;the residual metal ions removal from their aqueous solutions increased with 1) increasing the extent of grafting of PDMAeMRS i.e. from PDMAeMRS 1 to PDMAeMRS 6;2) increasing the pH of the metal ions solution complex from 1 to 8;3) increasing the rice straw dosage from 0.50 to 2.0 g, then leveled off thereafter;4) increasing the time of the reaction up to 20 minute then leveled off after that. On the other hand, Pb (II), Cd (II) and Hg (II) ions were also removed from their solutions with different extent. Furthermore, the prepared co-polymer could be recovered by washing the metal ions from the complex with weak acid 1 N HNO3 (pH 2) and the metal-binding activity of the rice straw was slightly reduced by this process. Finally, the ability of PDMAeMRS to remove three types of acid dyes from their solutions was also reported.

Vibrational, rotational, and triaxiality features in extended O(6) dynamical symmetry of IBM using three-body interactions

The shape transition between the vibrational U(5)and deformed c-unstable O(6)dynamical symmetries of sd interacting boson model has been investigated by considering a modified O(6)Hamiltonian,providing that the coefficients of the Casimir operator of O(5)are N-dependent,where N is the total number of *** modified O(6)Hamiltonian does not contain the number operator of the d boson,which is responsible for the vibrational *** addition,the deformation features can be achieved without using the SU(3)limit by adding to the O(6)dynamical symmetry the three-body interaction[QQQ]^(0),where Q is the O(6)symmetric quadrupole ***,triaxiality can be generated through the inclusion of the cubic d-boson interaction[d+d+d+]^(3)·∣ddd∣^(3).The classical limit of the potential energy surface(PeS),which represents the expected value of the total Hamiltonian in a coherent state,is studied and *** modified O(6)model is applied to the even–even^124-132 Xe *** parameters for the Hamiltonian and the PeSs are calculated using a simulated search program to obtain the minimum root mean square deviation between the calculated and experimental excitation energies and B(e2)values for a number of low-lying levels.A good agreement between the calculations and experiment results is found.

Performance enhancement of Discrete Multi-Tone Systems with a Trigonometric Transform

The sine transform can be used as a tool to conquer the problems of discrete multi-tone (DMT) systems to increase the bit rate. In the proposed discrete sine transform based discrete multi-tone (DST-DMT) system, we make use of the energy compaction property of the DST to reduce the channel effects on the transmitted signals. The mathematical model of the proposed DST system is presented in the paper. Simulation experiments have been carried out to test the effect of the proposed DST-DMT system. The results of these experiments show that the performance of the DST-DMT system is better than that of the traditional FFT-DMT system. The results also show that employing the proposed TeQ in the DST-DMT system can increase the bit rate by about 2.57 Mbps.

Spectophotometric method for determination of certain cephalosporins using 4-chloro-7-nitrobenzo-2-oxa-1,3-diazole (NBD-Cl)

A simple, accurate and precise spectrophotometric method has been proposed for the determination of eleven cephalosporins, namely;cefaclor monohydrate, cefadroxil monohydrate, cefalexin anhydrous, cefradine anhydrous, cefotaxime sodium, cefoperazone sodium, ceftriaxone sodium, ceftazidime penthydrate, cefazolin sodium, cefixime and cefpodoxime pro- xetil in bulk drug and in pharmaceutical formulations. The method depends on hydrolysis of the studied drugs using 0.5M NaOH at 100°C and subsequent reaction of the formed sulfide ions with NBD-Cl (4-chloro-7-nitrobenzo-2-oxa-1, 3-diazole) to form a yellow-colored chromogen measured at 390 nm. Different variables affecting the reaction (e.g. NaOH concentration, hydrolysis time, NBD-Cl concentration and diluting solvent) were studied and optimized. Under the optimum conditions, linear relationships with good correlation coefficients (0.9990- 0.9999) were found in the range of 5-160 μg mL-1 for all studied drugs. The limits of assay detection and quantitiation ranged from 0.289 to 5.867 and from 0.878 to 17.778 μg mL-1;respectively. The accuracy and precision of the proposed method were satisfactory. The method was successfully applied for analysis of the studied drugs in their pharmaceutical formulations and the recovery percentages ranged from 96.6 to 103.5%.

Features and Outcomes of Spontaneous Bacterial Peritonitis in egypt, Single Center experience

Spontaneous bacterial peritonitis (SBP) is a serious complication of liver cirrhosis, with a recurrence rate up to 70% at 1 yr. Diagnosis of SBP is based on a positive ascitic fluid (AF) culture and/or a polymorph nuclear leukocyte count of >250 cells/μL. Third generation cephalosporins had been viewed as the drug of choice. Aim of the work: to study features, and outcomes of spontaneous bacterial peritonitis in National Liver Institute (Menofia University), a single center study. Methods: This work was conducted on 60 patients had SBP. All patient had AF analysis, bedside AF culture, Blood culture (BC), Serum albumin, Ascitic fluid gradient (SAAG). Results: The more severe the liver disease, as assessed by Child-Turcotte-Pugh (CTP score), the higher the possibility to have positivity of AF culture. Patient who had positive BC had significant rising of Creatinine. The pattern of infection in AF culture is close to that of BC, but Streptococci take the upper hand in BC and Staphylococcus in AF culture. Gram-positive organisms are more common in both AF and BC than gram negative. Mortality rate was higher in culture positive SBP patients 46.2%, and BC positive patients, 62.5%. In all positive cultures patients, Staph Species represents 44.8% with mortality rate of 42% of all mortalities. Most of them were resistant to cephalosporin 62.5%. Strep species represents 31% with mortality rate of 20%. e. coli represents 24.2% with mortality rate of 20%. Conclusion: The microbial pattern of organisms responsible for SBP has changed with predominance of gram-positive organisms. Pattern of microbial infection is similar in both blood and ascetic fluid culture, so BC results can be relied upon to guide therapy, in negative AF culture.
