CDK5 in RNA Biology:New Clues that may Underlie Neuronal and Glial Diseases

Cyclin-dependent kinase 5(CDK5) plays key roles in governing normal neuronal development and synaptic function though phosphorylation of serine and threonine residues on diverse protein targets. Emerging evidence also revealed that tightly controlled CDK5 activity is also crucial for the development of oligodendrocytes(OLs) and myelinogenesis,which provide protection and trophic support to brain neurons. Aberrant activation of CDK5 contributes to the pathogenesis of an increasing list of brain disorders,including Alzheimer's disease,stroke,schizophrenia,and epilepsy. However,molecular mechanisms that control CDK5 activity and its downstream effectors in neurons and OLs still remain vastly elusive. Activation of CDK5 depends on the available amounts of p35 or p39,two distinct non-cyclin protein activators. Our recent studies identified distinct mechanisms that regulate CDK5 activators in neurons and OLs during brain development. Here we report posttranscriptional mechanisms that control CDK5 activity in postnatal brain neurons and OLs by a functional interplay of RNA-binding proteins and microRNAs implicated in Alzheimer's disease and intellectual disability. Moreover,we discovered that CDK5-dependnent phosphorylation of RNA-binding proteins controls mRNA homeostasis and non-coding RNA biogenesis,including microRNAs and long non-coding RNAs in human neurons and OLs. These results provide the first evidence connecting CDK5 function with regulation of coding and non-coding transcriptome,which revealed new clues for CDK5 pathway function in normal and diseased brains.

采用1-乙基-(3-二甲基氨基丙基)碳二亚胺(EDC)法将抗赭曲霉毒素A(OTA)腹水与量子点荧光微球偶联制备荧光探针,以牛血清白蛋白-OTA偶联物及驴抗鼠二抗喷涂硝酸纤维膜形成试纸条检测线(T线)和质控线(C线),建立了基于T/C荧光强度(FIT/FIC)比值法免疫层析试纸条高灵敏度、快速定量检测玉米中OTA的新方法。量子点荧光微球试纸条定量检测OTA线性范围为0.051.0ng/mL(Y=0.259lnX+0.897,R2=0.995),半数抑制浓度(IC50)为(0.215±0.023)ng/mL(n=5),玉米提取液中OTA的检出限为0.05 ng/mL,单个样品检测时间10min。加标回收实验表明,3个OTA加标浓度的平均回收率为107.2%-125.5%,相对标准偏差均小于10%。40个实际玉米样品检测结果表明,量子点荧光微球试纸条与酶联免疫吸附分析方法检测OTA的结果具有良好的相关性(R2=0.975)。

Introduction: Human leukocyte antigen G (HLA-G) is a non-classical major histocompatibility complex (MHC) class Ib antigen characterized by a limited polymorphism. The expression of HLA-G at immune privileged sites and its ability to inhibit the effectors functions of immune cells has set HLA-G as a molecule of immune tolerance. This expression pattern is unique among HLA genes and suggests that HLA-G may be involved in interactions that are critical in establishing and/or maintaining pregnancy. Methods: Soluble HLA-G (sHLA-G) levels were measured using a BioVendor sHLA-G ELISA kit following the manufacturer’s protocol. The study participants include women undergoing spontaneous abortion, non-pregnant women, males and an archive sample of women who had normal vaginal deliveries without any complications and any history of malaria infection from gestation to delivery. Results: Soluble HLA-G levels were higher among women undergoing spontaneous abortion as compared to women who had normal vaginal delivery and non-pregnant women. Soluble HLA-G levels were also higher in second trimester as compared to first trimester in both women who had spontaneous abortions and women who had normal delivery. Conclusion: Although sHLA-G levels were higher among women undergoing spontaneous abortion as compared to non-pregnant women and women who had normal delivery, this may be playing a role in the maintenance of maternal immune tolerance to fetal antigen, since plasma sHLA-G levels increased with increasing trimester in both women who had normal delivery and women undergoing spontaneous abortion.

Multiplexing multiple orbital angular momentum(OAM)channels enables high-capacity optical ***,optical scattering from ambient microparticles in the atmosphere or mode coupling in optical fibers significantly decreases the orthogonality between OAM channels for demultiplexing and eventually increases crosstalk in ***,we propose a novel scattering-matrix-assisted retrieval technique(SMART)to demultiplex OAM channels from highly scattered optical fields and achieve an experimental crosstalk of -13.8 dB in the parallel sorting of 24 OAM channels after passing through a scattering *** SMART is implemented in a self-built data transmission system that employs a digital micromirror device to encode OAM channels and realize reference-free calibration simultaneously,thereby enabling a high tolerance to *** successfully demonstrate highfidelity transmission of both gray and color images under scattering conditions at an error rate of <0.08%.This technique might open the door to high-performance optical communication in turbulent environments.

Dear Editor,In the United States,pancreatic cancer is considered the fourth most common reason for cancer-related deaths;over 53%of patients are diagnosed at end-stage with a 5-year survival rate of less than 3%[1].Incomplete removal of all cancerous cells generally leads to local recurrence of pancreatic cancer[2].Thus,the detection of tumor margin status plays an essential role in the complete resection of pancreatic *** of the existing imaging modalities,like computed tomography and magnetic resonance imaging,are primarily used for preoperative planning;these modalities also have issues of bulky equipment,radiation concerns,and slow imaging speed[2].Thus,these modalities can neither provide an intraoperative diagnosis of cancer with high sensitivity and specificity nor be used to define tumor margins[3].

To assist with the commissioning [1] of the Five-hundredmeter Aperture Spherical radio Telescope (FAST), we performed a pulsar search, with the primary goal of developing and testing the pulsar data acquisition and processing pipelines. We tested and used three pipelines, two (Pl and P2 hereafter) searched for the periodic signature of pulsars whereas the other one was used to search for bright single pulses (P3 hereafter).

Antimicrobial peptides(AMPs)are potentially powerful alternatives to conven-tional antibiotics in combating multidrug resistance,given their broad spectrum of *** mainly interact with cell membranes through surface electrostatic potentials and the formation of secondary structures,resulting in permeability and destruction of target microorganism *** earlier work showed that two leading AMPs,MSI-78(4–20)and pardaxin(1–22),had potent antimicrobial activ-ity against a range of *** is known that the attachment of moderate-length lipid carbon chains to cationic peptides can further improve the functionality of these peptides through enhanced interactions with the membrane lipid bilayer,inducing membrane curvature,destabilization,and potential ***,in this work,we aimed to investigate the antimicrobial activity,oligomerization propensity,and lipid-membrane binding interactions of a range of N-terminal lipidated analogs of MSI-78(4–20)and pardaxin(1–22).Molecular modeling results suggest that aggregation of the N-lipidated AMPs may impart greater structural stability to the peptides in solu-tion and a greater depth of lipid bilayer insertion for the N-lipidated AMPs over the parental *** experimental and computationalfindings provide insights into how N-terminal lipidation of AMPs may alter their conformations,with subsequent effects on their functional properties in regard to their self-aggregation behavior,membrane interactions,and antimicrobial activity.
